Abstract

1410926 1-Phenyl biguanides WILLIAM H RORER Inc 10 Oct 1972 [14 Oct 1971 (2)] 46579/72 Addition to 1410925 Heading C2C The invention comprises l-phenyl biguanide having the general formula where n is an integer from 1 to 3; Alk, Hal and R may be at any position in the ring; Alk is an alkyl or alkenyl group having 1 to 7 carbon atoms; Hal is halogen; R is hydrogen, halogen or alkyl having 1 to 7 carbon atoms; provided that Alk is not 2-methyl when Hal is chloro or 4- bromo and R is hydrogen or when Hal and R are 3,5-dichloro; and Alk is not 4-methyl or 4-butyl when Hal is 3-chloro and R is hydrogen; and their non-toxic acid addition salts. The invention further comprises a process for preparing compounds of the formula wherein n, Hal and R are as defined above but are not subject to the above provisos and X is Hal, or C 1 to C 7 alkyl or alkenyl by reacting an acid addition salt of an aniline of the formula with cyanoguanidine in the presence of a mildly acidic, non-nucleophilic solvent at from room temperature to 150‹ C. The compounds of Formula I exhibit gastric antisecretory and spasmolytic properties and antihypertensive and CNS depressant activity. They may be mixed with conventional pharmaceutical carriers and administered orally or parenterally.
