Abstract
1393256 Triazolobenzodiazepines CIBAGEIGYAG 14 Jan 1972 [21 Jan 1971] 1844/72 Heading C2C Compounds of the general formula (R 2,3 = H, C 1-6 alkyl, C 7-9 aralkyl or NR 2 R 3 = C 4-6 polymethyleneimino, morpholino; A= C 1-3 alkylene; R 4 , 5 = H, F, Cl, Br, CF 3 , NO 2 , C 1-6 alkyl or alkoxy), their acid addition salts and 5- oxides are prepared by (a) reacting R 2 R 3 NH or an alkali metal derivative thereof with a reactive ester of the corresponding 1-hydroxyalkyl compound, (b) condensing a compound of the formula (X = SH, SMe, SEt, OMe, OEt, alkylamino, the alkylthio and alkoxy groups being optionally activated by a substituent) with R 2 R 3 NACONHNH 2 , (c) hydrolysing a compound of the formula (R 3 <SP>a</SP> = acyl) or its 5-oxide, (d) reacting a compound of the formula (ring D optionally substituted) with hydrazine and/or a strong acid, (e) reducing the corresponding 1-azidoalkyl compound or (f) alkylating the corresponding compound in which R 3 = H, optionally followed in each case by salt formation. The preparation of the 1 -hydroxyalkyl compounds (process (a)) and their p-methoxybenzyl ethers, and starting materials for processes (c) and (d) is described. The above compounds are central depressants, and may be administered in the form of pharmaceutical preparations containing them in association with a carrier.
