Abstract

1529972 Basic alcohols ALLEN & HANBURYS Ltd 15 July 1976 [29 July 1975] 31678/75 Heading C2C Compounds of the general formula (R 1 (one or more present) = halogen, CF 3 , NR 5 R 6 ; R 5,6 = H, C 1-6 alkyl, acyl; X = C 2-6 alkylene; R 2-4 = H, C 1-4 alkyl) and their salts are prepared by standard methods, e.g. (a) reducing a corresponding compound in which the hydroxymethyl group is replaced by a C 2-5 alkoxycarbonyl group, (b) reductively alkylating a compound of the formula or the corresponding α-aminoketone or primary amine (the phenolic OH group being optionally protected in each case) with a compound of the formula or (c) reacting a compound of the formula with a 3-alkoxycarbonyl-4-aralkoxyphenylglyoxal hydrate and reducing the product in one or more steps, optionally followed by modification of the R 1 substituent (e.g. reduction of acylamino to alkylamino) and/or salt formation. The preparation of starting materials for each of the above processes and 5-(p-fluorophenyl)- pentan - 2 - ol, p - trifluoromethyl - α - chlorotoluene, methyl 5 - dichloroacetyl - 2 - benzyloxybenzoate and 4-(p-fluorophenyl)-2-methylbutan -2-ol is described. The above compounds are bronchodilators and gastric acid inhibitors, and may be administered in the form of pharmaceutical preparations containing them in association with a carrier.
