Abstract

1390014 Process for the preparation of carbocyclic fused pyrimidine derivatives KONINKLIJKE PHARMACEUTISCHE FABRIEKEN NV 5 May 1972 [7 May 1971] 13802/71 Heading C2C A novel process for the preparation of compounds of the Formula I wherein A represents an alkylene or alkenylen e group which together with the carbon atoms of the pyrimidine nucleus to which it is attached forms a homocyclic ring containing 5, 6- or 7-carbon atoms, which homo-cyclic ring may carry one or more substituents selected from halogen, alkyl and alkoxy, the total number of carbon atoms in the alkyl and/or alkoxy substituents being not more than eight, and R is dialkylamino, or a nitrogen-, oxygen- or sulphur containing saturated or unsaturated mononuclear heterocyclic group linked to the carbocyclic fused pyrimidine ring through a carbon atom or when the group contains a nitrogen atom, either through the nitrogen atom or carbon atom, said heterocyclic group optionally carrying one or more substituents selected from halogen, alkyl and alkoxy and/or when it contains an acylatable nitrogen atom, and N-acyl substituent, wherein alkyl, either alone or forming a part of a radical, have up to eight carbon atoms and acid addition salts thereof, which process comprises reacting a carbocyclic oaminonitrile of the Formula II with a nitrile RCN in the presence of a basic alkali metal-containing compound and decomposing the intermediate product of the Formula IV where M is an alkali metal atom, by addition of water and optionally forming an acid addition salt thereof. Pharmaceutical compositions of 4-amino-2-(2- furyl)quinazoline, a non-toxic acid addition or quaternary ammonium salt thereof, when administered orally or parenterally with the usual excipients or incorporated into an animal feedstuff, show activity against pleuropneumonia-like organisms and other gram negative bacteria and also exert antihypertensive activity. Certain novel compounds within the scope of the Formula I and pharmaceutical compositions thereof form the subject of Divided Specification 1,390,015.
