Abstract

1378134 Antigen purification F HOFFMANN-LA ROCHE & CO AG 11 May 1973 [12 May 1972] 22611/73 Heading A5B Human carcino-embryonic antigen is purified by (a) absorbing it on a mixed bed cellulose ion-exchange column comprising (i) microgranular diethylaminoethyl cellulose in the free base form having rod-shaped particles, a capacity of 1.0+0.1 meg/gm, a particle size distribution of 20Á to 60Á and a water regain of 2.3 to 2.8 gm/gm dry exchanger and (ii) a microgranular carboxymethyl cellulose with a capacity of 1.0 Œ 0.1 meg/gm, having rod-shaped particles, a particle size distribution of 20Á to 60Á and a water regain of 2.3 to 2.7 gm/gm dry exchanger, (b) eluting the column with an aqueous solution of ammonium acetate and sodium chloride, (c) subjecting the eluate to sequential chromatography first on an agarose gel containing about 6% agarose and having a particle size of 40 to 210Á and then on a hydrophilic water-insoluble cross-linked dextran polymer gel and (d) collecting and concentrating the eluate fractions which have a spectrophotometric absorption peak at 280 mÁ and a molecular weight of 200,000 to 500,000.
