Claims (7)

Having now particularly described and ascertained the nature of our said invention and in what manner the ..same is to be performed, we declare that what we claim is:
1. A process for the purification of human carcinoembryonic antigen which comprises subjecting carcinoembryonic antigen material to chromatography on a mixed bed cellulose ion exchange column comprising microgranular diethylamino-ethyl cellulose in the free base form having rod shaped particles, a capacity of 1.0 + 0.1 meq/gm, a particle size distribution of about 20 μ to about 60 μ and a water regain of 2.3 to 2.8 gm/gm dry exchanger, and a microgranular carboxymethyl cellulose with a capacity of 1.0 + 0.1 meq/gm, having rod shaped particles, a particle size distribution of about 20 μ to about 60 μ and a water regain of 2.3 to 2.7 gm/gm dry exchanger, and collecting the eluates resulting from eluting with a solution of ammonium acetate in sodium chloride followed by subjecting the resulting material to sequential chromatography on two different gel columns, the first of which is an agarose gel having about 6 o by weight agarose and a particle size of from 40-210 microns and the second of which is a hydrophilic water-insoluble cross-linked dextran polymer gel and collecting and concentrating the eluates which have a spectrophotometric absorption peak wave length of 280 ιημ and a molecular weight of about 200,000 to 500,000.
2. A process as claimed in claim 1 wherein the mixed bed column contains equal amounts by weight of the diethyl-aminoethyl cellulose and carboxymethyl cellulose.
3. A process as claimed in claim 1 or claim 2 wherein the solution of ammonium acetate in sodium chloride has a pH of 4.
4. A process as claimed in any one of claims 1 to 3 for the production of human carcinoembryonic antigen component A wherein the sodium chloride has a molarity of 0.05 M.
5. A process as claimed in any one of claims 1 to 3 for the production of human carcinoembryonic antigen i component B wherein the sodium chloride has a molarity 0.1 .

7. Human carcinoembryonic antigen, whenever purified by a process as claimed in any one of the preceding claims.
