Abstract

1356911 Purification and functionation of carcinoembryonic antigen F HOFFMANN-LA ROCHE & CO AG 1 June 1971 [1 June 1970 27 Jan 1971 12 April 1971] 18309/71 Heading A5B A purified human carcinoembryonic antigen is prepared by a process comprising: a) homogenizing adenocarcinoma tissue; b) removing solid from the homogenisate; c) treating the supernatant with a glycoprotein solvent (e.g. perchloric acid); d) discarding the precipitate and clarifying the solution; c) subjecting the clarified solution to osmosis against an aqueous solution of a polyethylene glycol which has a M.W. of approx. 15,000 to 20,000 and a softening point of about 60‹C; f) dissolving the resulting rentate in an aqueous buffer of pH 5-9; g) chromatographing the solution from f) sequentially on two different gel columns, the one being an agarose gel comprising approximately 6% agarose and having a particle size of 40-210 microns and the other being a water-insoluble, cross-linked, hydrophilic, dextran polymer gel; h) collecting the fractions having a spectrophotometric adsorption peak of 280mÁ and a M.W. of about 200,000 to about 500,000. These fractions may be combined and subjected to further treatment to produce two different active compounds, namely "Human carcinoembryonic antigen A and B". Suitable treatment comprises osmosis against a polyethylene glycol solution as described above, dissolution in a buffer, e.g. ammonium acetate at pH4, clarification and chromatography on a mixed bed ion-exchange column comprising microgranular diethylaminoethyl cellulose and carboxymethyl cellulose, using e.g. ammonium acetate/sodium chloride (pH 4) as eluant.
