Abstract

1353534 Pyrimido-[5,4-e]-1 ,2,4-triazines ROCHE PRODUCTS Ltd 12 July 1972 [13 July 1971 6 Aug 1971 21 March 1972] 32801/71 37049/71 and 13116/72 Heading C2C The invention comprises compounds of formula wherein R is CF 3 , C 1-6 alkyl, Ar or Ar-C 1-6 - alkyl, R<SP>1</SP> is H, C 1-6 alkyl, Ar or Ar-C 1-6 -alkyl, R<SP>2</SP> is H and R<SP>3</SP> is H, C 1-6 alkyl, C 2-6 alkenyl or alkynyl, C 3-6 cycloalkyl, Ar, Ar-C 1-6 -alkyl, HO, HO-C 1-6 alky or acyl, or R<SP>2</SP>, R<SP>3</SP> are each C 1-6 alkyl which may be joined directly, or through N, O or S to form (with the N shown) a 5- or 6-membered heteromonocyclic ring; and Ar is Ph, possibly substituted by halogen, C 1-6 alkyl and/or C 1-6 alkoxy. These compounds are prepared by reacting (1)a 5-amino-4-chloro-6- hydrazino-2-R-pyrimidine with R<SP>1</SP>C(OR<SP>4</SP>) 3 where R<SP>4</SP> is C 1-6 alkyl, and reacting the thus-formed 5- chloro - 1,2 - dihydro - 7 - R - 3 - R<SP>1</SP> - pyrimido- [5,4-e]-1,2,4-triazine with the appropriate amine under oxidizing conditions or with NH 3 and/or alkali metal azide; (2) a compound of Formula I (R<SP>2</SP> = R<SP>3</SP> = H) with a primary amine or with an acylating agent; or (3) a compound of Formula I (R<SP>2</SP> = R<SP>3</SP> = Me) with NH 3 , NH 2 OH or a primary amine. Therapeutic compositions having anti-inflammatory, and in some cases diuretic activity comprise compounds of the above formula, which may be administered enterally, parenterally or rectally.
