Abstract

1348258 Dioxinoquinoline derivatives LABORATOIRE ROGER BELLON 5 Aug 1971 [5 Aug 1970] 36846/71 Heading C2C Novel compounds I and II in which R is 1-4 C alkyl; R 1 is H, CH 3 or Cl; in compounds I R 2 is CH 3 and in compounds II R 2 is H or CH 3 with the proviso that in compounds II R 1 and R 2 are not both H; and their salts are prepared by hydrolysis and saponification of compounds IX and X in which X is such that X- is a halide ion or the anion of an esterifiable acid and R<SP>1</SP> and R<SP>11</SP> are 1-4 C alkyl or NR<SP>1</SP>R<SP>11</SP> is a 5-6 membered heterocyclic ring. Compounds IX and X are prepared by condensation of a compound III in which R 2 is attached to the 5 or 7 position (prepared by reduction of the corresponding nitro compound) with ethyl ethoxymethylenemalonate to give a compound IV cyclization of compound IV to give a compound V or VI depending on the position of R 2 ; condensation of V or VI with HNR<SP>1</SP>R<SP>11</SP> to give a compound VII or VIII respectively and quaternization. 5 - Methyl - 6 - nitro - 8 - chloro - 1,3 - benzodioxane is prepared by condensing formaldehyde with 3-hydroxy-4-chloro-6-nitro-toluene. Pharmaceutical compositions comprise a compound I or II together with a suitable carrier or diluent and are used as antibacterial agents.
