1336155 Preparation of 7-aminocephalosporanic acid derivatives FUJISAWA PHARMACEUTICAL CO Ltd 11 Nov 1970 [17 Nov 1969] 58753/70 Heading C2A A process for the preparation of 7-aminocephalosporanic acid derivatives of Formula I: where R 1 is azido, thiadiazolylthio, oxadiazolylthio or tetrazolylthio, X is thio, oxy or a single bond, R 2 is halogen or a sydnone, tetrazolyl, oxadiazolyl, thienyl, phenyl or thiadiazolyl, with the proviso that, if R 2 is halogen, X is a single bond, or a pharmaceutically acceptable salt thereof, which process comprises reacting a compound of general Formula II: in the presence of a base, with chlorotrimethylsilane, or reacting a salt of (II) with chlorotrimethylsilene, and further reacting the resultant compound III: with a halogenating agent, still further reacting the resulting compound IV: with an alkanol and still further reacting the resulting compound V: with a compound of Formula VI: in the presence of a condensing agent. Alternatively a compound of Formula V may be reacted with a compound of Formula VI having a reactive group in place of the carboxyl group, in the presence of an organic base, and then hydrolysing the resulting compound VII: Certain of the stages are those used in the imino halide or imide halide reaction. The invention also comprises compounds of the Formula I.
