Abstract

1492435 Cephalosporin derivatives E R SQUIBB & SONS Inc 19 May 1975 [30 Aug 1974] 21255/75 Heading C2C [Also in Division C3] Novel compounds I (A is tetrazolyl optionally substituted by 1-6 C alkyl, and linked through one of its N atoms; R is H, 1-6 C alkyl, phenyl-1-6 C alkyl, -CH(R 2 )OCOR 3 or a cation derived from an alkali metal, alkaline earth metal, tri-(1-6 C alkyl) amine or 5-6 C cycloalkylamine; R 2 is H or 1-6 C alkyl; R 3 is 1-6 C alkyl; phenyl or phenyl-1-6 C alkyl; and X is tetrazolyl optionally substituted by 1-6 C alkyl) are prepared by acylation of the corresponding 7- amino-cephem compound with a reactive derivative of an acid ACH 2 SCH 2 COOH or by reaction of the corresponding 7-haloacetamidocephem compound with ACH 2 SH or by conversion of the corresponding 3-acetoxymethyl cephem compound into the 3-tetrazolylthiomethyl derivative using an appropriate third XSH. Intermediates isolated are methyl [(2-tetrazolyl)methylthio]acetate and the corresponding free acid and its potassium salt. Pharmaceutical compositions comprise a compound I together with a suitable diluent and/or carrier and are useful as antibacterial agents.
