[Also in Division C3] A method for determining endohydrolases capable of hydrolysing polysaccharides, in aqueous samples, comprises contacting the sample with a water-insoluble, hydrophilic, swellable, enzymatically hydrolysable, three dimensional network of molecules of a polysaccharide or enzymatically hydrolysable derivatives thereof, the molecules being crosslinked by bridges with bonds of a covalent character, in which network there is also present indicatable groups or atoms (as defined) bound by means of bonds of a covalent character, a reaction taking place between the enzyme and reagent to release water-soluble fragments of the reagent and subsequent to the enzyme having reacted upon the reagent for a determined length of time, separating undissolved reagent from the liquid with the water-soluble fragments of the reagent dissolved therein, and then determining the indicatable groups or atoms in the liquid or undissolved reagent as a measure of enzyme activity. Specified enzymes are alpha-amylase and endodextranase. The indicatable groups or atoms may be colour-producing groups, fluorescent groups or radioactive isotope atoms or groups containing radioactive isotopes. The reagent can be mixed with paper pulp to form a reagent-containing paper.
