1287783 Determination of thrombin activity BAXTER LABORATORIES Inc 16 Sept 1969 [2 Oct 1968] 45581/69 Heading B1X [Also in Division A5] A gel plate for the quantitative determination of thrombin activity in blood plasma and plasma fractions comprises a plate carrying an agarose gel medium containing unclotted fibrinogen. In preparing the plate, from 1 % to 10% gelling agent may be dissolved in heated buffer volution, for example one comprising 0.15M sodium chloride, 0.04M imidazole, 0.02M EDTA and the resulting solution mixed with 10mg% to 2mg% unclotted fibrinogen. When the gel is set, holes may be punched in it for the samples to be tested and the plate may be covered with a protective plastics membrance and packed in plastics film or metal foil optionally together with control samples, buffer materials and capillary tubes for making the assay. The thrombin activity of a known plasma sample is determined by introducing the sample and a series of control samples of known concentration into individual wells in a gel medium containing unclotted fibrinogen, incubating for a predetermined period of time to produce in the gel opaque radial diffusion zones of reaction product, and measuring the diameters of the diffusion zones. The gel may be agarose (preferred), gelatin, pectin, silica gel, starch. agar, algin, carageenin, cross-linked polyacrylamide.
