Abstract
1,239,821. Penicillins. E. R. SQUIBB & SONS Inc. 25 June, 1968 [20 July, 1967; 16 May, 1968], No. 30222/68. Heading C2A. 6 - Acylaminopenicillanic acids, salts or esters thereof, are prepared by acylating the imine reaction product of 6-aminopenicillanic acid, or a salt or ester thereof, with an aldehyde and hydrolysing the acylated imine reaction product. The imine reaction product has the formula wherein R is H or an organic radical which forms an aldehyde of the formula RCHO and R<SP>1</SP> is H a group capable of replacing hydrogen in a caroxy group to form an ester or a saltforming ion. Suitable acylating agents have the formula R<SP>11</SP>COX wherein R<SP>11</SP>CO is an acyl group and X is OH, halogen, O-acyl, O-alkyl, O-aryl, amino, cyano or azido. The acylated imine reaction product has the formula wherein R, R<SP>1</SP>, R<SP>11</SP> and X have the above meaning. Examples relate to the preparation of phenoxymethylpenicillin, sodium oxacillin, potassium oxacillin, methicillin, ampicillin, cloxacillin, sodium phenoxymethylpenicillin and dicloxacillin. The imine reaction products are described in Specification 974,125.
