Abstract
1,226,350. 1,2α - Methylene - 7α - halosteroids. SCHERING A.G. 25 March, 1968 [23 March, 1967], No. 14163/68. Heading C2U. Novel 1,2α - methylene 7α - halo - steroids of the partial structure (wherein R is OH, Oacyl, Oalkyl, F, Cl or Br; and X is Cl or Br) are prepared by reacting the corresponding 1,2α - methylene - #<SP>4'6</SP> - 3 - ketosteriods which do not contain further double bonds simultaneously with elemental chlorine or bromine or compounds capable of releasing positive chlorine or bromine and an appropriate nucleophilic reagent, such as water in the presence of a strong acid in an inert solvent (to introduce OH), dimethylformamide in the presence of an anhydrous acid (to introduce O.CO.H), a carboxylic acid in the presence of an anhydrous acid (to introduce Oacyl), an alcohol in the presence of an anhydrous acid (to introduce Oalkyl), a chloride (or bromide) in the presence of an anhydrous carboxylic acid and a strong anhydrous acid or HCl (or HBr) in an inert anhydrous solvent (to introduce Cl or Br), or HF in an inert anhydrous solvent or a fluoride in the presence of an anhydrous carboxylic acid and a strong anhydrous acid (to introduce F). Preferred starting materials are 1,2α - methylene - #<SP>4</SP>'<SP>6</SP> - pregnadien - 17α - ol- 3,20 - dione and its esters and their 16α - methyl derivatives, 1,2α - methylene - #<SP>4</SP>'<SP>6</SP> - androstadien - 17# - ol - 3 - one and 16α,17α - isopropylidenedioxy - -1,2 α -methylene - #<SP>4</SP>'<SP>6</SP> - pregnadiene- 3,20-dione. The steroids of the invention are stated to be potent gestagens with only a slight anti-androgenic side effect, and they may be made up into pharmaceutical compositions with suitable carriers.
