Abstract
1,196,648. Moenomycin components. FARBWERKE HOECHST A.G. 2 Oct., 1967 [1 Oct., 1966], No. 44732/67. Heading C2A. Moenomycin components D, E and F are isolated from moenomycin B 1 and moenomycin components G and H are isolated from moenomycin B 2 , by subjecting moenomycin B 1 or B 2 to either (a) partition chromatography on silica gel using a mixture of chloroform, one or more C 1 to alkanols and water (40 : 70 : 20) or alcohol/water/borate buffer of pH 9À0 (70 : 25 : 5) as developing agent, or (b) adsorption chromatography on a strongly basic polystyrene anion exchange resin using a solution of potassium chloride in aqueous methanol as elution medium. Selected bands in the case of (a) are extracted with methanol, the solvent removed, the residue dissolved in methanol, made alkaline with ammoniacal methanol and the ammonium salt of the component precipitated with ether. As regards (b), selected fractions are evaporated to remove the methanol and dialysed to remove the potassium chloride from the residual aqueous solution. The inner dialysate is freeze dried, dissolved in methanol and the potassium salt precipitated by the addition of ether. The components D, E, F, G and H are colourless, amorphous, weak acids, soluble in water and polar organic solvents, forming salts with alkali metals and alkaline earth metals and further characterized as follows:- The Rf values are determined in thin layer chromatography on silica gel using chloroform/ ethanol/water 40 : 70 : 20 (I) and isopropanol/water/borate buffer pH9 70 : 25 : 5 (II).
