Abstract

1,191,384. Benzothioxanthene derivatives. MERCK & CO. Inc. 24 April, 1968 [27 April, 1967], No. 19394/68. Heading C2C. Novel benzothioxanthene derivatives of the general formulµ wherein B is a benzene ring fused in the [a] or [c] position, X is a chlorine or bromine atom attached to the left-hand ring and each R is a hydrogen atom or a C 1-6 alkyl group, and non- toxic acid addition salts thereof are prepared (a) by reaction of a hydroxybenzothioxanthene derivative of the general formula with phosphorus and hydrogen iodide; (b) by reaction of a benzothioxanthene derivative of the second general formula above with phosphorus and hydrogen iodide; (c) by catalytic hydrogenation of a benzothioxanthene derivative of the second general formula above, in each case producing a benzothioxanthene derivative of the first general formula above; or (d) by reaction of a hydroxy-benzothioxanthene derivative of the third general formula above with an acidic dehydrating agent to produce a benzothioxanthene derivative of the second general formula above; followed optionally by salification of the product. Pharmaceutical compositions having local anaesthetic activity comprise a non-toxic acid addition salt of a benzothioxanthene derivative of the first or second general formula above in sterile aqueous solution and may be administered by injection.
