Abstract

1,176,821. N-sulphomethyl polymyxin B. CHAS PFIZER & CO., Inc. 17 Feb., 1967 [2 June, 1966], No. 7808/67. Heading C2A. A pH and potency stabilized N-sulphomethyl polymyxin B is prepared by hydrolyzing an aqueous solution of N-sulphomethyl polymyxin B sodium salt at an elevated temperature, e.g. 50-60‹ C., while adding sufficient physiologically acceptable acid to the reaction mixture to maintain the pH thereof between 6À5 and 7À4 until the reaction mixture attains a constant pH between 7À0 and 7À2 and then recovering the resulting product. The preferred acid is sulphuric, but hydrochloric, succinic, citric, ascorbic, maleic and gluconic may be used. On completion, the reaction mixture may be filtered with filter aid, sterile filtered and lyophilized. A therapeutic composition for the treatment of coliform infections in animals comprises the above N-sulphomethyl polymyxin B in an injectable vehicle.
