Abstract

The invention comprises compounds of formula <FORM:1114584/C2/1> having an ethylenic double bond at the 4,5- or 5,6-position, wherein R is a carboxylic acyl group, R1 is a C2- 4 n-alkyl group and R2 is hydrogen or a C1- 6 alkanoyl group, and their preparation by acylation. Exemplified groups R include hemisuccinoyl and adamantane-1-carboxylic acid acyl groups. 3-Hydroxy-starting materials are prepared by (a) reduction of 3-ketones, optionally after 17-acylation with simultaneous 3-enol acylation, followed by partial hydrolysis, (b) reduction of a 3-acyloxy-3,5-diene, or (c) hydrolysis of a 17b -acyloxy-5-ene to a 17b -hydroxy-5-ene with lithium aluminium hydride. The compounds of the invention have progestational, pituitary gonadotrophin inhibiting and anti-estrogenic activity and may be used in pharmaceutical compositions in combination with a carrier, for example in forms suitable for oral, parental or rectal administration.
