Abstract

Crude preparations of the callicrein inactivator from the lungs of cattle are purified by stirring an aqueous solution containing the inactivator (and having a degree of activity of 80-140 ATU/mg. of dry material) with 3-10% by weight of MgO at 10-50 DEG C. for 1-16 hours, the MgO charged with contaminating proteins and colouring matter being then separated and the solution containing the inactivator so obtained being mixed with 2-6 times its volume of a C1-C3 aliphatic alcohol or acetone to precipitate the inactivator. The inactivator solution obtained after removal of MgO may be treated with a cation exchange resin to remove any Mg ions present. The aliphatic alcohol or acetone used for precipitation may first be cooled to -5 DEG C. The precipitate finally obtained may be worked up to a dry powder by washing with ethanol or acetone and then drying. Alternatively, the precipitate may be dissolved in pyrogen-free water or in physiological NaCl solution and the solution obtained may then be sterilised by filtration.
