Abstract
<PICT:1057194/C2/1> The invention relates to an anti-tumour antibiotic tuoromycin which is effective against gram positive microorganisms, is weakly acidic forming violet crystals of melting point 216-220 DEG C.; is insoluble in water, dilute mineral acid, diethyl ether and hexane; sparingly soluble in methanol, ethanol, n-butanol, carbon tetrachloride and dilute aqueous Na2CO3; moderately soluble in acetone, ethyl acetate, propylene glycol, polyethylene glycol and ethylene glycol mono-methyl ether; readily soluble in benzene, dioxane, methylene chloride, chloroform, dimethylsulphoxide and dilute aqueous alkali with which it forms salts; gives in elementary analysis C=57.17%, H=3.63%, O=39.20%; shows U.V. maximum <FORM:1057194/C2/1> <FORM:1057194/C2/2> <FORM:1057194/C2/3> shows Rf value 0.7 in paper chromatogram system benzene and chloroform (2:1), paper impregnated with 20% formamide in acetone; Rf 0.85 silica gel chromatogram in system chloroform and methanol (8:2) and Rf value 0.75 in system ethyl acetate and methyl ethyl ketone and formic acid and water (50:30:10:10). Tuoromycin is produced by cultivating Streptomyces tuirus N.R.R.L. 3120 under aerobic conditions in an aqueous nutrient medium containing a source of C, N and inorganic salts. A typical medium contains soybean flour-(C and N source), CaCO3 and NaCl in water. The antibiotic may be isolated from the separated mycelium by extraction with acetone followed by removal of the solvent or precipitation with a non-solvent, e.g. petroleum ether. Purification may be by recrystallization from solvents, chromatography or countercurrent distribution. Tuoromycin or a salt thereof may be formulated into compositions for human or animal use. Typical formulations are tablets, dragees, powders, suppositories, suspensions and injections. It may be used as a disinfectant, preserving agent or additive for animal feedstuffs.
