Abstract

The invention relates to an antibiotic ussamycin which is active against gram positive micro-organisms and tumours; is basic forming salts with inorganic and organic acids; soluble in methanol; insoluble in acetone, ether, benzene, chloroform and petroleum ether; hydrochloride is soluble in water; light-yellow colour, stable at pH 3 to 8; on acid hydrolysis produces glycine, alanine, leucine, serine, aspartic acid, glutamic acid, arginine and lysine; Rf value 0.74 in n-butanol, pyridine, acetic acid, water (15 : 10 : 3 : 12); no U.V. spectrum. Ussamycin is produced by cultivating streptomyces lavendulae NRRL 3119 (mutant) in an aqueous nutrient medium. It is isolated from the clarified broth by (A) adsorption on active carbon, activated earth or a weakly acidic ion exchanger. Elution is by means of aqueous acid or aqueous water-miscible organic solvents, e.g. aqueous methanol; (B) precipitation as picrate of Reineckate. Purification is by adsorption or distribution chromatography or by counter-current distribution between two immiscible solvent phases, e.g. in sec-butanol-3% acetic acid. The crude antibiotic may be dissolved in water and precipitated with Reinecke salt. Specified salts are the hydrochloride, sulphate, phosphate, nitrate, acetate, propionate, valerate, palmitate, oleate, succinate, citrate or mandelate. Pharmaceutical preparations for human or veterinary use comprise ussamycin or a physiologically tolerable salt thereof and a carrier. Formulations may be tablets, dragees, powders, solutions, suspensions, emulsions, suppositories and injections. The antibiotic may be added to an animal feed and may be used as a disinfectant or preservative.
