Abstract

1,053,339. New compounds of the lignan series. SANDOZ PATENTS Ltd. March 17, 1964 [March 29, 1963; Jan. 7, 1964; Feb. 3, 1964; Feb. 10, 1964 (3)1, No. 11121/64. Heading C2C. The invention comprises (1) novel compounds of Formula I in which each of R 1 and R 2 represents a hydrogen atom, or one of them is a hydroxyl radical or glucosido radical and the other a hydrogen atom, and R 3 is a hydrogen atom or a C 1 -C 4 alkyl group, and R 4 is a C 1 -C 4 alkyl radical, and (2) the preparation of compounds (a) of Formula I in which R 1 is a hydrogen atom, or an hydroxyl or glucosido radical, and R 2 is hydrogen, by reacting a compound of Formula II in which R 5 has the same meaning as R 1 with an alcohol R 4 OH in the presence of a Lewis acid; (b) of Formula I in which R 1 is a hydrogen atom and R 2 is a hydroxyl or glucosido radical, by reacting a compound of Formula IV in which R 6 has the same meaning as R 2 with an alcohol R 4 OH in the presence of a Lewis acid; (c) compounds of Formula I in which R 1 is an hydroxy group and R 2 is hydrogen, by subjecting the corresponding #-glucoside to enzymatic or acid hydrolysis, or by subjecting a compound of Formula II in which R 5 is a glucosido radical to enzymatic or mild acid hydrolysis, and reacting the product with an alcohol R 4 OH in the presence of an inorganic or strong organic acid or a Lewis acid, any mild acid hydrolysis being effected with an aqueous organic carboxylic acid at a temperature not exceeding 80‹ C.; (d) compounds of Formula I in which R 1 is a hydrogen atom and R 2 is a hydroxyl group, by subjecting the corresponding glucoside to enzymatic hydrolysis, or by subjecting a compound of Formula IV in which R 6 is a glucosido radical to enzymatic hydrolysis and reacting the product with an alcohol R 4 OH in the presence of a Lewis acid; and (e) compounds of Formula I in which R, is a hydrogen atom or hydroxyl radical and R 2 is hydrogen by reacting a compound of Formula II in which R 5 represents a hydrogen atom or a hydroxyl or glucosido radical with an alcohol R 4 OH in the presence of a strong organic or inorganic acid. Therapeutic compositions having anti-mitotic action which may be administered enterally or parenterally contain as active ingredient compounds of Formula I in which each of R 1 and R 2 represents a hydrogen atom, or one of them is a hydrogen atom, and the other is a hydroxyl radical, and R 3 and R 4 are as defined above.
