The skin is a vital organ in its own right that consists of three distinct tissues, each assuming different roles through different cell types and structures.
The most surface tissue and therefore the most exposed is the epidermis. This multilayered (Malpighian) and keratinizing epithelium, the outermost part of which is the stratum corneum, consists of different cells associated with many protective barrier functions. The majority are keratinocytes which, by their proliferation / differentiation processes, result in the formation of the horny layer known for its appearance and its hydrophobic properties, compact and tight. The major role of the epidermis is to provide the skin, and therefore the human body, with a first line of protection against external aggressions, such as physical, chemical, waterborne and bacteriological attacks, particularly via differentiation and the stratum corneum. ..
In the intermediate position, the dermis is also a connective tissue mainly invested with fibroblasts and matrix proteins giving the skin its qualities of compressibility and elasticity known. Within the conjunctive framework, other cells and structures are inserted, such as an important circulatory and nutritive network, consisting of blood vessels and lymphatic capillaries, as well as the epidermal appendages (hair, nails, pilosebaceous glands and sweat glands). ) that originate in the deep dermis. The hypodermis, located in depth and consisting for the most part of fatty lobules (adipocytes), provides a primary support function, mechanical and thermal protection and also plays a role in storing energy reserves quickly mobilized for all biological needs, such as for example cell renewal, defense of the body or muscle contraction.
Keratinocyte differentiation is an oriented maturation of epidermal keratinocytes into corneocytes (keratinocytes of the stratum corneum), during which cells change size, morphology, biochemical composition, membrane nature and biological activity. Corneocytes are dead anucleated keratinocytes, which pile up on the surface of the skin, before desquamating. They thus form a compact and highly hydrophobic layer through which the penetration of exogenous agents or water losses are not normally possible. This process of differentiation is also accompanied by modifications of the components and cellular structures such as interkeratinocyte junctions, which are increasingly represented as a function of differentiation.
Intercellular junctions are characteristic areas of epithelial cell membranes that allow strong cell adhesion to one another. They represent areas of interaction of the plasma membrane with the cytoskeleton and, in fact, areas of intercellular communication. They can be subdivided into three categories: anchor junctions, communicating junctions and impervious junctions.
Anchorage junctions are structures that mechanically join the cells together with more or less complex intracytoplasmic arrangements. These are (i) the intermediate junctions / adherent junctions (zonula adherens) directly responsible for the maintenance of the cellular form and directly involved in cellular communication, since at the origin of numerous cellular signals; and (ii) desmosomes and hemidesmosomes, which provide a strong and durable adhesion by acting as a "snap" (predominant junctions in the skin and the strongest). Participating directly in the barrier function of the epidermis, interkeratinocyte junctions are involved in the regulation of skin hydration and in the protection thereof. Water and hydration of living organisms is essential. It is also for the good general and cellular functioning of all organs. In the skin, water is 80% represented in the dermis, and in the epidermis there is a decreasing gradient from the basal layer to the stratum corneum of 70 to 10%. In depth, the water (from the diet) is conveyed via blood capillaries irrigating the dermis (hence the importance of good angiogenesis). It diffuses into the integument and is normally retained in the epidermis through the differentiation and sealing of the most differentiated layers, thus regulating the insensitive water loss (PIE); hence the importance of good keratinocyte differentiation.
In this context, the barrier function of the epidermis is essential. It tends to deteriorate with age or in certain transient situations, causing defects of desquamation, scars, dry and inhomogeneous appearance, alteration of complexion and color. Reinforcing it is therefore a protective process and a process that promotes hydration. Strengthening angiogenesis maximizes natural skin irrigation and tissue hydration. These two mechanisms combined give rise to cosmetic benefits of protection, hydration, smoothing, softness, radiance, and complexion, appearance and color homogeneous.
Regarding the barrier function and keratinocyte differentiation, the most common cosmetic solution is physical, it is to strengthen the surface barrier by a lipidic contribution (hydrophobic to limit the PIE). But this is transient and inefficient because it does not directly influence the natural biological process of epidermal differentiation. Molecules of the family retinoids directly influence this process, but also causing an inflammatory reaction. Moreover, these molecules are not of natural origin.
Regarding natural hydration, the most common cosmetic solution is physical: the water supply via emulsions or gels allows an immediate effect but that transient and lacking in effectiveness because it does not directly influence the natural biological process hydration.
There is therefore a real need to provide compositions and methods of cosmetic treatments for reinforcing the barrier function of the skin and its hydration with active ingredients of natural origin that are permanently effective.

The root of licorice currently has many applications, mainly in pharmacy, cosmetics, as well as in the food industry.
Thus because of its intense sweetness, licorice root preparations are widely used as a taste corrector in drugs and in certain cosmetics, such as toothpastes and mouthwashes, as well as in food products. Indeed, its sweetening power, which is 50 times greater than that of sugar, is mainly due to the ammonium glycyrrhizate salts present in the root ([2]).
Review of pharmacological effects of Glycyrrhiza sp. And its bioactive compounds ([3])·In addition, licorice is a medicinal plant both in Europe and Asia. Registered in the European Pharmacopoeia, it is traditionally used either orally, in the treatment of respiratory diseases, including cough, and digestive disorders, or in local use, as analgesic in diseases of the oral cavity. These uses are mainly due to the presence of saponins (ex: glycyrrhizinic acid, glycyrrhizin) with proven anti-inflammatory properties (Phytotherapy research, 2008, 709-724, Review of the pharmacological effects of Glycyrrhiza sp, and its bioactive compounds ([3]).
The hunt for natural skin whitening agents ([4]); International journal of cosmetic science, 2003, 25, 109-181. In cosmetics, licorice root is used mainly for its whitening action: it is allowed thanks to the mechanisms of action of certain flavonoids (glabridin, licochalcone, liquiritin, etc.) on melanogenesis (International Journal of Molecule Science, 2009, 10, 5326-5349, The hunt for natural skin whitening agents ([4]), International journal of cosmetic science, 2003, 25, 109-181. Skin-lightening products revisited ([5]) · The other applications in cosmetics are for products intended for sensitive skin ([3]) The use of liquorice extract (glabra species), or molecules present in licorice, such as glycyrrhizic acid or glabridin, for example, are described, for example for skin whitening (IL-109012 ([6])), skin radiance (FR3019040 ([7]), skin protection (ITRM20130542 ([8])) and anti-inflammatory action (WO200698006 ([9])).

EXEMPLESEXAMPLES
4- Réajustement en solvant de façon à obtenir un mélange dipropylène glycol/eau en proportion 70/30 (v/v). 5- Filtration stérilisanteIn the following examples, the licorice extract (Glycyrrhiza glabra L.) was prepared as follows, and according to the PAT-Traire® Plants protocol described in WO01 / 33942 ([10]). 1- Licorice plants are obtained by cuttings of mother plants and are cultured in buckets. These are cultured aeroponic system with spraying of a commercial nutrient solution N, P, K (nitrogen, potassium phosphorus in respective proportions 20/20/20) every 15 minutes. 2- Recovery of the metabolites released by the roots by means of a mixture of dipropylene glycol / water in proportion 70/30 (v / v) put in contact by immersion with the roots with a ratio fresh plant / solvent of extraction of 0, 5 kg / L. 3- Coarse filtration to collect the extract. 4- Readjustment solvent to obtain a mixture of dipropylene glycol / water in proportion 70/30 (v / v). 5- Sterilizing filtration
Various studies have been carried out on normal human keratinocytes, obtained from abdominal plasties, seeded in a monolayer (ref .: NHEK341) under the appropriate culture conditions, namely in a KSFM (keratinocyte serum free medium, KSFM invitrogen) medium. a temperature of 37 ° C under an atmosphere at 5% CO2 and saturated with moisture. After 24 h, the medium was replaced with medium containing Glycyrrhizza glabra extract (0.1%) or the reference element (1.5 mM CaCb, inducing differentiation). 24h after, the cultures were stopped and the expression of the markers of interest (PCR-array mQPA H-NHEK-64) was evaluated by RT-PCR on the mRNAs extracted from the cell mats.
The data in Table I below indicate the proteins whose RNA was overexpressed in this test.
Tableau I :Table I:
The markers listed in the table have been significantly overexpressed by the treatment and are involved in differentiation, interkeratinocyte junctions such as desmosomes; in a general way in the barrier function of the epidermis.
The epidermis (RHE BA 01015-294) was systemically treated with Glycyrrhizza glabra extract (0.3%) for 96 hours. Immunolabelling was performed on 5pm sections. A computer analysis then made it possible to quantify the expression of the markers relative to the base 100 of the untreated cultures.
These quantifications indicate an increase of 56%, 20% and 63% respectively for claudinel, desmogleinel and E-cadherin. They confirm the previous study (Example 1) at the protein level and indicate that the extract of Glycyrrhizza glabra makes it possible to stimulate the establishment of solid interkeratinocyte junctions and communication / exchanges. The extract is therefore of interest for the cohesion, the establishment of the barrier function and the water retention via tight junctions (Claudine).
In this experiment the cells are human umbilical vein endothelial cells (HUVEC) cultured in M200 medium (Invitrogen M200500) on matrigel (BD Biosciences 354234) for 6 hours. The cells are either as such in the medium (middle control), treated with suramin (negative control), treated with VEGF (positive control), or treated with Glycyrrhizza glabra extract (2.10'2 and 0, 1%). After 6 hours of incubation, the pseudo-tubes formed are observed in microcopy and images are taken randomly to then be quantified (total area of the network) by image analysis.
Tableau II :Table II:

Treatment with Glycyrrhizza glabra extract stimulates the formation of pseudo tubes 36-48% depending on the treatment.
These results validate the use of Glycyrrhizza glabra extract in a cosmetic treatment aimed at moisturizing the skin.
