Abstract
La présente invention se rapporte à une composition cosmétique ou dermatologique caractérisée en ce qu'elle comprend une huile essentielle de Ciste sp et une huile essentielle d’Iris sp, dans laquelle ladite huile essentielle de Ciste sp est une huile essentielle de Cistus ladaniferus, et ladite huile essentielle d’Iris sp est une huile essentielle choisie parmi iris germanica L. et iris pallida Lam. La présente invention se rapporte également à une utilisation cosmétique d’une association d’huile essentielle de Ciste sp et d’huile essentielle d’Iris sp. pour une action cosmétique d’amélioration de la couleur, du teint et/ou de l’éclat de la surface de la peau, ainsi qu’à un procédé cosmétique non-thérapeutique comprenant l’application sur la peau, ses annexes et/ou les muqueuses de la composition cosmétique. Dans cette expérience, des cellules endothéliales transduites (GFP-HUVEC) pour mise en évidence du cytoplasme par marquage fluorescent sont cultivées avec des fibroblastes humains normaux dans un mélange de milieu Endothelial Cell Growth 2 supplémenté avec hEGF, hbFGF, R3 IGF, hydrocortisone, vitamine C et de milieu DMEM supplémenté avec 2 mM L-glutamine, 50 U/ml pénicilline, 50µg/ml streptomycine et 1% de sérum de veau fœtal au ratio 50/50. Les cellules en co-culture sont traitées ou non avec le VEGF à 100 ng/ml (Sigma, réf. V7259), ou encore avec le mélange 10-3% d’huile essentielle de ciste+3.10-4% de beurre d’iris, puis les 2 dilutions aux 1/10esuivantes. Les traitements ont lieu à J1 et J4 en milieu faible en sérum. 3 paramètres sont mesurés pour évaluer les capacités des cellules endothéliales à former leur réseau : l’Aire du réseau, les Points de branchement entre les cellules et la Longueur du réseau en temps réel grade à Incucyte SIII. Les pourcentages sont calculés en fonction du contrôle milieu. Le VEGF (référence positive) valide les manipulations.In this experiment, transduced endothelial cells (GFP-HUVEC) for detection of the cytoplasm by fluorescent labeling are cultured with normal human fibroblasts in a mixture of Endothelial Cell Growth 2 medium supplemented with hEGF, hbFGF, R3 IGF, hydrocortisone, vitamin C and DMEM medium supplemented with 2 mM L-glutamine, 50 U/ml penicillin, 50 μg/ml streptomycin and 1% fetal calf serum at a 50/50 ratio. The cells in co-culture are treated or not with VEGF at 100 ng/ml (Sigma, ref. V7259), or with the mixture of 10 -3 % cistus essential oil + 3.10 -4 % butter of iris, then the 2 following 1/ 10th dilutions. The treatments take place on D1 and D4 in a medium low in serum. 3 parameters are measured to assess the capacities of endothelial cells to form their network: the Area of the network, the Branching points between the cells and the Length of the network in real time grade at Incucyte SIII. Percentages are calculated based on the medium control. The VEGF (positive reference) validates the manipulations.
Le mélange beurre d’iris / huile essentielle de ciste présente un effet stimulant sur le processus d’angiogenèse par augmentation des 3 paramètres mesurés. A savoir, pour la concentration la plus faible, +38% de points branchements entre cellules, +22% d’aire occupée par les cellules et la longueur du réseau plus élevée de 15%. On observe un effet synergique du mélange, surtout à ces concentrations.The iris butter / cistus essential oil mixture has a stimulating effect on the angiogenesis process by increasing the 3 parameters measured.
