Description

Protocole
Matériels et réactifs - Fluorimètre (À(excitation)=485nm ; l(émission)=528nm)
Microplaques 96 puits - AAPH (CAS : 1898-66-4 ; MW= 394.32 g.moF1) - Sodium Huorescein (CAS : 518-47-8 ; MW= 376 .27 g.mol1) - Trolox (CAS : 53188-07-1 ; MW= 250.29 g.rnoL)
ABBA ALBA FIR TREE OIL, COMPOSITIONS AND USES
FIELD OF THE INVENTION
The present invention relates to the field of dermatology, dermocosmetics, cosmetics, nutraceuticals and / or the hair field. Also known as fir or white fir, Abies alba, it is the main resinous species of the mountainous region of central and southern Europe. The fir tree of the Vosges produces male and female cones in April and May. After fertilization, the female cones remain on the tree for at least three years, the time it takes for the seed to grow. When the large female cones are no longer fed by the mother plant, the drought of the cone causes the opening of the scales, in September-October. The scales deviate from the central rachis of the cone thus releasing large winged seeds,

The cells in the skin are sensitive to oxidative stress. The skin contains a natural reserve of antioxidants that help it repair itself, however as the skin is subjected to environmental aggressions, this reserve runs out very quickly. These aggressions generate free radicals, which cause a chain reaction that damages the skin and destroys vital components of the cells. The main environmental factors contributing to skin defects are sunlight, infrared radiation, air pollution, high or low temperatures, and cigarette smoke. The chronic exposure to solar radiation is mainly responsible for the premature aging of the skin. The excessive production of UV-induced ROS is a key factor in photo-aging. ROS are indeed responsible for oxidative damage affecting nucleic acids, proteins and lipids. The so-called photoovilli skin then has particular clinical and histological characteristics, it is deeply wrinkled, spotted, reddened, dehydrated, rigid and weakened. In addition, the production of ROS induced by the other oxidizing environmental factors also contributes to skin aging.
According to one embodiment, the oil of the invention or the composition of the invention may therefore be useful for protecting the skin and / or for combating the oxidation induced by UV rays.

Figure 2 is a graph showing the reduction of free radicals of a DPPH solution in the presence of increasing concentrations of Abies alba seed oil.
EXAMPLES
The present invention will be better understood on reading the following examples which illustrate the invention in a nonlimiting manner.
EXAMPLE 1 Preparation of the Abies alba Seed Oil The Abies alba seed oil is prepared according to the method described in the patent application WO 2011/045387.
In the following examples, the term "Abies alba seed oil" refers to the Abies alba seed oil prepared according to the present Example 1.
Example 2: Antioxidant Properties of the Seed Oil at'Abies Alba
1. DPPH test
Principle
The anti-radical potential of a substance can be tested using a colorimetric method using substitute radicals such as the 1,1-diphenyl-2-picrylhydrazil radical called DPPH °. At ambient temperature and in solution, the DPPH0 radical exhibits an intense violet coloration. Its passage to the non-radicular form, after saturation of its electronic layers is accompanied by a disappearance of this coloration according to the following equation: DPPHÙ + AH _________ DPPH-H + Ac
The decrease in the intensity of the staining of a solution containing the DPPtT radical, called the DPPH solution, is followed by colorimetric measurement at 517 nm. It makes it possible to realize the trapping capacity of the compounds studied compared to that of the DPPH °. The decrease in absorbance thus highlights the anti-radieal properties of the substance under study.
Protocol
A volume of 25 pL of seed oil & Abies alba is injected into hemolysis tubes protected from light. A DPPH solution at 6.10'5 mol / L is added to obtain a final volume of 1 mL. After stirring and incubating for 30 min at room temperature and protected from light, the absorbance of the solution is measured at 517 nm. Results
The absorbance results are shown in FIG. 1. A marked decrease in absorbance at 517 nm is observed in the presence of seed oil & Abies alba of the invention. In addition, the effect of the oil is dose dependent, up to a more than 5-fold reduction in absorbance compared to the control without oil (corresponding to the 0% concentration).
The corresponding free radical reduction is shown in Figure 2. The reduction of free radicals increases sharply with the concentration of the oil. At a concentration of 0.5%, free radicals are reduced by about 38%. When the oil is at a concentration of 1.25%, the reduction in free radicals rises to about 58%. The reduction of free radicals reaches about 82% at an oil concentration of the invention of 2.5%.
Thus, Abies alba seed oil of the invention has a significantly strong anti-radical and antioxidant activity.
2. TestORAC
Principle
The ORAC test makes it possible to determine the antioxidant potential of an extract by the ORAC (Oxygen Radical Absorbance Capacity) kinetic fluorometric monitoring method. This method uses a fluorescent probe, fluorescein, to monitor antioxidant activity.
This method therefore consists in measuring the protection exerted by the extract against the oxidation of fluorescein, caused by a generator of free radicals, here Γ AAPH (2,2'-azobis-2-methyl-propanintidaniide, dihydrochloride) which produces free radicals of the peroxyl type. AAPH produces free radicals during the reaction that damage the fluorescent probe and decrease the intensity of fluorescence. It is the change in the degree of fluorescence intensity that testifies to the damage caused by free radicals, and which makes it possible to measure the potential for antioxidant protection. Kinetic monitoring is performed and continues until depletion of the antioxidant activity of the extract. The remaining free radicals then destroy the fluorescent probe until the total extinction of the fluorescence. In order to quantify the antioxidant protection potential of the extract,
Making under the curve of it is compared to making under the curve of a reference antioxidant, here Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid). Trolox is a water soluble analogue molecule of vitamin E. The results are given in Trolox equivalents.
Protocol
Materials and reagents - Fluorometer (At (excitation) = 485nm, l (emission) = 528nm)
96 well microplates - AAPH (CAS: 1898-66-4, MW = 394.32 g.moF1) - Sodium Huorescein (CAS: 518-47-8, MW = 376.27 molol) - Trolox (CAS: 53188-07 -1; MW = 250.29 g.rnoL)
The AAPH solution is prepared at a concentration of 34.8 mM in phosphate buffer. Trolox and fluorescein solutions are prepared at a concentration of 1 mM in phosphate buffer. All solutions are kept away from light. The fluorescein working solution is prepared extemporaneously from the 1 μM solution with phosphate buffer to obtain a final concentration of 100 nM. Method A 20 μl of Abies alba seed oil of known concentration are added to 120 μl of 100 nM fluorescein working solution. The kinetics is then triggered with the addition of 60 μl of AAPH solution (time 0) for a total reaction volume of 200 μl. A fluorescence measurement is performed every minute for 60 min at an excitation wavelength of 485 nm and emission of 528 nm. Each sample is prepared in triplicate and the result corresponds to the average of the 3 values recorded. If an absorbance value deviates by more than 10% from other values, it should not be considered for the calculation of mean fluorescence.
Negative and positive controls for which the extract of the invention is replaced in equivalent volume respectively by phosphate buffer and a Trolox solution. The area under the curve (AUC) of each sample concentration, oil or control, is calculated according to formula (1):
where fn is the fluorescence at time n and / 0 is the fluorescence at time 0. The area under the net curve (net AUC) is then calculated according to formula (2): (2) net AUC AUAUCfjianc
In order to obtain a Trolox equivalent result, a calibration straight line from the Trolox solution of different concentrations (from 0 to 100 μΜ) for which to make under the net curve is measured is carried out. The linear regression corresponding to this line makes it possible to determine the net AUC of the extract, that is to say its antioxidant potential, expressed in μ mol Trolox equivalent. Results
The results of the ORAC test of an alba seeds seed oil prepared according to Example 1 indicate a very high antioxidant potential (data not shown). The extract of the invention therefore has antioxidant properties.
