Trolox and fluorescein solutions are prepared at a concentration of 1 mM in phosphate buffer. All solutions are kept away from light. The fluorescein working solution is prepared extemporaneously from the 1 mM solution with phosphate buffer to obtain a final concentration of 100 nM. Method A 120 μl of fluorescein working solution at 100 nM are added 20 μl of Abies alba seed oil of known concentration. The kinetics are then triggered with the addition of 60 μl of AAPH solution (time 0) for a total reaction volume of 200 μl. A fluorescence measurement is performed every minute for 60 min at an excitation wavelength of 485 nm and emission of 528 nm. Each sample is prepared in triplicate and the result corresponds to the average of the 3 values recorded. If an absorbance value deviates by more than 10% from other values, it should not be considered for the calculation of mean fluorescence.
