Ces fonctions permettent de soulager une douleur chez des OH20 personnes souffrant d'une polyarthrite ou d'une arthrose. Compte tenu de ces propriétés pharmacologiques, la rhéine a été formulée en une composition pour traiter ces maladies. Poids sec du produit : 160 g Préparation de l'acide chromique : 500 g de Na2Cr207 sont dissous dans de l'eau (1500 ml) et H2SO4 conc. (333 ml) est ajouté à cette solution à 0-10 °c. Cette solution est utilisée telle quelle. EXEMPLE 2 Au mélange de rhéine (160 g) dans de l'anhydride propionique (1600 ml), on ajoute goutte à goutte de l'acide sulfurique (48 ml) à 25-40 °C. Après l'addition, la réaction est maintenue à 25-40 °C et suivie par CLHP. Au terme de la réaction, la masse réactionnelle est étouffée dans de l'eau à 5-10 °C, le produit est isolé par filtration et lavé avec un excès d'eau puis avec de l'acétone (160 ml). La rhéine dipropionylée est séchée sous vide à 25-30 °C. Poids sec du produit : 210 g. Données physiques : Point de fusion : 221 °C IR (dans KBr) : 1148 cm-1, 1255 cm-1, 1594 cm-1, 1685 cm-1, 1766 cm-1, 2941 cm-1. RMN H (DMSO-d6) : S 1,23-1,81 (6H), 2,54-2,81 (4H), 7,66-7,7 (1H), 7,9-8, 2 (3H), 8,5 (1H). The final aspect of the present invention is the purification of diacerein. The diacerein which is obtained at the end of the above process is crude and is subjected to a final purification. It is dissolved in N-methylpyrrolidone (NMP) at about 70 ° C. The filtrate obtained is contacted with methanol at about 60 ° C to precipitate the product. If necessary, repeat this procedure before dissolving the diacerein again in NMP and precipitating it with water. At the end of these purification steps, the diacerein is substantially pure and free of non-polar impurities as well. The impurity profile of the final product is: • monoacetylated rhein - 1: less than 0.15% • monoacetylated rhein - 2: less than 0.15% • Rhine: less than 0.15% • Any unknown impurity: less than 0 The diacerein that is obtained is 99.5% pure, as shown by HPLC analysis. The invention is now described by the following nonlimiting examples. The process of the invention can be illustrated by the use of dipropionyl rhein (of formula III) as a diacylated intermediate product in the synthesis of diacerein. EXAMPLE 1 To the solution of aloe-emodin (200 g) in tetrahydrofuran (2000 ml) was added chromic acid at 0-40 ° C. The reaction is maintained at 25-40 ° C and monitored by HPLC. At the end of the reaction, the THF is distilled completely, the reaction mass is diluted with water (2000 ml) and the solid filtered off. EXEMPLE 3 A la solution transparente de rhéine dipropionylée (210 g) dans du THF (1600 ml) à 60-65 °C, on ajoute du toluène (1680 ml). (160,5 ml) jusqu'à pH = 1. Le solide est séparé par filtration, lavé à l'eau et séché sous vide à 60-65 °C. Poids sec de rhéine : 111 g. EXEMPLE 4 Au mélange de rhéine (111 g) dans de l'anhydride acétique (1100 ml), on ajoute de l'acide sulfurique (33,3 ml) goutte à goutte tout en maintenant la température entre 25-45 °C. Au terme de l'addition, la température de la réaction est maintenue à 25-40 °C et elle est suivie par CLHP. Au terme de la réaction, la masse réactionnelle est stoppée dans de l'eau à 5-10 °C. Le produit est isolé par filtration et lavé avec un excès d'eau. Le solide mouillé est soumis à un traitement avec du charbon dans de la N-méthyl-2-pyrrolidone (charbon enlevé par filtration), et du méthanol (1100 ml) y est ajouté à 60-65 °C. La masse réactionnelle est refroidie à 25-30 °C et le solide séparé par filtration. Le solide mouillé est à nouveau dissous dans de la NMP (880 ml) à 70-80 °C et, à cette solution transparente, on ajoute du méthanol (880 ml) à 60-65 °C. La solution est refroidie à 25-30 °C pour précipiter le solide, qui est ensuite séparé par filtration. Cette purification est répétée jusqu'à ce que les impuretés monoacétylées soient en dessous de la limite recherchée. Ce solide mouillé est ensuite dissous dans de la N-méthyl-2-pyrrolidone (640 ml) à 70-80 °C et la solution transparente est lentement ajoutée sur de l'eau (3200 ml) préchauffée à 80-90 °C. This wet solid is then dissolved in N-methyl-2-pyrrolidone (640 ml) at 70-80 ° C and the clear solution is slowly added to water (3200 ml) preheated to 80-90 ° C. The suspension obtained is cooled to 25-30 ° C, the solid is filtered off and dried under vacuum at 50-55 ° C. Dry weight of diacerein: 55 g.
