Abstract
Description
Domaine Technique :
Technical area :
The invention relates to a microalgae extract having anti-free radical activity due to the presence of SOD (superoxidismutase) like, coupled with the presence of sulfated polysaccharides. These two activities are used alone or together to enter into compositions useful in the fields of pharmacy, cosmetics, dietetic or food nutrition and in the fight against culture pests.
Previous techniques:
Nowadays a lot of work is related to the obtaining and properties of anti-free radicals. Researchers recently found sources from the plant world because of infectious substances that may be encountered in animal or human extracts. Cloning techniques are being studied. A modern source is used with the culture of micro-algae. The usual goal is to extract algal biomass to find interesting components. These crops are expensive and take a long time. The culture medium is discarded, which explains a large part of the cost. The present invention stems from the unexpected properties that the Applicant has found not in the biomass, but in its culture medium. The biomass being preserved and used to obtain known components such as proteins, lipids, vitamins, mineral salts. Therefore, the invention by valuing an industrial waste in addition to the unexpected properties makes the exploitation of the micro-algae in question more economical.
Description sommaire de l'invention :
Brief description of the invention:
The invention consists in the preparation of extract contained in the culture medium of micro-algae. These cultures are known to those skilled in the art. However, in the particular case of the invention, arrangements are made to increase the defenses of the alga in order to increase the richness of the components that can be sought. The culture is carried out under controlled conditions and axenically, without a pesticide type product which would generate the quality of the objective extract of the invention. Particularly after proper culture and according to the invention, the alga undergoes a metabolic forcing. The physiological reaction of the alga to a supersaturation in O 2 would in fact be the cause of the overproduction of the anti-free radical compounds in order to protect themselves against oxygen in order to avoid structural degradations such as lipid peroxidation. The Applicant has developed its invention from cultures chosen from the Rhodophycae group and in particular the
Porphyridium cruentum (PC). The extract obtained has two biological activities due to superoxydismutases like SODL and to polysaccharides (PS). The interest of the extract lies in the simultaneous presence of the two activities. They act in synergy, the component
SODL acting as an anti-inflammatory thereby facilitating the tissue reconstruction brought by the PS, in particular for cosmetic applications. These two effects are distinct in fact, two extracts can be prepared, one containing only the SODL actor, the other containing the PS. The measures of specific activities show that they are present in isolation, but that their combination or simultaneous use amplifies each of the anti-inflammatory and regenerating effects.
1) Un extrait du milieu de culture de micro-algues présentant une activité SOD like d'au moins 30 U/ml.Thus and according to the invention, it is possible to conceive treating compositions containing in an effective amount:
1) An extract from the microalgae culture medium having an SOD like activity of at least 30 U / ml.
3) A microalgae culture extract having an SOD like activity of at least 30 U / ml and a content of sulphated polysaccharides at least equal to 1 mg per ml. The extract can therefore validly and in effective amount enter into compositions for cosmetic, dietetic or pharmaceutical use intended for! a prevention or treatment of aging and degeneration and in pathologies linked to oxidative stress for humans or animals. The extract can intervene by strengthening the antioxidant activities of other plant extracts. Thus with extracts with peroxidase activity, a net increase in this activity has been observed by adding about 10% of the extract according to the invention to them. This property is interesting in the biological fight against crop pests.
Ways to realize the invention:
The invention, although simple in its description, will be clearly understood with the aid of the examples which illustrate its multiple applications and its:
Example 1. Obtaining the extract from PC
In a 150 liter reactor, the algae is cultivated in a Conway type medium, artificial seawater. The inoculation is carried out by 5 l of culture at 5 million cells per milliliter.
The agitation and supply of C02 is done by air bubbling. The lighting is from 50 to 100 micromoles m-2 S-1. After 12 days, when the cell density is optimal, 100 mg per liter of KH C03 are added. The polyethylene sheath is closed, and the culture is allowed to continue for 2 days in order to create a metabolic forcing. The algae are separated by centrifugation from the culture medium. The extract is then obtained by filtration from the culture medium. This contains 30 liters of SOD like per liter, measured in the SOD-525 kit and approximately 1 mg of PS per ml. Half of the extract is dried by lyophilization to prepare compositions such as tablets. The other half is sterilized at 120 ° C. for 20 minutes and packaged in sterile bottles, without this operation modifying the SOD activity or the content of sulfated polysaccharides.
Example 2: Separation of activities:
From 10 liters of extracts of Example 1, the SOD like part is separated by solvent precipitation (ethyl alcohol). The extract is thus obtained in two sub-variants, one containing the anti-free radical activity not altered by the separation, the other the presence of the polysaccharides. As in Example 1 and by dehydration, the extract and its variants can be kept liquid or in the form of soluble powder.
