La présente invention concerne un procédé de prélèvement et de concentration sur une cartouche adsorbante, de produits volatils, notamment d'alcanes, tels l'éthane et le pentane issus de la peroxydation des acides gras polyinsaturés, en particulier de la peroxydation des acides gras polyinsaturés des membranes cellulaires, notamment de l'acide linoléique et de l'acide linolénique en vue du dosage de ces produits volatils, notamment de ces alcanes après désorption thermique et chromatographie en phase gazeuse.Method for sampling and concentrating

Le but recherché était de trouver un procédé de nature à pouvoir se substituer aux procédés classiques généralement mis en oeuvre pour la détermination du vieillissement cellulaire ces procédés qui consistaient en un dosage d'autres sous-produits issus de la peroxydation des acides gras polyinsaturés des membranes cellulaires, tels que des diènes conjugués ou des aldéhydes, notamment le malondialdéhyde (MDA), se sont en effet révélés peu fiables et présentent l'inconvénient d'être invasifs, c'est-à-dire de nécessiter la mise en oeuvre de prélèvements par piqûre, ce que l'on cherche de plus en plus à éviter pour limiter les extravations vasculaires chez le patient et pour protéger le personnel hospitalier contre les risques de transmission virales (hépatites, sida...). La mise en oeuvre de ce procédé, qui implique un dosage de l'air expiré par un sujet, présente l'inconvénient d'être particulièrement lourde et d'obliger de convoquer le sujet sur le lieu même de l'analyse, ce qui est parfois difficilement réalisable. De plus, on s'est rendu compte que les résultats obtenus sont faussés par une contamination par l'air ambiant, qui peut contenir un pourcentage non négligeable d'éthane et de pentane.Il serait donc souhaitable, pour pouvoir obtenir des mesures utilisables, d'éliminer cette source de contamination exogène et donc de recueillir exclusivement l'air alvéolaire ou endogène, c'est-à-dire l'air expiré par un sujet à la fin d'une expiration forcée.
