Abstract

L-(-)-4-Hydroxyproline (I) is obtd. by (a) hydrolysing an opt. pretreated collagen-contg. material; (b) decolorising the resulting protein hydrolysated and passing it through a weakly basic ion exchange to retain glutamic acid, aspartic acid and inorganic anions; (c) contacting the effluent with as strongly acidic ion exchanger to retain 75-95% of a amino acids present in the soln.; (d) eluting with H2O and concentrating the eluate to give a soln. with a (I) concn. of 180-350 (pref. 230-280) g/l; and (e) decolorising the soln., adjusting to pH 3.8-4.6, and isolating (I) in crystalline form. The process requires only small amts. of ion exchanger, avoids the use of buffers, and can be applied to materials with a low collagen content.
