Claims (10)

1. Förfarande för bestämning av protrombin i biologiskt material, säsom blodplasma, genom omvandling tili trombin, enzymatisk spjälkning av ett trombinsubstrat och mätning av en spjälkningsprodukt, kännetecknat av att inku-bationen av testlösningen sker genom att tillsätta faktor Xa, företrädesvis faktor Xa frän människa.
2. Förfarande enligt patentkravet 1, kännetecknat av att faktorn Xa är framställd sä att man behandlar blodplasma, företrädesvis blodplasma frän människa, med en protrombinaktivator, efter centrifugering tillsätter ett pro-teinadsorbens, eluerar ett precipitat med ett proteineluerings-medel och försätter eluatet med en faktor-X-aktivator samt even-tuellt med ett lösligt kalciumsalt.
3. Förfarande enligt patentkravet 1 eller 2, kännetecknat av att trombinsubstratet är ett syntetiskt peptid-p-nitroanilid-derivat.
4. Förfarande enligt patentkravet 3, kännetecknat av att trombinsubstratet är N-Tos-Gly-Pro-Arg-pNA, N-Cbz-Gly-Pro-Arg-pNA, H-D-Phe-Pip-Arg-pNA eller Bz-Phe-Val-Arg-pNA.
5. Reagens för bestämning av protrombin i biologiskt material, säsom blodplasma, innehällande ett trombinsubstrat och en protrombinaktivator, kännetecknat av att protrom-binaktivatorn utgöres av faktor Xa, företrädesvis faktor Xa frän människa.
6. Reagens enligt patentkravet 5, kännetecknat av att faktorn Xa är framställd sä att man behandlar blodplasma, företrädesvis blodplasma frän människa, med en protrombinaktivator, efter centrifugering tillsätter ett proteinadsorbens, eluerar ett precipitat med ett proteinelueringsmedel och försätter eluatet med en faktor-X-aktivator samt eventuellt med ett lösligt kalciumsalt.
