Claims (2)
1, Förfarande för pävisande av hepatitis B ytantigen i manniskoblod, varvid ett blodprov, fr«in vilket de röda blodkropparna har avlSgsnats, blandas med en d(agnostisk sammansSttnihg.vilken best§r av en vattensuspension som inneh$ller praktiskt taget likadana kulformiga polystyrenpartikJar med en diameter av 0,05-2,0 fjm och belagda med en renad antikropp specifik mot hepatitis B ytantigen, varvid denna antikropp har isolerats ur serum av marsvin, vilka har immuniserats med renat hepatitis B ytantigen isolerat ur minniskoblod och varvid denna antikropp inte innehSlIer andra antikroppar specifika mot mSnniskoserumsproteiner, och varvid vattensuspensionen Sven inne-hSller i Iöst form normalt marsvinserum, vilket inte innehäller en antikropp specifik mot hepatitis B ytantigen och varvid polystyrenpartiklarna har be-lagts med en s&dan mängd av nSmnda antikropp, som just Sr tillräcklig för att förhindra autoaggregation av partiklarna, och blandningen av blodprovet och suspensionen undersöks, huruvida dSri förekommer agglutination, k a n n e -t e c k n a t dSrav, att (a) antikroppen, som har använts som belSggningen av polystyrenpartiklarna, har renats pä ett kSnt sätt genom att bringa en lösning av antigen/ antikropp-komplexet, som har framställts genom att blanda marsvinserum inne-h31lande antikroppen med människoserum, vilket innehSller antigenhaltiga antikroppar specifika mot hepatitis B ytantigen, i kontakt med ändan med den större tStheten av ett flytande medium innehSllande en täthetsgradient, varefter det flytande mediet uitracentrifugeras , tills distributionen av serimkomponenterna i tSthetsgradienten har nätt jämvlktsti11ständ, och därefter separeras frak-tionerna, vilka innehSller antikroppen specifik mot hepatitis B ytantigen. (b) alla blodprov, väri agglutination observeras, neutraliseras gencm att blanda dem med en tili människans hepatitis bunden antikropp, (c) frän proven erhillna ί steget (b) separeras reumafaktorn genom att blanda proven med latexkulor, vilka har belagts med mSnniskans gammaglobulin, och kulorna tillsammans med reumafaktorn absorberad dSri separeras, och (d) de nSmnda proven var? agglutination observerats, Sterblandas med den nSmnda diagnostiska sammansättningen.
