Abstract
Procedure for the preparation of controlled oral gastric residence oral drug formulations, of the type comprising a drug core coated with a pharmaceutically acceptable polymeric film, crosslinked, hydrohydratable, water permeable and adapted to swell in the presence of gastric fluids, characterized in that it comprises the steps of: (a) coating, for example by immersion, a drug core with a solution, for example ethyl acetate, consisting of a prepolymer and a crosslinking agent in a ratio of 8 parts by weight of the prepolymer to 0.15 - 4 parts by weight of crosslinking agent; and (b) crosslinking the resulting coated drug core, under crosslinking conditions, for example by subjecting the coated drug core to a temperature between 10 and 90ºC. at a relative humidity of 30-95%, preferably between and 200 hours; and in that the prepolymer is prepared by copolymerization of (i) 20-90 parts in step of a substituted or unsubstituted dicarboxylic acid or carboxylic acid anhydride, with 1-3 carbon atoms between the 2 carboxylic groups, preferably maleic anhydride; and (ii) 10-80 weight carriers of an alkene of 2 to 6 carbon atoms, styrene or an alkyl vinyl ether in which the alkyl radical contains 1 to 16 carbon atoms, preferably methyl vinyl ether.
