Abstract

Procedure for the preparation of bis-basic xanthene and xanthene-9-one ketones, having antiviral, having the formula **(See formula)** where Z is a member selected from the group consisting of oxygen or H2; each A is a straight or branched chain of alkylene, containing from about 1 to 6 carbon atoms; and each Y is a selected member of the group consisting of **(See formula)** individually selected from the group consisting of hydrogen, lower alkyl, having from 1 to 6 carbon atoms, cycloalkyl, having from 3 to 6 carbon atoms, alkenyl from 3 to 6 carbon atoms and having the vinyl unsaturation in a position other than that position -1 of the alkenyl group; or **(See formula)** whole 4 to 6 and is a member selected from the group consisting of hydrogen or lower alkyl of from about 1 to about 4 carbon atoms and can bind to any of the carbon atoms in the heterocyclic group; or (C) the group (see formula), wherein X is a member selected from the group consisting of oxygen or NR4, and R4 is hydrogen or lower alkyl of from 1 to about 4 carbon atoms, or a pharmaceutically acceptable acid addition salt thereof, characterized by comprising the selected processes of (I) reacting in a solvent, a xanthene derivative of the formula **(See formula)** With a compound of the formula Y-h where Z, A and Y are as defined above and Hal is chlorine, bromine or iodine; (II) when A is an alkylene chain of 3 to 6 carbon atoms and Z is H2, reacting a xanthene derivative of the formula **(See formula)** with a Grignard reagent of the formula XMg (CH2) p Y where Y is as defined above first, except those containing a hydrogen attached to the nitrogen atom, X is chlorine or bromine, p is an integer from 3 to 6, (see formula) where R5 and R6 are hydrogen or lower alkyl or -NR5R6 taken together form a heterocyclic monocyclic saturated group, and R7 is lower alkyl or aryl; (III) when A is -CH2CH2- and Y is one of the groups first defined herein, except those containing two hydrogens on the nitrogen atom, react, in the presence of formaldehyde, a xanthene derivative of the formula **(See formula)** with a compound of the formula Y-h where Z is oxygen or H2; (IV) when Z is oxygen, oxidizing the corresponding bis-basic xanthene ketone compound of the formula **(See formula)** where A and Y are as first defined above; (V) when Y is -NH2, Z is H2 and A is an alkylene chain of 2 to 6 carbon atoms, hydrolyzing and decarboxylating a bis-phthalimido derivative of the formula **(See formula)** where R8 is lower alkyl; and (VI) Whenever the pharmaceutically acceptable acid addition salt thereof is desired, react a pharmaceutically acceptable acid with the base compound.
