Abstract
The invention comprises 2a -alkyl-19-nortestosterone derivatives having the formula: <FORM:0947000/C2/1> in which Y represents an alkyl group having from 1-7 carbon atoms and R is an acyl radical derived from a lower organic carboxylic acid or Y represents an alkyl group having from 2-7 carbon atoms and R is an acyl radical derived from a lower organic carboxylic acid or a hydrogen atom and a process for the production thereof in which a D 1(9), 7-3-methyl-7-amino-hexahydro-(31-acyloxy) -cyclopentano - [2,1,C] napthalene derivative having the formula: <FORM:0947000/C2/2> (wherein R1 and R11 which may be the same or different each represents an alkyl group having 1-6 carbon atoms or together with the nitrogen atom represent a heterocyclic ring structure, optionally mono-oxygenated and Ac denotes the acyl radical derived from a lower organic carboxylic acid) is reacted with a 2-alkyl - 1,3 - dichloro -2-butene to form a D 1(9)-3-methyl-7-keto-8-(b -alkyl - g - chloro - 211 - butenyl)-octahydro -(31-acyloxy)-cyclopentano-[2,1,C]- napththalene having the formula: <FORM:0947000/C2/3> which is then subjected to hydrolysis to form a corresponding D 8 - 3 - methyl - 7 - keto - 8 - (b -alkyl - g - keto - butyl) - octahydro - (31 - acyloxy) -cyclopentano - [2,1,C]-naphthalene which is then subjected to treatment with a reducing agent to eliminate the double bond at the 8-position and form a corresponding 3-methyl - 7 - keto - 8 - (b -alkyl - g - keto - butyl) - decahydro - (31 - acyloxy) -cyclopentano - [2,1,C] - naphthalene which is then cyclised by treatment with an acid dehydrating agent to yield the corresponding 17-ester of a 2a - alkyl - 19 - nor - testosterone which is optionally thereafter saponified to the corresponding 2a -alkyl - 19 - nor - testosterone. Reduction may be effected by hydrogenation in the presence of palladinized charcoal, cyclisation with concentrated hydrochloric acid and the subsequent optional saponification with methanolic potassium hydroxide.
