Abstract
Un fragmento de anticuerpo que reconoce específicamente un complejo inmunitario entre el tetrahidrocannabinol (THC) y el anti-THC, y que comprende las regiones determinantes de complementariedad (CDR) de una cadena ligera y de una cadena pesada de anticuerpo, en donde dichas regiones de la cadena ligera tienen las secuencias aminoacídicas de los aminoácidos 23 a 35, 51 a 57 y 93 a 100 de la SEQ ID n.º 3 y dichas regiones de la cadena pesada tienen las secuencias aminoacídicas de los aminoácidos 29 a 35, 50 a 60 y 99 a 109 de la SEQ ID n.º 4.
Description
inmovilizados (IMAC). El Fab 104 se purificó mediante Sepharose quelante con Ni2+ (Amersham Pharmacia Biotech). La pureza del Fab 104 se comprobó por SDS-PAGE.
Ejemplo 3
Un inmunoensayo homogéneo para el THC por transferencia de energía por resonancia de fluorescencia resuelta en 5 el tiempo (TR-FRET, por su nombre en inglés)
El fragmento Fab T3 anti-THC se marcó con europio mediante el kit LANCE Eu-W1024 ITC chelate (Wallac). El tampón del T3 marcado se cambió a Tris a 50 mM, pH 7,8, y NaCl al 0,9%. El fragmento Fab 104 contra el complejo inmunitario T3+THC se marcó con Alexa Fluor 647 con el kit de marcación de proteínas con Alexa Fluor (Molecular Probes, Inc.).
10 A los pocillos de microtitulación con 60 µl de saliva se les añadieron 1 µg del Fab T3 anti-THC marcado con Eu y 1,5 µg del Fab 104 contra el complejo inmunitario marcado con Alexa Fluor 647. A los pocillos se les añadieron muestras de 40 µl de saliva enriquecida con 0, 3,75, 7,5, 15, 30, 60 y 120 ng/ml de las siguientes drogas: D9-THC, 11-OH-D9-THC, 11-nor-D9-THC-COOH, heroína y anfetamina. Después de una mezcla breve, se midió la transferencia de energía por resonancia de fluorescencia resuelta en el tiempo (TR-FRET) con un fluorímetro Victor
15 V (Perkin Elmer Wallac). Los resultados se muestran en la figura 3.
La muestra de saliva enriquecida con THC da un valor fluorescente alto, mientras que los metabolitos del THC dan valores de fluorescencia significativamente más bajos, y la heroína y la morfina dan valores de fluorescencia cercanos a la fluorescencia de fondo detectada en el control (fragmentos T3 y 104 marcados a lo que no se ha añadido ninguna droga). El Fab 104 se fija al complejo inmunitario formado entre el T3 y el THC con una
20 especificidad y sensibilidad extremadamente altas. La prueba es capaz de detectar el THC muy por debajo del nivel de umbral (20 ng/ml en la saliva) de las pruebas rápidas que actualmente están disponibles en el mercado.
