Abstract
Procedimiento para preparar una fracción plaquetaria, que comprende: - preparar sangre placentaria, - centrifugar la sangre placentaria a una rotación que oscila de 300 a 900 g durante un periodo que varía de 5 a 20 minutos; - centrifugar la sangre placentaria a una rotación que oscila de 50 a 200 g durante un periodo que varía de 5 a 15 minutos.

El sedimento resultante se resuspendió en 5,5 ml de dicha porción de plasma a fin de obtener una concentración final de 2x109 plt/ml.
A continuación, la muestra de PRP resultante se dejó reposar a temperatura ambiente durante 30 minutos.
El protocolo del ejemplo 1 se repitió a fin de obtener una muestra de 5,5 ml de fracción plaquetaria de PRP a partir de sangre placentaria que tiene una concentración de 2x109 plt/ml.
Dicha muestra se activó añadiendo 1 ml de gluconato de calcio y un comprimido de batroxobina liofilizada correspondiente a 5 unidades Bethesda y aproximadamente 0,9 unidades NIH trombínicas.
Se esperó durante 10 minutos a fin de obtener un gel plaquetario.
El gel plaquetario preparado de acuerdo con el ejemplo 2 se centrifugó a 2000 g durante 10 minutos y el sobrenadante se filtró con un filtro que tenía poros de 20 micrómetros a fin de obtener un lisado plaquetario. Un correspondiente lisado plaquetario a partir de sangre periférica se preparó adicionalmente del mismo modo.
Se aislaron 2x105 células madre mesenquimatosas a partir de tejido adiposo de adultos humanos (con consentimiento informado). Dichas células madre mesenquimatosas se sembraron en un cultivo de (medio de Eagle modificado por Dulbecco) y un 10 % v/v de lisado plaquetario obtenido a partir de sangre placentaria y un 1 % v/v de penicilina/estreptomicina. Un cultivo que comprendía un 10 % v/v de lisado que derivaba de sangre periférica se prepararó del mismo modo. Las células se mantuvieron en cultivo durante tres semanas, reemplazando la mitad del medio de cultivo una vez por semana.
Después de tres semanas, las células se caracterizaron mediante análisis citofluorimétrico con el dispositivo Cytomics C500 y el software CXP de Beckam Coulter.
Las células madre mesenquimatosas obtenidas después de 3 semanas se sembraron en DMEM+ un 20 % v/v de FBS (suero fetal bovino) a 2,1x104 células/cm2 y se incubaron a 37 °C con un 5 % v/v de CO2.
Una vez han alcanzado la confluencia, se estimulan las células con tres ciclos de diferenciación. Cada ciclo consiste en cultivar las células obtenidas en dichos medios de cultivo con medio de inducción de adipogénesis (Lonza) durante tres días, seguido de 1-3 días de cultivo en medio de mantenimiento adipogénico (Lonza).
Después de tres ciclos completos, las células se mantienen en cultivo con medio de mantenimiento adipogénico durante siete días, reemplazando el medio cada 2-3 días. Finalmente, con el propósito de observar la diferenciación real las células se lavan con PBS, se fijan en un 10 % v/v de formaldehído durante 10 minutos y se colorean con el aceite rojo O durante 5 minutos.
Solo se colorean las vacuolas lipídicas. Los resultados se pueden comparar visualmente en la figura 1, en la que la figura 1A muestra el crecimiento de la fracción plaquetaria de acuerdo con la invención y la figura 1B muestra el crecimiento con la fracción plaquetaria a partir de sangre periférica.
Más allá de la prueba con diferenciación adipogénica, la diferencia en eficacia de la fracción plaquetaria también se midió con diferenciación osteogénica. Las células madre mesenquimatosas obtenidas a partir de los cultivos después de 3 semanas, como anteriormente, se sembraron en una concentración de 2,1x104 células/cm2 en DMEM + un 20 % v/v de FBS (suero fetal bovino) y se incubaron a 37°C con un 5 % v/v de CO2.
Después de 24 horas, el medio de cultivo se reemplazó con medio de inducción de osteogénesis (Lonza). Las células se mantienen en cultivo durante 3 semanas, reemplazando el medio con medio recién preparado cada 2-3 días.
Finalmente, con el propósito de observar la diferenciación real las células se lavan con PBS y se fijan con un 70 % v/v de alcohol etílico (almacenado a -20°C) durante 1 hora y se colorean con rojo de alizarina S 40 mM (pH = 4,1).
Solo se colorean los depósitos de calcio. Los resultados se pueden comparar visualmente en la figura 2, en la que la figura 2A muestra el crecimiento de la fracción plaquetaria de acuerdo con la invención y la figura 2B muestra el crecimiento con la fracción plaquetaria a partir de sangre periférica.
La comparación para ambos tipos de diferenciación muestra que el uso de la fracción plaquetaria que deriva de sangre placentaria tiene un efecto evidente sobre el potencial de dichas células madre mesenquimatosas.
