LA INVENCION TRATA DE CONJUGADOS A PARTIR DE UNA COMPOSICION FLUORESCENTE, UN LINKER Y UNA PROTEINA. LA INVENCION TRATA ADEMAS DE LA PRODUCCION DE DICHOS CONJUGADOS, ASI COMO DE SU USO.
Description
Conjugado de albúmina y un colorante de xanteno
para diferenciar tejido enfermo de tejido sano.
La invención se refiere a conjugados para
diferenciar tejido enfermo de tejido sano, a procedimientos para su
preparación y a la utilización de los mismos.
En el tratamiento de tejidos enfermos, la
extirpación de los mismos es con frecuencia un paso esencial. Para
ello es necesario que el cirujano identifique con exactitud dónde
termina el tejido enfermo y comienza el tejido sano. Sin embargo,
frecuentemente esto no es posible. Por ello se pasan por alto
ramificaciones del tejido enfermo que constituyen después la base
para una nueva formación de tejido enfermo.
Rushfeldt et al. (Cancer Res., Vol. 53, Nº 3, pp.
658-662, 1993) describen un método para marcar
carcinomas de colon con fluoresceína utilizando receptores
especiales similares a la lectina para la endocitosis.
El documento ``Biochemicalien Organische
Verbindungen Voor Research en Diagnostica, 1994, Sigma, NL, p. 71''
menciona un conjugado de albúmina sérica humana con
fluoresceína.
Por consiguiente, la presente invención tiene por
objetivo poner a disposición un producto con el que se pueda
diferenciar tejido enfermo de tejido sano.
Esto se logra de acuerdo con la invención
mediante los objetos de las reivindicaciones. Por consiguiente, un
objeto de la presente invención consiste en un conjugado de un
colorante de xanteno, un engarce y albúmina sérica humana nativa,
siendo el engarce cloruro cianúrico.
Como representantes de los colorantes de xanteno
se mencionan: fluoresceína, amino-fluoresceína
(AFI), eritrosina, amino-eritrosina (AEros), eosina
amarillenta y amino-eosina amarillenta (AEs). Estos
compuestos también pueden presentar fotoactividad, como por ejemplo
la AEros. Típicamente, la actividad fotodinámica se excita con una
longitud de onda entre 500 y 550 nm (AEros: 533 nm).
Los conjugados según la invención pueden
presentar varios colorantes de xanteno, que pueden ser iguales o
diferentes entre sí.
Los conjugados según la invención presentan
albúmina sérica humana (HSA). Esta proteína produce una acumulación
del conjugado en tejidos enfermos, en particular en tejido tumoral y
en pequeños vasos superficiales, por ejemplo de neovascularizaciones
en el área de la córnea.
La expresión ``engarce'' comprende cloruro
cianúrico (Cy).
Los anteriores componentes de un conjugado según
la invención se indican como eductos. En el conjugado se encuentran
en forma derivada.
La Figura 1 muestra conjugados preferentes según
la invención.
Los conjugados según la invención se pueden
producir de acuerdo con procedimientos habituales mediante los
cuales el colorante de xanteno, el engarce y la albúmina sérica se
unen entre sí, en particular de forma covalente. Véase por ejemplo
la preparación de los conjugados de los Ejemplos 1 a 3.
Los conjugados según la invención se caracterizan
por una vida media prolongada en el organismo. Además, los
conjugados según la invención se acumulan en tejidos enfermos, en
particular en tejido tumoral y en pequeños vasos superficiales, por
ejemplo de neovascularizaciones en el área de la córnea. El
compuesto fluorescente se excita mediante luz, por ejemplo una
lámpara UV, con lo que los tejidos enfermos se hacen visibles,
mientras que, por el contrario, los tejidos sanos, en los que no se
acumulan los conjugados según la invención, no se hacen visibles.
Por consiguiente, se posibilita la delimitación de tejidos enfermos
con respecto a tejidos sanos.
Los conjugados según la invención también se
caracterizan porque en ellos se puede excitar una actividad
fotodinámica con una longitud de onda entre 500 y 550 nm. La luz con
esta longitud de onda sólo tiene una pequeña profundidad de
penetración en el cuerpo. Por consiguiente, los conjugados según la
invención son sumamente adecuados para la terapia de tejidos sanos
superficiales, por ejemplo neovascularizaciones y tumores
esofágicos.
La Figura 1 muestra los conjugados según la
invención AEs-Cy-HSA,
AEros-Cy-HSA y
AFI-Cy-HSA, y
la Figura 2 muestra la acumulación de
AFI-Cy-HSA en tejido tumoral.
Los siguientes ejemplos aclaran la invención.
El conjugado
AEros-Cy-HSA está representado en la
Figura 1.
Cincuenta mg de
5-[(4,6-dicloro-triazín-2-il]-amino)-fluoresceína
(= AFI-Cy) se disolvieron en 5 ml de DMSO y se
sometieron a reacción mediante adición de una cantidad
correspondiente de succinimida de yodo en 2 ml de DMSO y 1 ml de Bic
0,17M (bicarbonato sódico) para obtener AEros-Cy. La
reacción finalizó en unos segundos. El AEros-Cy
producido se añadió lentamente bajo agitación continua a 4 g de HSA
disuelta en 20 ml de solución original, 20 ml de Bic y 20 ml de
DMSO. La solución adquirió un color rojo intenso; permaneció clara.
Aproximadamente 45 minutos después, la solución proteínica se diluyó
con 1 l de agua destilada y a continuación se purificó por
ultrafiltración (YM 30, Amicon). El control analítico de pureza se
llevó a cabo mediante HPLC. Se obtuvo el conjugado según la
invención AEros-Cy-HSA.

AEs-Cy-HSA está representado en la
Figura 1.
El conjugado
AEs-Cy-HSA se preparó análogamente
al procedimiento descrito en el Ejemplo 1 utilizando succinimida de
bromo en lugar de succinimida de yodo.
El conjugado
AFI-Cy-HSA está representado en la
Figura 1.
Cuarenta y cinco mg de AFI-Cy se
disolvieron en 4 ml de DMSO y se añadieron lentamente bajo agitación
continua a 4 g de HSA disuelta en 20 ml de solución original, 20 ml
de Bic y 20 ml de DMSO. Durante la adición de
AFI-Cy, la solución adquirió un color amarillo
intenso; permaneció clara. Aproximadamente 45 minutos después, la
solución obtenida se diluyó con 1 l de agua destilada y a
continuación se purificó por ultrafiltración (YM 30, Amicon). El
control de pureza se llevó a cabo mediante HPLC. Se obtuvo
AFI-Cy-HSA.
Una rata (240 g) que presentaba un carcinoma
Walker 256 (peso del tumor 4,2% del peso corporal) recibió una
inyección intravenosa de AFI-Cy-HSA
del Ejemplo 4. Éste había sido previamente marcado radiactivamente.
La rata fue sacrificada 24 horas después. Se extrajeron todos los
órganos y el tumor. La cantidad de conjugado presente en los órganos
y en el tumor se determinó midiendo la radiactividad.
Tal como se puede observar en la Figura 2, el
conjugado según la invención
AFI-Cy-HSA se acumuló en el
tumor.
Claims (6)
1. Conjugado de un colorante de xanteno, un
engarce y albúmina sérica humana, siendo el engarce cloruro
cianúrico.
2. Conjugado según la reivindicación 1,
caracterizado porque el colorante de xanteno es fluoresceína,
amino-fluoresceína, eritrosina,
amino-eritrosina, eosina amarillenta y
amino-eosina amarillenta.
3. Conjugado según la reivindicación 1 ó 2,
caracterizado porque el colorante de xanteno presenta
fotoactividad.
4. Conjugado según una de las reivindicaciones 1
a 3, caracterizado porque presenta varios colorantes de
xanteno.
5. Procedimiento para preparar un conjugado según
una de las reivindicaciones 1 a 4, caracterizado porque el
colorante de xanteno, el cloruro cianúrico y la albúmina sérica
humana se unen de forma covalente.
6. Utilización de un conjugado según una de las
reivindicaciones 1 a 4 para la preparación de un producto para
diferenciar tejido enfermo de tejido sano.
