Abstract

Una proteasa extracelular de Acremonium chrysogenum con actividad CPC-acetilhidrolasa y su utilización para la síntesis de derivados desacetilados de cefalosporina C e inactivación del gen para aumentar la producción de cefaloporina. El procedimiento incluye la preparación de una enzima con actividad CPC- acetilhidrolasa (CPC-AH) a partir de caldos de cultivo del hongo filamentoso A.chrysogenum y la caracterización bioquímica de dicha enzima. Asimismo, describe la clonación y secuenciación de un fragmento de ADN genómico y otro de ADN complementario (ADNc) que contienen el gen cahB que codifica para dicha enzima. Mediante el uso del gen en forma de ADNc se describe la producción de la actividad enzimática CPC-AH B en Escherichia coli. Finalmente, se incluye un procedimiento para aumentar la producción de cefalosporina en A. chrysogenum mediante la inactivación del gen cahB.
