UNA RESPUESTA POSITIVA INDICA LA PRESENCIA DE EXPOSICION ANTERIOR A ENFERMEDADES PREVIAS O CONDICIONES CLINICAS COMO POR EJEMPLO ENFERMEDADES PARASITARIAS, TUBERCULOSIS, SALMONELOSIS, GONORREA, INFECCIONES CON HONGOS, INFECCIONES RIQUETSIAS, ENFERMEDAD DE LYME O ALERGENOS. LA TOTALIDAD DE LA SANGRE DEL PACIENTE ESTA INCUBADA CON EL ANTIGENO DE LA ENFERMEDAD O CONDICION PARA LA QUE SE ESTA PROBANDO AL PACIENTE. DESPUES DE UN TIEMPO APROPIADO, UN TINTE O COLORANTE FLUORESCENTE QUE TIENE UNA AFINIDAD PARA UNA CARACTERISTICA DISCRIMINANTE DE LOS LINFOCITOS O LINFOBLASTOS ACTIVADOS, COMO POR EJEMPLO: CALCIO INTRACELULAR; ANTIGENOS DE ACTIVACION DE SUPERFICIE COMO POR EJEMPLO RECEPTOR CESIONARIO; HLADR; LEU-23; Y SIMILARES. LA SANGRE INCUBADA SE INTRODUCE ENTONCES DENTRO DE UN TUBO TRANSPARENTE QUE CONTIENE UN FLOTADOR QUE CONCENTRA LAS CAPAS CONSTITUTIVAS DE LA ENVOLTURA AMORTIGUADORA DESPUES DEL CENTRIFUGADO DE LA MUESTRA DE SANGRE.
