Abstract

Die vorliegende Erfindung betrifft Verfahren zum Erhalt eines Fucoxanthin-Kristall-haltigen Präzipitats, insbesondere Fucoxanthin-Kristallen, insbesondere Fucoxanthin, und einer Fettsäurehaltigen Lipidfraktion, insbesondere Fettsäuren, aus Algenbiomasse, insbesondere Verfahren zur integrierten Extraktion und Separation von Fucoxanthin und Fettsäuren aus Algenbiomasse, sowie die aus diesen Verfahren erhaltenen Produkte, insbesondere Fucoxanthin-Kristalle und Fettsäuren.

BESCHREIBUNG
Verfahren zum Erhalt von Fucoxanthin und Fettsäuren aus Algenbiomasse
Die vorliegende Erfindung betrifft Verfahren zum Erhalt eines Fucoxanthin-Kristall-haltigen Präzipitats, insbesondere Fucoxanthin-Kristallen, insbesondere Fucoxanthin, und einer Fettsäure haltigen Lipidfraktion, insbesondere Fettsäuren, aus Algenbiomasse, insbesondere Verfahren zur integrierten Extraktion und Separation von Fucoxanthin und Fettsäuren aus Algenbiomasse, sowie die aus diesen Verfahren erhaltenen Produkte, insbesondere Fucoxanthin-Kristalle und Fettsäuren. Verschiedene Makroalgen sowie Mikroalgen, insbesondere Diatomeen (auch bekannt als Bacillariophyta oder Kieselalgen) und Vertreter der Klasse Prymnesiophyceae enthalten industriell relevante Mengen des Carotinoids Fucoxanthin (FX) sowie verschiedene meist mehrfach ungesättigte Fettsäuren (zum Beispiel Eicosapentaensäure EPA oder Docosahexaensäure DHA). Eine chemische Synthese von Fucoxanthin ist möglich, jedoch sehr aufwändig (Kajikawa et al., 2012). EPA und DHA wird für gewöhnlich aus Fischöl hergestellt (Lee et al., 2009).
Bei Fucoxanthin handelt es sich um ein Lichtsammelpigment, welches in Form von Fucoxanthin-Chlorophyll-a/c-Proteinkomplexen (FCP) in den Membranzwischenräumen der Thylakoidmembranen der Algenzellen vorkommt, wo es Lichtenergie im grünen bis blauen Wellenlängenbereich an die intrinsischen Primärpigmente in den Photosystemen der Zellen weiterleitet. Neben seiner färbenden Eigenschaft als orange-rötliches Pigment ist Fucoxanthin aus kommerzieller Sicht auch aufgrund seiner gesundheitsfördernden Eigenschaften interessant, welche im Wesentlichen auf der Struktur und den funktionalen Gruppen des Moleküls (unter anderem der Epoxylgruppe und kumulierte Kohlenstoffdoppelbindungen) basieren (Peng et al., 2011). Es wurden bereits eine Vielzahl von Studien durchgeführt, welche unter anderem die anti oxidative (Airanthi et al., 2011), anti-inflammatorische (Heo et al., 2012), krebshemmende (Hosokawa et al., 1999) und gewichtsreduzierende Wirkung (Maeda et al., 2005) von Fucoxanthin dokumentieren. Aufgrund dieser Eigenschaften ist es nicht nur aus ernähmngsphysiologischer Sicht interessant, sondern auch in Kosmetikprodukten wie zum Beispiel Sonnencremes (Heo and Jeon, 2009) vorteilhaft einsetzbar.
Die in den Algen enthaltenen (größtenteils einfach- und mehrfach ungesättigten) Fettsäuren liegen je nach Algenspezies entweder in Form von Glyco- und Phospholipiden als Bestandteil der Chloroplastenmembran oder als Speicherlipide in Form von Triacylgylceriden (TAG) vor. Wirtschaftlich am interessantesten sind essentielle Fettsäuren wie EPA und DHA. Sie besitzen ebenfalls verschiedene gesundheitsfördernde Eigenschaften.
Fettsäuren, wie EPA oder DHA, können sowohl mit polaren als auch mit unpolaren Lösungsmitteln klassisch oder mit druckbasierten Verfahren gewonnen werden (Pieber et al., 2012). Polare Lösungsmittel (Ethanol, Ethylacetat, Aceton) werden ebenfalls genutzt, um
Fucoxanthin zusammen mit weiteren lipophilen Komponenten wie Fettsäuren bei Raumtemperatur oder mittels druckbasierten Verfahren bei höheren Temperaturen zu extrahieren.
Die klassischerweise eingesetzten polaren Lösungsmittel liefern zwar hohe Ausbeuten von Fucoxanthin, sind jedoch sehr unselektiv. Daher enthalten die gewonnenen Extrakte sehr hohe Anteile von anderen Lipiden (unter anderem Fettsäuren, Phytosterole und andere Carotinoide) und müssen aufwändig aufgereinigt werden, um hochreines Fucoxanthin zu gewinnen.
Im Fall von Mikro- und Makroalgen entsteht dabei ein Extrakt, der Fucoxanthin, Fettsäuren und viele weitere Lipide enthält. Die weiteren Lipide können dabei im Hinblick auf die Trockenmasse einen Hauptteil des Extraktes darstellen.
Bisher kennt die Fachwelt insbesondere folgende Möglichkeiten die Fettsäuren (zum Beispiel EPA) und das Fucoxanthin in der extrahierten Fraktion voneinander zu separieren und so die Komponenten aufzureinigen:
Es können unpolare Lösungsmittel, welche nicht oder nur bedingt mit den zuvor verwendeten Lösungsmitteln mischbar sind (zum Beispiel Hexan in Kombination mit Ethanol und Wasser), genutzt werden, um die Fettsäuren und das Fucoxanthin in den zuvor gewonnenen Extrakten mittels Flüssig/Flüssig-Extraktion im Zwei-Phasensystem (Kim et al., 2012a, 2012b) zu trennen. Hierfür sind große Mengen des zusätzlichen Lösungsmittels notwendig und die Trennung der Komponenten ist meist nicht vollständig möglich, wodurch keine benötigte hohe Reinheit der beiden Komponenten bezogen auf die Gesamttrockenmasse der einzelnen Fraktionen erreicht wird.
Beispielsweise wird in einem Verfahren nach Kim et al. 2012a n-Hexan verwendet, um Lipide/Fettsäuren aus dem Ethanol extrakt, welcher beide Stoffklassen (Fettsäuren und Fucoxanthin) enthält, zu entfernen. Es wurde festgestellt, dass Fucoxanthin in n-Hexan nahezu unlöslich ist und n-Hexan das Fucoxanthin damit deutlich schlechter aufnehmen kann als die zuvor genannten polaren Lösungsmittel. Insgesamt dauerte mit 3 Stunden die Extraktion zudem sehr lange.
Durch die Zugabe von Phosphatsalzen in so genannten ATPS-Systemen kann das Fucoxanthin durch„Verdrängung“ zum Ausfallen gebracht werden (Gömez-Loredo et al., 2014). Hier besteht das Problem, dass die große Menge an Phosphatsalz wieder aus dem System entfernt werden muss, zum Beispiel mittels Diafiltration, um beide Stoffe nutzen zu können, was wiederum aufwändig ist und einen weiteren Prozessschritt zur Folge hat.
Es gibt ebenfalls Untersuchungen zur chromatographischen Trennung der Komponenten mit Silica-Gel-Säulen (Xia et al., 2013). Dabei wird zwar eine hohe Reinheit für das Fucoxanthin erreicht, das Verfahren ist jedoch teuer und sehr zeitaufwändig.
In der WO 2015/101410 Al wird ein Verfahren zur kontinuierlichen Extraktion von Fucoxanthin offenbart, wobei eine Flüssig-/Fest-Extraktion des Fucoxanthins mit mindestens einem Alkan erfolgt und das Fucoxanthin durch Kristallisation aus dem Extrakt isoliert wird. In diesem Verfahren werden insbesondere chromatographische Säulen oder eine gepackte Säule mit Festbett zur Isolation des Fucoxanthins eingesetzt. Nachteilhaft an dem Verfahren ist es, dass keine hohe Reinheit des Fucoxanthin erreicht wird und dass insbesondere der Einsatz von chromatographischen Säulen oder eines gepackten Festbetts einen hohen Aufwand, insbesondere einen hohen zeitlichen Aufwand, darstellt. Ferner ist das in WO 2015/101410 offenbarte Verfahren weniger wirtschaftlich, da die Fettsäure enthaltende Fraktion aus dem Algenextrakt nicht weiter verarbeitet wird.
Der vorliegenden Erfindung liegt das technische Problem zugrunde, sowohl Fucoxanthin als auch Fettsäuren, insbesondere EPA und/oder DHA, in hoher Ausbeute, in besonders hoher Reinheit und ohne Produktschädigung zu gewinnen und dabei ein besonders kostengünstiges und wirtschaftlich attraktives Verfahren bereitzustellen. Die vorliegende Erfindung löst das ihr zugrunde liegende technische Problem durch die Bereitstellung der Lehren der unabhängigen Ansprüche.
Erfmdungsgemäß bereitgestellt wird ein Verfahren zum Erhalt eines Fucoxanthin-haltigen Präzipitats und einer Fettsäure-haltigen Lipidfraktion aus Algenbiomasse, umfassend die folgenden Verfahrensschritte: a) Bereitstellen von mindestens einem apolaren organischen Lösungsmittel und
Algenbiomasse, b) Extraktion von Fucoxanthin und Fettsäuren mittels des in Verfahrensschritt a)
bereitgestellten mindestens einen apolaren organischen Lösungsmittels aus der in
Verfahrensschritt a) bereitgestellten Algenbiomasse in mindestens einer
Extraktionsreaktoreinheit, der mindestens ein erstes Filterelement zugeordnet ist, bei einer Temperatur von 60 bis 150 °C und einem Druck von 1,5 bis 100 bar zum Erhalt eines Gemisches umfassend einen Algenextrakt und eine extraktionsunterzogene Algenbiomasse, c) Abtrennen der extraktionsunterzogenen Algenbiomasse aus dem in Verfahrensschritt b) erhaltenen Gemisch mittels Filtrierung durch das mindestens eine erste Filterelement zum
Erhalt eines filtrierten Algenextraktes und Einleiten des erhaltenen filtrierten Algenextraktes in mindestens eine Präzipitationsreaktoreinheit, der mindestens ein zweites Filterelement zugeordnet ist, d) Absenken der Temperatur des filtrierten Algenextrakts auf -80 bis 50 °C in der
Präzipitationsreaktoreinheit zur Präzipitation von Fucoxanthin-Kristallen zum Erhalt eines
Gemisches aus Fucoxanthin-Kristall-haltigem Präzipitat und Fettsäure-haltiger
Algenextraktfraktion, e) Abtrennen des Fucoxanthin-Kristall-haltigen Präzipitats aus dem Gemisch aus Fucoxanthin- Kristall-haltigem Präzipitat und Fettsäure-haltiger Algenextraktfraktion mittels Filtrierung durch das mindestens eine zweite Filterelement zum Erhalt eines Fucoxanthin-Kristall- haltigen Präzipitats und einer Fettsäure-haltigen Algenextraktfraktion, und f) Abtrennen einer Fettsäure-haltigen Lipidfraktion aus der Fettsäure-haltigen
Algenextraktfraktion durch Verdampfen des mindestens einen apolaren organischen
Lösungsmittels zum Erhalt der Fettsäure-haltigen Lipidfraktion. Die vorliegende Erfindung stellt daher ein Verfahren zum , insbesondere diskontinuierlichen, Erhalt eines Fucoxanthin-Kristall-haltigen Präzipitats, insbesondere von Fucoxanthin, und einer Fettsäure-haltigen Lipidfraktion aus Algenbiomasse bereit, wobei in einem ersten
Verfahrensschritt a) mindestens ein apolares organisches Lösungsmittel und Algenbiomasse bereitgestellt wird und in einem zweiten Verfahrensschritt b) Fucoxanthin und Fettsäuren mittels des im ersten Verfahrensschritt bereitgestellten mindestens einen apolaren organischen Lösungsmittels aus der im ersten Verfahrensschritt bereitgestellten Algenbiomasse extrahiert werden, insbesondere bei einer Temperatur von 60 bis 150 °C und einem Druck von 1,5 bis 100 bar zum Erhalt eines Gemisches umfassend einen Algenextrakt und eine extraktionsunterzogene Algenbiomasse.
In einem dritten Verfahrensschritt c) wird die extraktionsunterzogene Algenbiomasse aus dem im zweiten Verfahrensschritt erhaltenen Gemisch mittels Filtrierung zum Erhalt eines filtrierten Algenextrakts abgetrennt. In einem vierten Verfahrensschritt d) wird die Temperatur des filtrierten Algenextrakts auf -80 bis 50 °C zur Präzipitation von Fucoxanthin-Kristallen und somit zum Erhalt eines Gemisches aus Fucoxanthin-Kristall-haltigem Präzipitat und Fettsäure haltiger Algenextraktfraktion abgesenkt.
In einem fünften Verfahrensschritt e) wird das Fucoxanthin-Kristall-haltige Präzipitat aus dem Gemisch aus Fucoxanthin-Kristall-haltigem Präzipitat und Fettsäure-haltiger
Algenextraktfraktion mittels Filtrierung zum Erhalt eines Fucoxanthin-Kristall-haltigen
Präzipitats und einer Fettsäure-haltigen Algenextraktfraktion abgetrennt. In einem sechsten Verfahrensschritt f) wird eine Fettsäure-haltige Lipidfraktion aus der Fettsäure-haltigen
Algenextraktfraktion durch Verdampfen des mindestens einen apolaren organischen
Lösungsmittels zum Erhalt der Fettsäure-haltigen Lipidfraktion abgetrennt. Die vorliegende Erfindung sieht insbesondere vor, die vorgenannten sechs Verfahrensschritte a) bis f) in einem Reaktorsystem durchzuführen, welches mindestens eine Extraktionsreaktoreinheit und mindestens eine der Extraktionsreaktoreinheit nachgeschaltete Präzipitationsreaktoreinheit aufweist und wobei jeder Extraktionsreaktoreinheit mindestens ein erstes Filterelement und jeder Präzipitationsreaktoreinheit mindestens ein zweites Filterelement zugeordnet sind und wobei das mindestens eine erste Filterelement - in Prozessrichtung vom ersten zum letzten Schritt (Verfahrensschritt a) zum sechsten Schritt, Verfahrensschritt f)) gesehen - vor der mindestens einen Präzipitationsreaktoreinheit angeordnet ist und das mindestens eine zweite Filterelement nach der mindestens einen Präzipitationsreaktoreinheit angeordnet ist. Die vorliegende erfindungsgemäße, insbesondere diskontinuierliche, Verfahrensweise zeichnet sich demgemäß insbesondere dadurch aus, ein Fucoxanthin-haltiges Präzipitat, insbesondere Fucoxanthin, und eine Fettsäure-haltige Lipidfraktion aus Algenbiomasse mittels mindestens eines apolaren organischen Lösungsmittels zu extrahieren und durch die erfmdungsgemäßen Reaktionsparameter und die, insbesondere diskontinuierliche, Verfahrensweise unter Nutzung mindestens zweier Filterelemente, nämlich zur Abtrennung einer in einem zweiten Verfahrensschritt erhaltenen extraktionsunterzogenen Algenbiomasse und eines in einem vierten Verfahrensschritt erhaltenen Fucoxanthin-Kristall-haltigen Präzipitats, hohe Ausbeuten und Reinheiten an Fucoxanthin und Fettsäuren erhalten zu können.
Das Verfahren kann kontinuierlich oder diskontinuierlich durchgeführt werden.
Die Extraktion von Fucoxanthin und Fettsäuren aus der Algenbiomasse erfolgt erfmdungsgemäß bei einer Temperatur von 60 bis 100 °C und einem Druck von 1,5 bis 100 bar in mindestens einer Extraktionsreaktoreinheit, welcher mindestens ein erstes Filterelement zugeordnet ist. Erhalten wird durch die Extraktion ein Gemisch umfassend einen Algenextrakt sowie eine extraktionsunterzogene Algenbiomasse. Die extraktionsunterzogene Algenbiomasse wird durch Filtration mittels des mindestens einen ersten Filterelements aus dem Gemisch entfernt. Es wird ein filtrierter Algenextrakt erhalten, welcher in mindestens eine Präzipitationsreaktoreinheit eingeleitet wird, der mindestens ein zweites Filterelement zugeordnet ist. In dieser mindestens einen Präzipitationsreaktoreinheit wird die Temperatur des filtrierten Algenextraktes abgesenkt auf -80 bis 50 °C, um Fucoxanthin-Kristalle aus dem filtrierten Algenextrakt zu präzipitieren und demgemäß ein Gemisch aus Fucoxanthin-Kristall-haltigem Präzipitat und einer nicht präzipitierten Fettsäure-haltige Algenextraktfraktion zu erhalten. Das Fucoxanthin-Kristall- haltige Präzipitat wird aus dem zuvor gebildeten Gemisch aus Fucoxanthin-Kristall-haltigem Präzipitat und einer Fettsäure-haltigen Algenextraktfraktion durch Filtration mittels des mindestens einen zweiten Filterelements abgetrennt, wobei ein Fucoxanthin-Kristall-haltiges Präzipitat sowie eine Fettsäure-haltige Algenextraktfraktion erhalten werden. Aus der Fettsäure haltigen Algenextraktfraktion wird in einem weiteren Verfahrensschritt durch Verdampfen des mindestens einen apolaren organischen Lösungsmittels eine Fettsäure-haltige Lipidfraktion erhalten. Das Fucoxanthin-Kristall-haltige Präzipitat kann in mindestens einem weiteren Verfahrensschritt, zum Beispiel Auflösen, Trocknen, Umkristallisieren, Zerkleinern, insbesondere mit Ultraschallbehandlung, oder dergleichen, weiterverarbeitet werden, um Fucoxanthin-Kristalle, insbesondere Fucoxanthin, insbesondere mit einem definierten Durchmesser, zu erhalten.
Das erfindungsgemäße Verfahren stellt vorteilhafterweise Fucoxanthin-Kristalle, insbesondere Fucoxanthin, sowie Fettsäuren, insbesondere DHA und EPA, in besonders hoher Reinheit sowie besonders hoher Ausbeute her. Insbesondere die besonders hohe Ausbeute der Fettsäuren ist mit Blick darauf, dass die Fettsäuren in den eingesetzten Algen in Form von Glycolipiden Vorkommen, welche einen eher polaren Charakter aufweisen, nicht ohne weiteres vorhersehbar. Ohne an die Theorie gebunden zu sein, kann es sein, dass thermisch bedingte Umwandlungen der Glycolipide zu freien Fettsäuren während der Extraktion zu dieser besonders hohen Ausbeute führen.
Das erfindungsgemäße Verfahren stellt die Fettsäuren und auch das Fucoxanthin in besonders hoher Reinheit her, wobei vorteilhafterweise beide Komponenten in einem einzigen Verfahrensgang gewonnen werden können und somit das Verfahren auch besonders wirtschaftlich und kostengünstig ist. Insbesondere können weitere Aufreinigungsschritte entfallen. Ein besonderer Vorteil der hohen Reinheit der Fettsäuren sowie der Fucoxanthin- Kristalle liegt in deren besserer Weiterverarbeitung. Zum Beispiel weisen die Fettsäuren, gewonnen nach dem erfindungsgemäßen Verfahren, geringe oder keine Verfärbungen auf, weswegen sie insbesondere in der Kosmetikindustrie vorteilhafterweise verwendet werden können.
In einer besonders bevorzugten Ausführungsform wird das erfindungsgemäße Verfahren in einem erfmdungsgemäß für die Durchführung der Erfindung eingerichteten Reaktorsystem durchgeführt. Ein erfmdungsgemäß eingerichtetes Reaktorsystem umfasst mindestens eine Extraktionsreaktoreinheit, welcher mindestens ein erstes Filterelement zugeordnet ist, und mindestens eine zweite Präzipitationsreaktoreinheit, welcher mindestens ein zweites Filterelement zugeordnet ist. Vorzugsweise weist das Reaktorsystem eine dem zweiten Filterelement nachgeordnete und mit der Präzipitationsreaktoreinheit verbundene Verdampfungsvorrichtung, insbesondere Destillationsapparat auf.
In einer besonders bevorzugten Ausführungsform sind einer Extraktionsreaktoreinheit mindestens zwei, insbesondere mindestens drei, insbesondere zwei, insbesondere drei, erste Filterelemente zugeordnet. In einer besonders bevorzugten Ausführungsform sind einer Präzipitationsreaktoreinheit mindestens zwei, insbesondere mindestens drei, insbesondere zwei, insbesondere drei, zweite Filterelemente zugeordnet.
In einer besonders bevorzugten Ausführungsform sind die mindestens eine
Extraktionsreaktoreinheit und die mindestens eine Präzipitationsreaktoreinheit zwei räumlich voneinander getrennte, durch eine Verbindung, insbesondere Fluidverbindung, verbundene Reaktoren.
In einer besonders bevorzugten Ausführungsform liegen die mindestens eine
Extraktionsreaktoreinheit und die mindestens eine Präzipitationsreaktoreinheit in einem Reaktor integriert vor und stellen zwei getrennte durch eine Verbindung, insbesondere Fluidverbindung, verbundene Einheiten des Reaktors dar.
In einer besonders bevorzugten Ausführungsform ist die Verbindung zwischen beiden
Reaktoreinheiten mindestens ein Schlauch oder mindestens ein Rohr.
In einer besonders bevorzugten Ausführungsform ist die Verbindung zwischen beiden
Reaktoreinheiten eine Öffnung in einer räumlichen Trennvorrichtung. Die räumliche Trennvorrichtung kann eine Wand sein.
In einer besonders bevorzugten Ausführungsform ist in mindestens einer der Reaktoreinheiten ein Rührwerk enthalten.
In einer besonders bevorzugten Ausführungsform ist in der mindestens einen
Extraktionsreaktoreinheit ein Rührwerk enthalten.
In einer besonders bevorzugten Ausführungsform ist in der mindestens einen
Präzipitationsreaktoreinheit ein Rührwerk enthalten.
In einer besonders bevorzugten Ausführungsform ist in der mindestens einen
Extraktionsreaktoreinheit und der mindestens einen Präzipitationseinheit jeweils ein Rührwerk enthalten.

In einer besonders bevorzugten Ausführungsform weist das erfindungsgemäße Verfahren Ausbeuten von 65 bis 95 Gew.-% (TS, Trockensubstanz) (Ausbeute bezogen auf Trockengewicht der Fettsäuren im Ausgangsmaterial) für Fettsäuren auf.
In einer besonders bevorzugten Ausführungsform weist das erfindungsgemäße Verfahren Ausbeuten von 65 bis 98 Gew.-% (TS) (Ausbeute bezogen auf Trockengewicht des EPA im Ausgangsmaterial) für EPA auf.
In einer besonders bevorzugten Ausführungsform weist das erfindungsgemäße Verfahren Ausbeuten von 30 bis 75 Gew.-%, insbesondere 40 bis 75 Gew.-% (TS) (Ausbeute bezogen auf Trockengewicht des Fucoxanthins im Ausgangsmaterial) für Fucoxanthin auf. In einer besonders bevorzugten Ausführungsform des erfmdungsgemäßen Verfahrens ist vorgesehen, dass die in Verfahrensschritt a) bereitgestellte Algenbiomasse vor der Bereitstellung zerkleinert, insbesondere durch Zellaufschluss, wurde.
In einer besonders bevorzugten Ausführungsform des erfmdungsgemäßen Verfahrens ist vorgesehen, dass die in Verfahrensschritt a) bereitgestellte Algenbiomasse vor der Bereitstellung getrocknet, insbesondere mittels Sprüh- oder Gefriertrocknung, wurde.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass die in Verfahrensschritt a) bereitgestellte Algenbiomasse vor der Bereitstellung zerkleinert, insbesondere durch Zellaufschluss, und getrocknet, insbesondere mittels Sprüh- oder Gefriertrocknung, wurde.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist die in Verfahrensschritt a) bereitgestellte Algenbiomasse eine partikuläre Algenbiomasse.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Zerkleinerung, insbesondere der Zellaufschluss, der Algenbiomasse vor dem Bereitstellen in Verfahrensschritt a) durch eine Kugelmühle, insbesondere eine Rührwerkskugelmühle, oder durch einen Hochdruckhomogenisator, insbesondere durch eine Kugelmühle, insbesondere eine Rührwerkskugelmühle, durchgeführt wird.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass das mindestens eine apolare organische Lösungsmittel ein Alkan, insbesondere ein n-Alkan ist oder eine Kombination aus verschiedenen Alkanen, insbesondere verschiedenen n-Alkanen ist.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass das mindestens eine apolare organische Lösungsmittel Pentan, Hexan, Heptan und/oder eine Kombination dieser ist, insbesondere n-Hexan, n-Pentan, n-Heptan und/oder eine Kombination dieser, insbesondere Cyclopentan, Cyclohexan, Cycloheptan und/oder eine Kombination dieser ist.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass das mindestens eine apolare organische Lösungsmittel Hexan oder Heptan ist, insbesondere Hexan, insbesondere Heptan ist. In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass das mindestens eine apolare organische Lösungsmittel n-Hexan oder n-Heptan ist, insbesondere n-Hexan, insbesondere n-Heptan ist.
In einer besonders bevorzugten Ausführungsform wird das mindestens eine apolare organische Lösungsmittel vor der Bereitstellung in Verfahrensschritt a) mit geringen Mengen mindestens eines polaren Lösungsmittels, insbesondere 0,1 bis 10 Vol.-%, insbesondere 0,1 bis 5 Vol.-%, insbesondere 0,1 bis 1 Vol.-%, insbesondere 0,1 bis 0,5 Vol.-% (jeweils bezogen auf das Gesamtvolumen der Mischung aus dem mindestens einen apolaren organischen Lösungsmittel und dem mindestens einen polaren Lösungsmittel), gemischt.
In einer besonders bevorzugten Ausführungsform ist das mindestens eine polare Lösungsmittel in der Mischung aus dem mindestens einen apolaren organische Lösungsmittel und dem mindestens einen polaren Lösungsmittel ein Lösungsmittel ausgewählt aus der Gruppe bestehend aus Ethanol, Ethylacetat, Aceton oder Mischungen davon.
Das Mischen des mindestens einen apolaren organischen Lösungsmittels vor der Bereitstellung in Verfahrensschritt a) mit geringen Mengen mindestens eines polaren Lösungsmittels führt insbesondere in vorteilhafter Weise zu einer Steigerung der Ausbeuten an Fucoxanthin und/oder Fettsäuren.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass die Algenbiomasse eine Algenbiomasse von Makroalgen oder Mikroalgen ist, insbesondere Diatomeen, insbesondere Bacillariophyceae, insbesondere Algen der Klasse Prymnesiophyceae, insbesondere Phaeodactylum tricornutum , insbesondere Cylindrotheca fusiformis , insbesondere Odontella aurita , insbesondere Haptophyta, insbesondere Isochrysis spec., insbesondere Isochrysis aff galbana.
In einer ganz besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass die Algenbiomasse eine Algenbiomasse von Phaeodactylum tricornutum ist.
In einer besonders bevorzugten Ausführungsform ist das Verfahren zum Erhalt eines Fucoxanthin-haltigen Präzipitats und einer Fettsäure-haltigen Lipidfraktion aus Algenbiomasse ein Verfahren zum diskontinuierlichen Erhalt eines Fucoxanthin-haltigen Präzipitats und einer Fettsäure-haltigen Lipidfraktion aus Algenbiomasse. In einer besonders bevorzugten Ausführungsform sieht das erfindungsgemäße Verfahren vor, dass die Temperatur im Verfahrensschritt b) 100 bis 120 °C, insbesondere genau 100 °C, beträgt.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung beträgt die Temperatur im Verfahrensschritt b) 70 bis 120 °C, insbesondere 70 bis 100 °C, insbesondere 70 bis 95 °C, insbesondere 70 bis 90 °C, insbesondere 70 bis 80 °C, insbesondere genau 70 °C.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung beträgt die Temperatur im Verfahrensschritt b) 80 bis 150 °C, insbesondere 80 bis 120 °C, insbesondere 80 bis 100 °C, 85 bis 150 °C, insbesondere 85 bis 120 °C, insbesondere 85 bis 100 °C, insbesondere 90 bis 150 °C, insbesondere 90 bis 100 °C, insbesondere genau 80 °C, insbesondere genau 90 °C, insbesondere genau 95 °C.

In einer ganz besonders bevorzugten Ausführungsform ist vorgesehen, dass der Druck in Verfahrensschritt b) 100 bar beträgt.
Der in Verfahrensschritt b) eingesetzte Druck kann insbesondere synergistisch eingesetzt werden, um die Extraktion gemäß Verfahrensschritt b) zu bewirken und um gemäß einer besonderen Ausführungsform mit Hilfe des Drucks über das erste Filterelement einen filtrierten Algenextrakt zu erhalten und über das zweite Filterelement ein Fucoxanthin-haltiges Präzipitat abzutrennen sowie eine Fettsäure-haltige Algenextraktfraktion zu erhalten.
In einer besonders bevorzugten Ausführungsform kann mit Hilfe des in Verfahrensschritt b) eingesetzten Drucks über das erste Filterelement ein filtrierter Algenextrakt erhalten und/oder über das zweite Filterelement ein Fucoxanthin-haltiges Präzipitat abgetrennt sowie eine Fettsäure-haltige Algenextraktfraktion erhalten werden.
In einer besonders bevorzugten Ausführungsform kann mit Hilfe des in Verfahrensschritt b) eingesetzten Drucks über das erste Filterelement ein filtrierter Algenextrakt erhalten und über das zweite Filterelement ein Fucoxanthin-haltiges Präzipitat abgetrennt sowie eine Fettsäure haltige Algenextraktfraktion erhalten werden.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Verweilzeit in der Extraktionsreaktoreinheit in Verfahrensschritt b) 1 bis 60 Minuten, insbesondere 1 bis 45 Minuten, insbesondere 1 bis 30 Minuten, insbesondere 1 bis 25 Minuten, insbesondere 1 bis 20 Minuten, insbesondere 1 bis 15 Minuten, insbesondere 1 bis 10 Minuten, insbesondere 1 bis 5 Minuten, beträgt.
In einer ganz besonders bevorzugten Ausführungsform ist vorgesehen, dass die Verweilzeit in der Extraktionsreaktoreinheit in Verfahrensschritt b) 1 bis 5 Minuten beträgt.
Durch die besonders kurzen Verweilzeiten in Verfahrensschritt b) kann erreicht werden, dass im Vergleich zu bisher bekannten Verfahren weniger Produktschädigungen von Fettsäuren und/oder Fucoxanthin auftreten und/oder die Fettsäure-haltige Lipidfraktion und/oder das Fucoxanthin- Kristall-haltige Präzipitat eine höhere Reinheit aufweisen.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass der Verfahrensschritt b) in einer Batch-Betriebsweise durchgeführt wird.
In einer besonders bevorzugten Ausführungsform sieht das erfindungsgemäße Verfahren vor, dass Verfahrensschritt b) vor Durchführung von Verfahrensschritt c) mindestens einmal, insbesondere mindestens zweimal, insbesondere mindestens dreimal, insbesondere mindestens viermal, wiederholt wird. In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass der in Verfahrensschritt b) erhaltene Algenextrakt weitere Carotinoide, Polyphenole und/oder Phytosterole umfasst. Insbesondere ist vorgesehen, dass die zuvor genannten Substanzen, das heißt weitere Carotinoide, Polyphenole und/oder Phytosterole, in einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung mit der Fettsäure-haltigen Lipidfraktion in Verfahrensschritt f) erhalten werden, das heißt in der erhaltenen Fettsäure haltigen Lipidfraktion vorhanden sind.
In einer besonders bevorzugten Ausführungsform wird das mindestens eine apolare organische Lösungsmittel vor dem Verfahrensschritt b) in einem Verfahrensschritt bO) vortemperiert auf die in Verfahrensschritt b) benötigte Temperatur, insbesondere in der Extraktionsreaktoreinheit, vorzugsweise in einer separaten der Extraktionsreaktoreinheit vorgelagerten und mit ihr verbundenen Vorrichtung.
In einer besonders bevorzugten Ausführungsform wird das mindestens eine apolare organische Lösungsmittel vor dem Verfahrensschritt b) in einem Verfahrensschritt bO) in der Präzipitationsreaktoreinheit, welche vorzugsweise mit einer Vorrichtung zum Temperieren ausgestattet ist, vortemperiert und anschließend in die Extraktionsreaktoreinheit überführt.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung wird der Druck nach Durchführung der Extraktion in Verfahrensschritt b) in Verfahrensschritt c) gesenkt.

Der Druck, welcher in Verfahrensschritt b) zur Extraktion eingesetzt wird, kann auch eingesetzt werden, um über die Druckdifferenzen an dem mindestens einen ersten und/oder mindestens einen zweiten Filterelement die Filtrationen in den Verfahrensschritten c) und/oder e) zu bewirken, insbesondere energetisch sinnvoll zu bewirken. Insbesondere kann der Druck gemäß Verfahrensschritt b) eingesetzt werden, um über die Druckdifferenzen an dem mindestens einen ersten und mindestens einen zweiten Filterelement die Filtrationen in den Verfahrensschritten c) und e) zu bewirken, insbesondere energetisch sinnvoll zu bewirken.

In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Verweilzeit in der Präzipitationsreaktoreinheit in Verfahrensschritt d) 1 bis 60 Minuten, insbesondere 1 bis 45 Minuten, insbesondere 1 bis 30 Minuten, insbesondere 1 bis 20 Minuten, insbesondere 1 bis 15 Minuten, insbesondere 1 bis 10 Minuten, insbesondere 1 bis 5 Minuten, beträgt.
Durch die besonders kurzen Verweilzeiten in Verfahrensschritt d) kann erreicht werden, dass im Vergleich zu bisher bekannten Verfahren weniger Produktschädigungen von Fettsäuren und/oder Fucoxanthin auftreten und/oder die Fettsäure-haltige Lipidfraktion und/oder das Fucoxanthin- Kristall-haltige Präzipitat eine höhere Reinheit aufweisen.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass der Verfahrensschritt d) in einer Semi-kontinuierlichen-Betriebsweise, insbesondere in einer Gegenstrombetriebsweise, durchgeführt wird.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass der Verfahrensschritt d) in einer Batch-Betriebsweise durchgeführt wird. In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Verfahrensschritte b) und d) in einer Batch-Betriebsweise durchgeführt werden.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass Verfahrensschritt b) in einer Batch-Betriebsweise und Verfahrensschritt d) in einer Semi-kontinuierlichen- Betriebsweise, insbesondere in einer Gegenstrombetriebsweise, durchgeführt werden.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass die Abtrennung des Fucoxanthin-Kristall-haltigen Präzipitats in Verfahrensschritt e) mittels Überdruck-, Unterdrück- oder Vakuum- unterstützter Filtration erfolgt.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Filtration von Verfahrensschritt e) eine Querstromfiltration ist.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass die Filtration von Verfahrensschritt e) eine Siebfiltration ist.
In einer besonders bevorzugten Ausführungsform sieht das erfindungsgemäße Verfahren vor, dass das in Verfahrensschritt f) verdampfte mindestens eine apolare organische Lösungsmittel in einem zusätzlichen Verfahrensschritt g) verflüssigt wird und das verflüssigte mindestens eine apolare organische Lösungsmittel in Verfahrensschritt a) wieder bereitgestellt wird.
In einer besonders bevorzugten Ausführungsform sieht das erfindungsgemäße Verfahren vor, dass der Verfahrensschritt g) in einer Vorrichtung zur Rückführung des Lösungsmittels durchgeführt wird.
In einer besonders bevorzugten Ausführungsform betrifft die vorliegende Erfindung auch Fucoxanthin-Kristalle herstellbar, insbesondere hergestellt, durch ein erfindungsgemäßes Verfahren.
In einer besonders bevorzugten Ausführungsform betrifft die vorliegende Erfindung auch die Fettsäuren, insbesondere EPA und/oder DHA, herstellbar, insbesondere hergestellt, durch ein erfindungsgemäßes Verfahren. In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 40 bis 90 Gew.-%, insbesondere 45 bis 75 Gew.-%, Fettsäuren (bezogen auf Gesamtgewicht der Trockensubstanz) auf.
In einer besonders bevorzugten Ausführungsform besteht die Fettsäure-haltige Lipidfraktion im Wesentlichen aus Fettsäuren, insbesondere besteht aus diesen.
In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 10 bis 50 Gew.-%, insbesondere 20 bis 35 Gew.-%, EPA (bezogen auf Gesamtgewicht der Trockensubstanz) auf.
In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 50 bis 70 Gew.-%, insbesondere 55 bis 65 Gew.-%, EPA (bezogen auf Gesamtgewicht der Trockensubstanz an Fettsäuren in der Fettsäure-haltigen Lipidfraktion) auf.
In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 50 bis 70 Gew.-%, insbesondere 55 bis 65 Gew.-%, DHA (bezogen auf Gesamtgewicht der Trockensubstanz an Fettsäuren in der Fettsäure-haltigen Lipidfraktion) auf.
In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion mehr EPA als DHA auf, insbesondere eine 1,2-fache, insbesondere 1,5-fache, insbesondere 2-fache, insbesondere 5-fache, insbesondere 10-fache, Menge von EPA im Vergleich zu DHA (bezogen auf das Gesamtgewicht an EPA und DHA) auf. Dies ist insbesondere von Vorteil, wenn höhere Anteile an EPA benötigt werden. Aus der bisher verwendeten Quelle für EPA und DHA Fischöl, müssen hierzu beide aufwendig mittels chromatographischer Verfahren voneinander getrennt werden.
In einer besonders bevorzugten Ausführungsform weist die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion eine ölige Konsistenz auf.
In einer besonders bevorzugten Ausführungsform weist das in Verfahrensschritt e) erhaltene Fucoxanthin-Kristall-haltige Präzipitat 50 bis 100 Gew.-%, insbesondere 60 bis 99,999 Gew.-%, Fucoxanthin, insbesondere Fucoxanthin-Kristalle (bezogen auf Gesamtgewicht der Trockensubstanz) auf. Es ist insbesondere in erfindungsgemäß bevorzugter Ausführungsform vorgesehen, dass durch die Variation der Dauer der Verweilzeit, der Geschwindigkeit der Abkühlung oder von beiden Parametern in Verfahrensschritt d) die Reinheit der Fucoxanthin-Kristalle, das heißt der Anteil an Fucoxanthin-Kristallen in dem Fucoxanthin-Kristall-haltigen Präzipitat, beeinflusst wird.
In einer weiteren bevorzugten Ausführungsform kann vorgesehen sein, dass durch die Variation der Dauer der Verweilzeit, durch die Geschwindigkeit der Abkühlung oder durch beide Parameter in Verfahrensschritt d), den Durchmesser der erhaltenen Fucoxanthin-Kristalle in dem Fucoxanthin-Kristall-haltigen Präzipitat zu beeinflussen.
In einer Ausführungsform der vorliegenden Erfindung liegen die in dem Fucoxanthin-Kristall- haltigen Präzipitat enthaltenen Fucoxanthin-Kristalle in Form von Agglomeraten, zum Beispiel einer Größe von 10 bis 100 pm, insbesondere 10 bis 80 pm, insbesondere 10 bis 70 pm, insbesondere 10 bis 50 pm, insbesondere 10 bis 25 pm, insbesondere 20 bis 100 pm, insbesondere 20 bis 80 pm, insbesondere 20 bis 70 pm, insbesondere 20 bis 50 pm, insbesondere 20 bis 25 pm, insbesondere 30 bis 80 pm, insbesondere 30 bis 70 pm, insbesondere 30 bis 50 pm, vor. In besonders bevorzugten Ausführungsformen lassen sich diese Agglomerate durch Ultraschall in einzelne Kristalle überführen.
In einer besonders bevorzugten Ausführungsform werden die Fucoxanthin-Kristalle aus dem Fucoxanthin-Kristall-haltigen Präzipitat erhalten durch Zerkleinerung des Fucoxanthin-Kristall- haltigen Präzipitats.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass das Fucoxanthin-Kristall- haltige Präzipitat mittels Ultraschall zum Erhalt von Fucoxanthin-Kristallen zerkleinert wird.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass das Fucoxanthin-Kristall- haltige Präzipitat mittels mechanischer Scherkräfte zum Erhalt von Fucoxanthin-Kristallen zerkleinert wird.
In einer besonders bevorzugten Ausführungsform ist vorgesehen, dass das Fucoxanthin-Kristall- haltige Präzipitat zum Erhalt von Fucoxanthin-Kristallen umkristallisiert wird.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung ist vorgesehen, dass die in Verfahrensschritt e) im Fucoxanthin-Kristall-haltige Präzipitat enthaltenen Fucoxanthin-Kristalle einen Durchmesser von 0,1 bis 10 pm aufweisen, insbesondere von 0,5 bis 10 mih, insbesondere von 1 bis 10 mih, insbesondere von 2 bis 10 mih, insbesondere von 3 bis 10 mih, insbesondere von 4 bis 10 mih, insbesondere von 5 bis 10 mih, insbesondere von 6 bis 10 mih, insbesondere von 8 bis 10 mih, insbesondere von 0,1 bis 8 mih, insbesondere von 0,1 bis 6 mih, insbesondere von 0,1 bis 4 mih, insbesondere von 0,1 bis 2 mih, insbesondere von 0,1 bis 1 mih, insbesondere von 0,5 bis 8 mih, insbesondere von 0,5 bis 6 mih, insbesondere von 0,5 bis 4 mih, insbesondere von 0,5 bis 2 mih, insbesondere von 0,5 bis 1 mih, insbesondere von 1 bis 9 mih, insbesondere von 1 bis 8 mih, insbesondere von 1 bis 6 mih, insbesondere von 1 bis 5 mih, insbesondere von 1 bis 4 mih, insbesondere von 1 bis 2 mih, insbesondere von 3 bis 8 mih, insbesondere von 3 bis 6 mih, insbesondere von 3 bis 5 mih, insbesondere von 3 bis 4 mih aufweisen.
Im Zusammenhang mit der vorliegenden Erfindung wird unter„Algenbiomasse“ ein Material biologischen Ursprungs verstanden, welches Algen, insbesondere Makroalgen, insbesondere Mikroalgen, umfasst, insbesondere im Wesentlichen aus diesen besteht, insbesondere aus diesen besteht.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einer„extraktionsunterzogenen Algenbiomasse“ eine Algenbiomasse verstanden, aus welcher mittels Extraktion Stoffe entfernt worden sind. Insbesondere werden darunter die Materialien verstanden, welche sich nicht im eingesetzten Lösungsmittel lösen.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem „Algenextrakt“ eine Zusammensetzung aus mindestens einem apolaren Lösungsmittel und in dem Lösungsmittel gelösten Stoffen aus Algenbiomasse verstanden.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem„filtrierten Algenextrakt“ ein Algenextrakt verstanden, welcher einer Abtrennung, insbesondere einer Filtration, unterzogen wurde.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem„Fucoxanthin-Kristall- haltigen Präzipitat“ ein aus einem filtrierten Algenextrakt ausgefallenes, das heißt präzipitiertes, Material verstanden, welches Fucoxanthin-Kristalle umfasst, insbesondere im Wesentlichen aus diesen besteht, insbesondere aus diesen besteht. Insbesondere kann das Fucoxanthin-Kristall- haltige Präzipitat Agglomerate von Fucoxanthin-Kristallen umfassen, insbesondere im Wesentlichen aus diesen bestehen, insbesondere aus diesen bestehen. Im Zusammenhang mit der vorliegenden Erfindung wird unter einer „Fettsäure-haltigen Algenextraktfraktion“ eine Algenextraktfraktion verstanden, welche Fettsäuren umfasst, insbesondere im Wesentlichen aus diesen besteht, insbesondere aus diesen besteht und welche größtenteils, insbesondere ganz, von ausgefallenen Fucoxanthin-Kristallen abgetrennt wurde.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einer „Fettsäure-haltigen Lipidfraktion“ eine Fraktion verstanden, welche Lipide, unter anderem Fettsäuren, umfasst, insbesondere im Wesentlichen aus diesen besteht, insbesondere aus diesen besteht. Insbesondere kann die Fettsäure-haltige Lipidfraktion Fettsäuren umfassen, insbesondere im Wesentlichen aus diesen bestehen, insbesondere aus diesen bestehen. Diese kann auch als Algen-Öl bezeichnet werden.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem„Filterelement“ eine Vorrichtung verstanden, welche zum Filtrieren geeignet ist. Diese kann in jedweder Bauart, Form, Größe oder Typ ausgestaltet sein, solange sie zum Filtrieren geeignet ist.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem„ersten Filterelement“ ein Filterelement verstanden, das in der Lage ist eine extraktionsunterzogene Algenbiomasse von einem Algenextrakt, gewonnen durch Extraktion von Fucoxanthin und Fettsäuren mittels mindestens einen apolaren organischen Lösungsmittels, zu trennen, wobei der Algenextrakt, als filtrierter Algenextrakt, das Permeat und die extraktionsunterzogene Algenbiomasse das Retentat darstellt.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem„zweiten Filterelement“ ein Filterelement verstanden, das in der Lage ist ein Fucoxanthin-Kristall-haltiges Präzipitat von einer Fettsäure-haltigen Algenextraktfraktion zu trennen, wobei das Fucoxanthin-Kristall-haltige Präzipitat das Retentat und die Fettsäure-haltige Algenextraktfraktion das Permeat darstellt.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einer„Extraktionsreaktoreinheit“ eine Vorrichtung verstanden, die als Reaktionsraum für eine Lösungsreaktion, das heißt Extraktion, dient.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einer „Präzipitationsreaktoreinheit“ eine Vorrichtung verstanden, die als Reaktionsraum für eine Ausfällungsreaktion, das heißt Präzipitation, dient. Im Zusammenhang mit der vorliegenden Erfindung wird unter einer„Batch-Betriebsweise“ eine diskontinuierliche Betriebsweise einer Reaktoreinheit verstanden, das heißt die Edukte werden in die nahezu leere, insbesondere leere, Reaktoreinheit gegeben und die Produkte nach einer bestimmten Verweilzeit aus der Reaktoreinheit genommen.
Im Zusammenhang mit der vorliegenden Erfindung wird unter„Abtrennen“ verstanden, dass ein Teil eines Gemisches von dem restlichen Teil oder den restlichen Teilen, räumlich getrennt wird und der Teil des Gemischs somit physisch nicht mehr mit dem restlichen Teil oder den restlichen Teilen in Kontakt steht.
Im Zusammenhang mit der vorliegenden Erfindung wird unter„diskontinuierlichem Erhalt“ verstanden, dass die erfindungsgemäße Verfahrensweise dergestalt ist, dass kein kontinuierlicher Produktstrom zum Erhalt der erfmdungsgemäßen Produkte führt, sondern diese in diskreten Mengeneinheiten erhalten werden.
Im Zusammenhang mit der vorliegenden Erfindung wird unter„Extraktion“ verstanden, dass eine oder mehrere Substanzen aus einem Material oder Stoffgemisch mittels eines Extraktionsmittels, das heißt Lösungsmittels, gelöst werden, das heißt im Extraktionsmittel in Lösung gehen.
Im Zusammenhang mit der vorliegenden Erfindung wird unter Druck der Druck nach Abzug des Atmosphärendrucks verstanden.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem Absenken oder einer Absenkung eines ersten Druckes aus einem für einen bestimmten Verfahrensschritt angegebenen ersten Druckbereich zu einem zweiten Druck, der in einen für einen nachfolgenden weiteren, zweiten Verfahrensschritt angegebenen weiteren, zweiten Druckbereich fällt verstanden, dass der erfmdungsgemäß im Verfahren eingesetzte erste Druck ausgewählt aus dem ersten Druckbereich auf einen abweichenden, nämlich niedrigeren, Druck ausgewählt aus den im zweiten weiteren Druckbereich, abgesenkt wird.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einer Absenkung des Drucks vorzugsweise verstanden, dass der Druck auf > 0 bar abgesenkt wird.

Im Zusammenhang mit der vorliegenden Erfindung sind in Bezug auf das im erfmdungsgemäßen Verfahren eingesetzte Reaktorsystem angegebene relative räumliche Angaben, insbesondere „vor“ und„nach“ in Richtung der erfmdungsgemäßen Verfahrensweise zu verstehen, das heißt ausgehend von Verfahrensschritt a) zu Verfahrensschritt f).
Weitere vorteilhafte Ausgestaltungen ergeben sich aus den Unteransprüchen.

Beispiel:
Im Folgenden soll schematisch eine beispielhafte Durchführung des erfindungsgemäßen Verfahrens anhand der Figur 1 dargestellt werden.
Mikroalgenbiomasse (Al), zum Beispiel aus Bacillariophyceae oder Prymnesiophyceae, wird in einem optionalen Verfahrensschritt in einer Vorrichtung zum Zellaufschluss der Algenbiomasse (51) zerkleinert.
Die zerkleinerte Mikroalgenbiomasse wird in einem optionalen Verfahrensschritt anschließend in einer Vorrichtung zur Trocknung der Algenbiomasse (52) getrocknet und so eine partikuläre Algenbiomasse erzeugt.
Die partikuläre Mikroalgenbiomasse wird in eine Extraktionsreaktoreinheit (10) überführt, in der bei einem Druck von 3 bis 100 bar sowie einer Temperatur von 60 bis 150 °C aus der Algenbiomasse mittels des mindestens einen apolaren organischen Lösungsmittels, zum Beispiel n-Hexan, ein Gemisch umfassend einen Algenextrakt sowie eine extraktionsunterzogene Algenbiomasse erhalten wird. Die extraktionsunterzogene Algenbiomasse wird durch das erste Filterelement (11), welches der Extraktionsreaktoreinheit (10) zugeordnet ist, auf dem Weg zur Präzipitationsreaktoreinheit (20) von dem Algenextrakt abgetrennt. Hierbei wird die Druckdifferenz, welche zwischen der Extraktionsreaktoreinheit (10) sowie der Präzipitationsreaktoreinheit (20) herrscht, für die Filtration ausgenutzt.
Der Druck wird demgemäß auf 3 bis 20 bar abgesenkt. Anschließend erfolgt in der Präzipitationsreaktoreinheit (20) eine weitere Absenkung des Druckes auf 3 bis 10 bar sowie auf eine Temperatur von 50 bis -80 °C, wobei Fucoxanthin-Kristalle aus dem Algenextrakt ausfallen. Diese ausgefallenen Fucoxanthin-Kristalle werden anschließend mittels Filtration, insbesondere unter Ausnutzung des in der Präzipitationsreaktoreinheit (20) vorhandenen Drucks von 3 bis 10 bar, durch das zweite Filterelement (21) abgetrennt, wobei ein Fucoxanthin-Kristall-haltiges Präzipitat (PI) sowie eine Fettsäure-haltige Algenextraktfraktion gewonnen werden.
Alternativ kann die Filtration durch das zweite Filterelement (21), insbesondere wenn der Druck in der Präzipitationsreaktoreinheit auf 0 bar abgesenkt wird, eine Vakuum-Filtration sein. Die Fettsäure-haltige Lipidfraktion (P2) aus dieser Fettsäure-haltige Algenextraktfraktion wird in einem weiteren Verfahrensschritt mittels Destillation in einem Destillationsapparat (30) von dem mindestens einen apolaren organischen Lösungsmittel getrennt.
Optional wird das verdampfte Lösungsmittel in einer Vorrichtung zur Lösungsmittelrückführung (40) wieder abgekühlt und zurückgeführt, um für eine erneute Extraktion im Extraktionsreaktor eingesetzt zu werden.
Nachfolgend wird eine konkrete Ausführungsform der schematischen Darstellung aufgeführt.
Als Mikroalgenbiomasse (Al) wurde die Kieselalge Phaeodactylum tricornutum (Bacillariophyceae) verwendet, welche zuvor in 180 L Flachplatten-Photobioreaktoren unter photoautotrophen Bedingungen in einer Outdoor-Pilotanlage erzeugt wurde.
Die Zerkleinerung der Algenbiomasse erfolgte mittels Rührwerkskugelmühle (PML-2, Bühler) mit einer Mahlkammer Centrex S1 und einem Mahlvolumen von 0,22 dm3 als Vorrichtung zum Zellaufschluss (51). Als Mahlkörper wurden stabilisierte Keramikkugeln (Draison Ytterium Ultra Power, Bühler) mit einem Durchmesser 0,3 mm und einer Dichte von 5.95 g cm 3 verwendet. Die in einem wässrigen Medium suspendierten Algenzellen wurden kontinuierlich aus einem Vorlagebehälter durch eine Pumpe (PSF 3, Ragazzini) in den Mahlraum der Rührwerkskugelmühle gefördert. Die Kontrolle des Aufschlussgrades erfolgte über einen Zeitraum von 3 h durch Messung des Proteingehalts im Überstand mittels FTIR-Spektroskopie.
Anschließend wurde die Algenbiomasse mit einer Gefriertrocknungsvorrichtung der Fa. Christ über einen Zeitraum von 24 h unter Vakuum getrocknet. Die partikuläre im Vakuum getrocknete Algenbiomasse wurde danach in eine Extraktionsreaktoreinheit (10) überführt.
Für die Extraktion in der Extraktionsreaktoreinheit (10) wurde ein Druck von 100 bar und eine Temperatur von 100 °C verwendet. Die Verweilzeit während der Fest-Flüssig-Extraktion betrug 20 min. Als Lösungsmittel diente n-Hexan und als Extraktionsreaktoreinheit (10) ein druckfester Edelstahlbehälter. Als erstes Filterelement (11) wurde ein gesinterter Edelstahlfilter mit einer Porengröße von 0,25 pm zur Abtrennung der extraktionsunterzogenen Algenbiomasse.
Die extraktionsunterzogene Algenbiomasse wurde durch das erste Filterelement (11), welches der Extraktionsreaktoreinheit (10) zugeordnet ist, auf dem Weg zur Präzipitationsreaktoreinheit (20) von dem Algenextrakt abgetrennt, wobei der Druck von ursprünglich 100 auf 30 bar in der Präzipitationsreaktoreinheit abgesenkt wurde.
Nachfolgend wurde der Druck in der Präzipitationsreaktoreinheit (20) auf 3 bar und die Temperatur auf 25 °C gesenkt, wobei Fucoxanthin-Kristalle in Form eines Präzipitats aus dem Algenextrakt ausfallen.
Diese ausgefallenen Fucoxanthin-Kristalle wurden anschließend mittels Filtration, insbesondere unter Ausnutzung des in der Präzipitationsreaktoreinheit (20) vorhandenen Drucks von 3 bar, durch das zweite Filterelement (21) abgetrennt, wobei ein Fucoxanthin-Kristall-haltiges Präzipitat sowie eine Fettsäure-haltige Algenextraktfraktion gewonnen wurden. Als zweites Filterelement (21) diente ein Filter aus Polytetrafluorethylen (PTFE) mit einer Porengröße von 1 pm. Die Fettsäure-haltige Lipidfraktion aus dieser Fettsäure-haltige Algenextraktfraktion wurde in einem weiteren Verfahrensschritt mittels Destillation in einem Destillationsapparat (30) von dem mindestens einen apolaren organischen Lösungsmittel, n-Hexan, getrennt. Das n-Hexan wurde in der Extraktionsreaktoreinheit (10) anschließend für nachfolgende Durchführungen wiederverwendet. Die im Beispiel eingesetzte P. tricornutum Algenbiomasse wies, bezogen auf die Gesamttrockensubstanz der Algenbiomasse, einen Gesamtfettsäuregehalt von 104,7 mg/g, einen Eicosapentaensäure (EPA)-Gehalt von 54,5 mg/g und einen Fucoxanthingehalt von 17,6 mg/g auf.
Als Fucoxanthin-Kristall-haltiges Präzipitat (PI) konnten im Beispiel bezogen auf 1 g der eingesetzten P. tricornutum Algenbiomasse 7,1 mg Fucoxanthin-Kristall-haltiges Präzipitat gewonnen werden (siehe Figur 3). Das Fucoxanthin-Kristall-haltige Präzipitat wies einzelne Fucoxanthin-Kristalle (Figur 4 B und 4C) in Form von zusammengelagerten Agglomeraten (Figur 4A) auf. Das Fucoxanthin-Kristall-haltige Präzipitat wies einen Massenanteil, das heißt eine Reinheit, von Fucoxanthin von 98,3 Gew.-% (UHPLC-MS) (bezogen auf Gesamttrockensubstanz) auf (siehe Figur 2). Bezogen auf die in der eingesetzten Ausgangsbiomasse verfügbare Menge an Fucoxanthin entspricht dies einer Ausbeute 40 Gew.-% (TS) (Ausbeute bezogen auf Trockengewicht des Fucoxanthins im Ausgangsmaterial).
Bezogen auf 1 g der eingesetzten P. tricornutum Algenbiomasse konnten 87,85 mg Fettsäuren und 49,0 mg Eicosapentaensäure (EPA) gewonnen werden. Somit sind etwa 55,8 Gew.-% (bezogen auf Gesamtgewicht der Trockensubstanz an Fettsäuren in der Fettsäure-haltigen Lipidfraktion) der Fettsäuren in der Fettsäure-haltigen Lipidfraktion (P2) EPA. Bezogen auf die in der eingesetzten Ausgangsbiomasse verfügbare Menge an Fettsäuren entspricht dies einer Ausbeute von 83,9 Gew.-% (TS) (Ausbeute bezogen auf Trockengewicht der Fettsäuren im Ausgangsmaterial). Bezogen auf die in der eingesetzten Ausgangsbiomasse verfügbare Menge an Eicosapentaensäure (EPA) wurde eine Ausbeute von 89,9 Gew.-% (TS) (Ausbeute bezogen auf Trockengewicht des EPA im Ausgangsmaterial) erreicht. In Anbetracht der Gesamttrockensubstanz der Fettsäure-haltigen Lipidfraktion (P2) von 134 mg (für 1 g Biotrockenmasse) stellen die Fettsäuren somit einen Anteil von 65,6 Gew.-% (bezogen auf Gesamttrockensubstanz) in der Fraktion dar.

Claims

ANSPRÜCHE
1. Verfahren zum Erhalt eines Fucoxanthin-haltigen Präzipitats und einer Fettsäure-haltigen Lipidfraktion aus Algenbiomasse, umfassend die folgenden Verfahrensschritte: a) Bereitstellen von mindestens einem apolaren organischen Lösungsmittel und Algenbiomasse, b) Extraktion von Fucoxanthin und Fettsäuren mittels des in Verfahrensschritt a) bereitgestellten mindestens einen apolaren organischen Lösungsmittels aus der in Verfahrensschritt a) bereitgestellten Algenbiomasse in mindestens einer
Extraktionsreaktoreinheit (10), der mindestens ein erstes Filterelement (11) zugeordnet ist, bei einer Temperatur von 60 bis 150 °C und einem Druck von 1,5 bis 100 bar zum Erhalt eines Gemisches umfassend einen Algenextrakt und eine extraktionsunterzogene Algenbiomasse, c) Abtrennen der extraktionsunterzogenen Algenbiomasse aus dem in
Verfahrensschritt b) erhaltenen Gemisch mittels Filtrierung durch das mindestens eine erste Filterelement (11) zum Erhalt eines filtrierten Algenextraktes und Einleiten des erhaltenen filtrierten Algenextraktes in mindestens eine Präzipitationsreaktoreinheit (20), der mindestens ein zweites Filterelement (21) zugeordnet ist, d) Absenken der Temperatur des filtrierten Algenextrakts auf -80 bis 50 °C in der Präzipitationsreaktoreinheit (20) zur Präzipitation von Fucoxanthin-Kristallen zum Erhalt eines Gemisches aus Fucoxanthin-Kristall-haltigem Präzipitat und Fettsäure-haltiger Algenextraktfraktion, e) Abtrennen des Fucoxanthin-Kristall-haltigen Präzipitats aus dem Gemisch aus Fucoxanthin-Kristall-haltigem Präzipitat und Fettsäure-haltiger
Algenextraktfraktion mittels Filtrierung durch das mindestens eine zweite Filterelement (21) zum Erhalt eines Fucoxanthin-Kristall-haltigen Präzipitats und einer Fettsäure-haltigen Algenextraktfraktion, und f) Abtrennen einer Fettsäure-haltigen Lipidfraktion aus der Fettsäure-haltigen
Algenextraktfraktion durch Verdampfen des mindestens einen apolaren organischen Lösungsmittels zum Erhalt der Fettsäure-haltigen Lipidfraktion.
2. Verfahren nach Anspruch 1, wobei die Temperatur in Verfahrensschritt b) 85 bis 120 °C beträgt.
3. Verfahren nach einem der vorhergehenden Ansprüche, wobei der Druck nach
Durchführung der Extraktion in Verfahrensschritt b) in Verfahrensschritt c) gesenkt wird.
4. Verfahren nach Anspruch 3, wobei der Druck nach Durchführung der Extraktion in
Verfahrensschritt b) in Verfahrensschritt c) auf 1 bis 30 bar gesenkt wird.
5. Verfahren nach einem der vorhergehenden Ansprüche, wobei der Druck nach
Durchführung der Präzipitation in Verfahrensschritt d) in Verfahrensschritt e) gesenkt wird.
6. Verfahren nach einem der vorhergehenden Ansprüche, wobei das in Verfahrensschritt f) verdampfte mindestens eine apolare organische Lösungsmittel in einem zusätzlichen Verfahrensschritt g) verflüssigt wird und das flüssige mindestens eine apolare organische Lösungsmittel in Verfahrensschritt a) wieder bereitgestellt wird.
7. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt a) bereitgestellte Algenbiomasse einem Zellaufschluss zugeführt und/oder getrocknet wurde.
8. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt a) bereitgestellte Algenbiomasse eine partikuläre Algenbiomasse ist.
9. Verfahren nach einem der vorhergehenden Ansprüche, wobei das mindestens eine
apolare organische Lösungsmittel ein Alkan, insbesondere ein n-Alkan, ist.
10. Verfahren nach einem der vorhergehenden Ansprüche, wobei der in Verfahrensschritt b) erhaltene Algenextrakt weitere Carotinoide, Polyphenole und/oder Phytosterole umfasst, welche in Verfahrensschritt f) von dem mindestens einen apolaren organischen
Lösungsmittel zusammen mit der Fettsäure-haltigen Lipidfraktion aus der Fettsäure haltigen Algenextraktfraktion abgetrennt werden.
11. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt e) erhaltenen Fucoxanthin-Kristalle im Fucoxanthin-Kristall-haltigen Präzipitat einen Durchmesser von 0,1 bis 10 mih aufweisen.
12. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt e) erhaltenen Fucoxanthin-Kristalle im Fucoxanthin-Kristall-haltigen Präzipitat in Form von Agglomeraten, insbesondere in einer Größe von 10 bis 100 mih, vorliegen.
13. Verfahren nach einem der vorhergehenden Ansprüche, wobei die Abtrennung des
Fucoxanthin-Kristall-haltigen Präzipitats in Verfahrensschritt e) mittels Überdruck-, Unterdrück- oder Vakuum-unterstützter Filtration erfolgt.
14. Verfahren nach einem der vorhergehenden Ansprüche, wobei die Verweilzeit im
Extraktionsreaktor in Verfahrensschritt b) 1 bis 60 Minuten beträgt.
15. Verfahren nach einem der vorhergehenden Ansprüche, wobei die Verweilzeit im
Präzipitationsreaktor in Verfahrensschritt d) 1 bis 60 Minuten beträgt.
16. Verfahren nach einem der vorhergehenden Ansprüche, wobei die mindestens eine
Extraktionsreaktoreinheit (10) und/oder die mindestens eine Präzipitationsreaktoreinheit (20) ein Wirbelbettreaktor ist oder Wirbelbettreaktoren sind.
17. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 50 bis 70 Gew.-% EPA (bezogen auf Gesamtgewicht der Trockensubstanz an Fettsäuren in der Fettsäure-haltigen
Lipidfraktion) aufweist.
18. Verfahren nach einem der vorhergehenden Ansprüche, wobei die in Verfahrensschritt f) erhaltene Fettsäure-haltige Lipidfraktion 50 bis 70 Gew.-% DHA (bezogen auf Gesamtgewicht der Trockensubstanz an Fettsäuren in der Fettsäure-haltigen
Lipidfraktion) aufweist.
