Alcoholic compositions with a content of octenidine dihydrochloride
The invention relates to special alcoholic compositions with a content of octenidine dihydrochloride. The compositions are used in particular for hygienic hand disinfection and for surgical hand disinfection. Furthermore, the invention relates to the use of octenidine dihydrochloride for improving the effectiveness of alcoholic agents for skin antisepsis, for hygienic and surgical hand washing, and for hygienic and surgical hand disinfection.
The effectiveness of alcoholic compositions for skin antisepsis, mucosa antisepsis, wound disinfection, hygienic hand disinfection, hygienic hand washing, surgical hand disinfection and surgical hand washing is known. On account of their effectiveness against a large number of bacteria, fungi and viruses, aliphatic alcohols such as ethanol, n-propanol (1 -propanol) and isopropanol (2-propanol), for example, are used for this purpose. Their effectiveness starts after just a short time, particularly if the specified alcohols are present in a comparatively high concentration. By contrast, compositions with a lower content of aliphatic alcohols are more skin compatible and consequently advantageous for hand disinfection or hand washing.
The combination of aliphatic alcohols with active ingredients with demonstrated remanence effect in principle offers advantages for skin antisepsis, hand disinfection and hand washing. These combinations can, for example, be advantageous since smaller amounts of alcohol are then required and thus effective but fewer degreasing preparations are available. However, it has been found that active ingredients with remanence effect cannot be formulated directly to give disinfectants based on aliphatic alcohols. For example, the active ingredient chlorhexidine is not sufficiently stable in ethanol (cf. inter alia US 5,800,827 B).
Octenidine dihydrochloride is an active ingredient with demonstrated remanence effect. For example, the product octeniderm® (Schijlke & Mayr GmbH, Norderstedt, Federal Republic of Germany) comprises, as medically effective constituents, 0.1 % by weight of octenidine dihydrochloride, 30.0% by weight of 1 - propanol and 45.0% by weight of 2-propanol. The product octeniman® comprises 0.1 % by weight of octenidine dihydrochloride, 40.0% by weight of 1 -propanol and 30.0% by weight of 2-propanol. On account of the comparatively high content of aliphatic alcohols, they are only suitable to a limited extent as washing preparations. They are also only virucidal to a limited extent. By contrast, it is advantageous for hygienic hand disinfection if a formulation also has good effectiveness against non-enveloped viruses such as noroviruses, adenoviruses and polioviruses. This would be present in particular in formulations with a high ethanol content.
DE 10 2010 054 155 A1 discloses the use of aliphatic alcohols in the production of a disinfectant for preventing contamination of the disinfectant with bacterial spores. An effect of the disinfectant against typical germs of transient and resident skin flora (such as fungi and bacteria, including Staphylococcus aureus) and an effect against viruses, as are important in particular for hygienic and surgical hand disinfection, are not discussed in DE 10 2010 054 155 A1.
DE 10 201 1 077 432 A1 discloses the use of bispyridiniumalkanes. The compositions are used on inanimate surfaces, for example for surface or instrument disinfection, or on animate surfaces, such as the human skin.
The object of the present invention was to provide compositions, especially for hygiene hand disinfection and for surgical hand disinfection, which do not automatically have a high content of aliphatic alcohol (but, if desired, can be formulated with a high content of aliphatic alcohol). These compositions should thus also be suitable as washing preparations and, moreover, have good virucidal effectiveness.
This object is achieved according to the invention by a composition which comprises a) at least 20% by weight of ethanol, b) at least 1 % by weight of n-propanol and c) at least 0.02% by weight of octenidine dihydrochloride.
Surprisingly, for the combination of aliphatic alcohols with octenidine dihydrochloride, a mixture of ethanol with 1 -propanol [mixture (i)] has proven to be antimicrobially more effective than a mixture of ethanol with isopropanol [mixture (ii)].
Furthermore, the advantageous results of the quantitative suspension experiments were able to be confirmed in experiments that simulate conditions in practice relating to hygienic hand disinfection in accordance with EN 1500.
The combination of aliphatic alcohols with octenidine dihydrochloride offers the option, particularly when using ethanol in relatively high concentrations, to utilize its virucidal properties and, by virtue of the content of octenidine dihydrochloride, to obtain a virucidal composition with broad antibacterial effectiveness and a remanence effect.
Furthermore, this surprising finding of the improved effectiveness due to the content of specifically 1 -propanol is also interesting with regard to washing and antiseptic preparations.
Accordingly, in a first aspect, the invention relates to a composition which comprises:
a) - At least 20% by weight of ethanol,
b) - At least 1 % by weight of n-propanol and
c) - At least 0.02% by weight of octenidine dihydrochloride. In the description of the present invention, the stated content of ethanol refers to the pure substance (and does not include denaturant present for example in ethanol of technical or medical purity or water).
According to a particular aspect, the composition as hereinbefore defined comprises up to 95% by weight of ethanol.
A preferred amount of a) ethanol is 23 to 95% by weight, preferably 25 to 85% by weight, in particular 30 to 80% by weight, such as for example 34% by weight to 75% by weight of ethanol.
In a further preferred embodiment, the composition comprises less than 78% by weight of ethanol, such as at most 75% by weight of ethanol.
According to another particular aspect, the composition as hereinbefore defined, comprises up to 30% by weight of n-propanol.
A preferred amount of b) n-propanol is at least 1 .5% by weight, preferably at least 1 .8% by weight, in particular at least 2.0% by weight, such as 2.0 to 30% by weight or 2.2 to 30% by weight, for example 2.5 to 25% by weight, of n-propanol. Particular preference is given to an amount of 2.5 to 20% by weight of n-propanol, such as 2.5 to 15% by weight of n-propanol.
According to a particular aspect, the composition as hereinbefore defined, comprises up to 0.25% by weight of octenidine dihydrochloride.
In preferred embodiments, the amount of c) octenidine dihydrochloride is 0.025 to 0.25% by weight, preferably 0.03 to 0.2% by weight, in particular 0.035 to 0.18% by weight, such as 0.04 to 0.16% by weight, for example about 0.05 to about 0.15% by weight, of octenidine dihydrochloride.
According to a more particular aspect, the composition as hereinbefore defined, comprises:
a) - From 25 to 85% by weight of ethanol,
b) - From 2% to 30% by weight of n-propanol and
c) - From 0.03 to 0.2% by weight of octenidine dihydrochloride; and still more particularly:
a) - From 30 to 80% by weight of ethanol,
b) - From 2.5% to 25% by weight of n-propanol and
c) - From 0.05 to 0.15% by weight of octenidine dihydrochloride;
Besides the obligatorily prescribed components a) ethanol, b) n-propanol and c) octenidine dihydrochloride, the composition according to the invention optionally further comprises:
d) - One or more surfactants, preferably amphoteric or nonionic surfactants. As nonionic surfactant, all suitable nonionic surfactants can be used, where (i) (fatty) alcohol ethoxylates, (ii) sorbitan esters, (iii) alkyl glycosides (in particularly alkyl polyglucosides), (iv) amine oxides and (v) ethylene oxide/propylene oxide block copolymers are preferred.
The (i) alcohol polyalkoxylates include fatty alcohol alkoxylates, e.g. isodecyl ethoxylates with various fractions of ethylene oxide, isotridecyl ethoxylates, polyethylene glycol ethers of stearyl, lauryl and cetyl and oleyl alcohol. Here, the alcohols can have been alkoxylated with ethylene oxide, propylene oxide or any desired mixtures of ethylene oxide and propylene oxide. Alcohol polyalkoxylates are known inter alia under the names Lutensol®, Marlipal®, Marlox®, Brij® and Plurafac®. As nonionic surfactant, particular preference is given to lauryl alcohol ethoxylates.
Furthermore, the nonionic surfactants (ii) used are sorbitan esters, which are mostly present as oleates, stearates, laurates and palmitates and which are referred to as polysorbates (e.g. Tween®).
Moreover, the nonionic surfactant can be a (iii) alkyl glycoside, such as an alkyl glucoside (i.e. an alkyl glycoside of glucose), more preferably a C8-C2o- alkylpolyglucose, in particular a C8-Ci6-alkylpolyglucose of a fatty alcohol, where a laurylpolyglucose, a decylpolyglucose or a mixture thereof is preferred. The carbon chain length in the case of the cocoylpolyglucose is 8 to 16 atoms, in the case of the laurylpolyglucose 12 to 16 carbon atoms and in the case of the decylpolyglucose likewise 8 to 16 carbon atoms.
A typical amount of alkyl glycoside is 0.03 to 10% by weight, preferably 0.06 to 5% by weight, in particular 0.1 to 2% by weight.
According to the invention, in principle all suitable amine oxides can be used as (iv) amine oxide. The amine oxides, which are N-oxides of tertiary amines, include aliphatic amine oxides, cyclic amine oxides (such as N-alkylmorpholine oxide) and aromatic amine oxides (such as pyridine N-oxides). In a preferred embodiment, the amine oxide has the general formula

in which R1 is methyl, ethyl or 2-hydroxyethyl, R2 is methyl, ethyl or 2- hydroxyethyl, R1 and R2 together can be morpholine, R3 is alkyl having 8 to 18 carbon atoms or R4CONH(CH2)n, where R4 is alkyl having 8 to 18 carbon atoms and n is in the range from 1 to 10, preferably 1 to 5, more preferably 2 to 4, and in particular 3, and 2- hydroxyethyl can be condensed with 1 to 2000 ethylene oxide, ethylene oxide/propylene oxide or propylene oxide units.
Examples of amine oxides are cocamidopropylamine oxide, N-cocomorpholine oxide, decyldimethylamine oxide, dimethylcetylamine oxide, dimethylcocamine oxide, dimethyl hydr. tallow-amine oxide, dimethyllaurylamine oxide, dimethylmyristylamine oxide, (2-hydroxyethyl)cocamine oxide and oleamine oxide. See also "International Cosmetic Ingredient Dictionary and Handbook", 10th edition 2004, volume 3, pages 2268-2275 (Surfactants - Cleansing Agents).
In a preferred embodiment, the amine oxide is cocamidopropylamine oxide, i.e. R4CO is the acyl radical derived from the fatty acids of coconut oil, n = 3, and R1 and R2 are methyl. This product is sold as Rewominox B 204 by Goldschmidt, Federal Republic of Germany.
A typical amount of amine oxide is 0.03 to 10% by weight, preferably 0.06 to 5% by weight, in particular 0.1 to 2% by weight.
Likewise of suitability as surfactant are amphoteric surfactants, for example betaines. Suitable betaines are described in EP 560 1 14 A2. Particular preference is given to cocamidopropylbetaine. A typical amount of betaine is 0.03 to 10% by weight, preferably 0.06 to 5% by weight, in particular 0.1 to 2% by weight.
Moreover suitable as surfactant are cationic surfactants such as quaternary ammonium salts. In principle, all suitable quaternary ammonium compounds can be used according to the invention. Preferably, the quaternary ammonium compound is a dialkyldimethylammonium salt.
Quaternary ammonium salts used according to the invention are given by the formula [R1R2R3(CH3)N]+[X]", where R1 to R3 can be identical or different and are selected from d- to C30-alkyl, -aralkyl, -alkenyl and mixed groups which can have one or more atoms selected from O, S, N and P, where R1 to R3 are, for example, C8- to Cis-alkyl, benzyl or methyl, preferably C9- to Ci8-alkyl, benzyl or methyl, such as Ci6- alkyl, benzyl or methyl. X is an anion (of an inorganic or organic acid). Here, both anion and cation of the quaternary ammonium salt can be polyvalent ions, which gives rise to a stoichiometry [A(n+)]m[K(m+)]n.
According to the invention, suitable quaternary ammonium salts are all quaternary ammonium salts of the aforementioned formula known in the prior art, as are disclosed for example in WO 00/63337, to which reference is made here. However, preference is given to using dialkyldimethylammonium salts, for example dialkyldimethylammonium chlorides, the alkyl chains of which are selected independently of one another from C8- to Ci8-alkyl, preferably C9- to Ci8-alkyl, such as Ci6-alkyl. In the dialkyldimethylammonium salts, one of the methyl groups can be an alkoxylated, for example ethoxylated, hydromethyl group.
Quaternary ammonium salts preferably used according to the invention are compounds of the formulae [R1N(CH3)3]+[X]", [R1R2N(CH3)2]+[X]" and [R1R2R3(CH3)N]+[X]", where R1 to R3, independently of one another, are selected from C8- to Cie-alkyl and -(CH2-CHR40)n-R5, where n is a number from 1 to 20, preferably 1 to 5, and R4 and R5, which can be identical or different, are H and/or d- to C4-alkyl, preferably H.
Examples of anions and classes of anions of the quaternary ammonium salts used according to the invention are hydroxide, sulphate, hydrogen sulphate, methosulphate, ethosulphate, lauryl sulphate, lauryl ether sulphate, cellulose sulphate, sulphamate, halide (fluoride, chloride, bromide, iodide), nitrite, nitrate, carbonate, hydrogen carbonate, phosphate, alkyl phosphate, metaphosphate, polyphosphate, thiocyanate (rhodanide), carboxylic acid salt such as benzoate, lactate, acetate, propionate, citrate, succinate, glutarate, adipate, toluenesulphonate (tosylate) and salicylate. Particularly preferred anions are chloride and propionate.
Particularly preferably, the quaternary ammonium salts mecetronium etilsulphate (hexadecyl(ethyl)dimethylammonium ethylsulphate) and benzalkonium chloride are used.
Besides the obligatorily prescribed components a) ethanol, b) n-propanol and c) octenidine dihydrochloride, and the optional component d) the composition according to the invention optionally further comprises:
e) - One or more solvents.
Preferred solvents are glycols and water, and mixtures thereof. A preferred solvent is water.
Besides the obligatorily prescribed components a) ethanol, b) n-propanol and c) octenidine dihydrochloride, and the optional components d) and or e), the composition according to the invention optionally further comprises:
f) - One or more active ingredients and/or auxiliaries.
Examples of active ingredients and/or auxiliaries which can optionally be present in compositions according to the invention are skincare additives, refatting agents, perfumes, fragrances, thickeners, pH regulators, emollients, humectants and dyes. These are inter alia:
polyols, which act as skincare additives, refatting agents and humectants, such as glycerol, erythritol, 1 ,2,6-hexanetriol, inositol, lactitol, maltitol, mannitol, methylpropanediol, phytantriol, polyglycerols, sorbitol and xylitol, where glycerol is particularly preferred,
- glycerol esters, preferably coconut fatty acid glycerol monoesters, fatty alcohol esters such as octanoic acid cetearyl ester (cetearyl octanoate) myristic acid isopropyl ester (isopropyl myristate), palmitic acid isopropyl ester (isopropyl palmitate) and fatty acid glyceryl triesters, which act as refatting agents,
allantoin, which acts as skincare additive, and/or
sodium gluconate, which acts as buffer.
For example, the composition according to the invention preferably comprises one or more refatting agents.
Refatting agents
Compositions according to the invention preferably comprise d) one or more fatty acid esters of monohydric C to C6-alkyl alcohol.
Preferably, the alkyl alcohol of the fatty acid ester is selected from methanol, ethanol, n- and isopropanol. Particularly preferably, the alkyl alcohol of the fatty acid ester is specifically isopropanol.
It is also preferred that the fatty acid of the fatty acid ester is selected from d0- to Cie-fatty acids, preferably Ci2- to Ci6-fatty acids, where the fatty acid is preferably myristic acid.
Consequently, it is preferred in all embodiments of the present invention that the refatting component is isopropyl myristate.
Component, preferably myristic acid isopropyl ester, is present in the composition according to the invention preferably in an amount of from 0.02 to 5% by weight, based on the weight of the composition, preferably 0.1 to 4% by weight, more preferably 0.2 to 3% by weight, in particular 0.3 to 2.0% by weight, such as 0.5 to 1 .8% by weight, for example 0.6 to 1 .6% by weight, such as about 0.75 or about 1 .4% by weight.
Furthermore, it is preferred that compositions according to the invention have no content of aromatic alcohol (as are obligatorily prescribed according to
DE 10 201 1 077 432 A1 , if aliphatic alcohol is present).
According to a further preferred embodiment, the content of isopropanol in the compositions is limited and is, for example, less than 25% by weight, preferably less than 20% by weight, in particular less than 15% by weight, such as less than 10% by weight, for example less than 5% by weight, of isopropanol.
Furthermore, the content of chlorhexidine derivative is preferably limited and is less than 0.3% by weight, such as less than 0.2% by weight, where compositions according to the invention are particularly preferably free from chlorhexidine derivative. The chlorhexidine derivative, the presence of which is preferably limited according to the invention and the presence of which is particularly preferably excluded, is preferably chlorhexidine digluconate. Moreover, in a further embodiment, the invention relates to the composition for use in a method for skin antisepsis, hygienic hand disinfection, hygienic hand washing, surgical hand disinfection or surgical hand washing.
The invention thus also relates to the use of the composition for skin antisepsis, hygienic hand disinfection, hygienic hand washing, surgical hand disinfection or surgical hand washing.
Also thus in accordance with the invention are methods for skin antisepsis, hygienic hand disinfection, hygienic hand washing, surgical hand disinfection or surgical hand washing, in which the composition is left to act on the area of skin in question or hands.
The application of the compositions according to the invention takes place in particular in methods for:
i) Skin antisepsis,
ii) Hygienic hand disinfection (in accordance with EN 1500, ASTM 1 174), iii) Hygienic hand washing (in accordance with EN 1499),
iv) Surgical hand disinfection, or
v) Surgical hand washing (in accordance with EN 12791 , ASTM 1 1 15).
In one embodiment, the invention thus relates to the application during hygienic hand disinfection in accordance with EN 1500, preferably in accordance with EN 1500:2013. In a further embodiment, the invention relates to the application of the compositions for hygienic hand disinfection in accordance with ASTM 1 174, preferably in accordance with ASTM 1 174-13.
In a further embodiment, the invention relates to the application of the compositions during hygienic hand washing in accordance with EN 1499, preferably in accordance with EN 1499:2013.
In a further embodiment, the invention relates to the application of the compositions during surgical hand washing in accordance with EN 12791 , preferably in accordance with EN 12791 :2013. Moreover, the invention relates to the application of the compositions during surgical hand washing in accordance with ASTM 1 1 15, preferably in accordance with ASTM 1 1 15-1 1 .
Furthermore, the invention relates to the use of octenidine dihydrochloride for improving the remanence effect of a composition which comprises
a) at least 20% by weight of ethanol and
b) at least 1 % by weight of n-propanol.
The compositions according to the invention are used in particular for controlling transient pathogens in hygienic hand disinfection. For example, compositions according to the invention are effective against human pathogenic bacteria such as Escherichia coli or yeast-like fungi such as Candida albicans. Furthermore, compositions according to the invention are effective against enveloped viruses such as HIV, HBV or HCV and surrogate viruses thereof (BVDV or vaccinia virus) and, in a preferred embodiment, also against non-enveloped viruses such as noroviruses, adenoviruses and polio viruses. The compositions according to the invention are also used for reducing the resident skin flora during surgical hand disinfection. These are preferably bacterial Gram-positive microorganisms which are characterized by not being able to form any endospores, such as Staphylococcus spp. or Corynebacterium spp.
The advantages of the present invention are evident in particular from the following examples. Stated percentages refer to the weight, unless stated otherwise.
Examples
Experiment 1 : Effectiveness of various combinations of aliphatic alcohols with octenidine dihydrochloride (quantitative suspension experiment according to the German Society for Hygiene and Microbiology (DGHM) 2001 , dirty conditions, test organism: Candida albicans)
40g denatured ethanol in 100g solution correspond to about 37.2% by weight ethanol. ** MEK: methyl ethyl ketone.
The results of the effectiveness for a contact time of 15s, 30s and 60s are shown graphically in Fig. 1 (CT = contact time; Oct = octenidine dihydrochloride; EtOH = ethanol; 1 -propOH = 1 -propanol; 2-propOH = 2-propanol). This includes data in the kinetic effectiveness range, in which saturation of the effect is still not present, for which reason the different effectiveness becomes clear:
- Experiment A shows the effectiveness of 46.5% by weight ethanol.
- Experiment G shows the effectiveness of 0.05% by weight octenidine dihydrochlonde.
- Experiment B (46.5% by weight ethanol and 0.05% by weight octenidine dihydrochlonde) demonstrates that octenidine dihydrochlonde in ethanol inhibits the effectiveness for example at 30s.
- Experiment C shows the effectiveness of 37.2% by weight ethanol in combination with 10% by weight isopropanol.
- Experiment D shows that an inhibition due to octenidine dihydrochlonde also arises for this mixture of alcohols.
- Experiment E shows the effectiveness of 37.2% by weight ethanol in combination with 10% by weight n-propanol.
- Experiment F demonstrates then that no inhibition arises upon adding octenidine dihydrochlonde.
Experiment 2
Moreover, the following formulations were tested in accordance with EN 1500:1997 (30 sec, 3 ml). The reference alcohol used in each case was the standard provided in EN 1500:1997 (2 x 3 ml, 60% (v/v) 2-propanol, 60 sec). The tests were carried out in each case as envisaged in EN 1500:1997, with 12-15 healthy subjects and evaluated statistically.

- Formulation I with content of ethanol and n-propanol is effective. Upon adding 0.1 % by weight octenidine dihydrochloride (formulation II), the effectiveness is retained.
- Formulation III with content of ethanol and isopropanol is effective. This effectiveness is not retained on adding 0.1 % by weight of octenidine dihydrochloride (formulation IV), and specifically also not if the added amount of octenidine dihydrochloride is reduced to 0.05% by weight (formulation V).

1 . Composition which comprises
a) at least 20% by weight of ethanol,
b) at least 1 % by weight of n-propanol and
c) at least 0.02% by weight of octenidine dihydrochloride.
2. Composition according to Claim 1 characterized in that it comprises up to 95% by weight of ethanol.
3. Composition according to Claim 1 or 2, characterized in that the amount of ethanol is 25 to 85% by weight.
4. Composition according to Claim 3, characterized in that the amount of ethanol is 30 to 80% by weight.
5. Composition according to Claim 1 or 2, characterized in that it comprises up to 30% by weight of n-propanol.
6. Composition according to anyone of Claims 1 to 5, characterized in that the amount of n-propanol is 2% to 30% by weight.
7. Composition according to Claim 6, characterized in that the amount of n- propanol is 2.5% to 25% by weight.
8. Composition according to Claim 7, characterized in that the amount of n- propanol is 2.5% to 15% by weight.
9. Composition according to anyone of Claim 1 , 2 or 5, characterized in that it comprises up to 0.25% by weight of c) octenidine dihydrochloride.
10. Composition according to any one of claims 1 to 9, characterized in that the amount of octenidine dihydrochloride is 0.03 to 0.2% by weight.

d) - One or more surfactants selected from lauryl alcohol ethoxylates, laurylpolyglucose, decylpolyglucose, or a mixture thereof, cocamidopropylamine oxide and cocamidopropylbetaine.
13. Composition according to any one of claims 1 to 12, characterized in that it further comprises:
e) - One or more solvents, preferably water.
14. Composition according to any one of claims 1 to 13, characterized in that it further comprises:
f) - One or more active ingredients and/or auxiliaries selected from glycerol, erythritol, 1 ,2,6-hexanetriol, inositol, lactitol, maltitol, mannitol, methylpropanediol, phytantriol, sorbitol and xylitol, preferably glycerol, coconut fatty acid glycerol monoesters, myristic acid isopropyl ester, octanoic acid cetearyl ester, palmitic acid isopropyl ester, and fatty acid glyceryl triesters, allantoin, and sodium gluconate.
15. Composition according to Claim 14 characterized in that it further comprises: f) 0.02 to 5% by weight of isopropyl myristate.
16. Composition according to one of the preceding claims, characterized in that it has no content of aromatic alcohol.
17. Composition according to one of the preceding claims, characterized in that it comprises less than 25% by weight of isopropanol, preferably less than 20% by weight, in particular less than 15% by weight, such as less than 10% by weight, for example less than 5% by weight, of isopropanol.
18. Composition according to one of the preceding claims, characterized in that it comprises less than 0.3% by weight of chlorhexidine derivative, such as less than 0.2% by weight of chlorhexidine derivative, where the composition is particularly preferably free from chlorhexidine derivative.
19. Composition according to one of the preceding claims for use in a method for skin antisepsis, hygienic hand disinfection, hygienic hand washing, surgical hand disinfection or surgical hand washing.
20. Use of octenidine dihydrochloride for improving the remanence effect of a composition which comprises
a) - At least 20% by weight of ethanol and
