Abstract
Die Erfindung betrifft wirkstoffhaltige Pellets mit Polymerüberzug und einer mittleren Teilchengröße im Bereich von 300 bis 1100 μm, enthaltend eine pharmazeutisch wirksame Substanz, eingebettet im einer Polymermatrix aus einem oder mehreren Polymeren, dadurch gekennzeichnet, dass die Pellets eine Friabilität von höchstens 0,1 %, gemessen mit 200 g Pellets auf einer Siebmaschine mit 200 m Sieb, einem Siebdurchmesser von 20 cm und 1,5 mm Schüttel-Amplitude bei einer Schüttel-Frequenz von 50 1/sec für 10 min in Gegenwart von sechs Gummiwürfeln mit 1,8 cm Kantenlänge, aufweisen und mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen sind mit der Maßgabe, dass die Pellets im Freisetzungstest nach USP in künstlichem Magensaft bei pH 1,2 nach 120 min nicht mehr als 10 % des enthaltenen Wirkstoffs freisetzen

Pellets mit einer Wirkstoff-Matrix und einem Polymerüberzug, sowie ein Verfahren zur Herstellung der Pellets
Technischer Hintergrund
WO 01/68058 beschreibt die Verwendung einer mehrschichtigen Arzneiform, die im wesentlichen aufgebaut ist aus einem Kern mit einem pharmazeutischen Wirkstoff, einem inneren Überzug aus einem Copolymeren oder einer Mischung von Copolymeren, die sich aus 85 bis 98 Gew.-% radikalisch polymehsierten C1 - bis C4- Alkylestern der Acryl- oder der Methacrylsäure und 15 bis 2 Gew.-% (Meth)acrylat- Monomeren mit einer quaternären Ammoniumgruppe im Alkylrest zusammensetzen und einem äußeren Überzug aus einem Copolymeren, das sich aus 75 bis 95 Gew.- % radikalisch polymehsierten d- bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 5 bis 25 Gew.-% (Meth)acrylat-Monomeren mit einer anionischen Gruppe im Alkylrest zusammensetzt. Der Mengenanteil des äußeren Überzugs soll im Bereich von 10 bis 50 Gew.-% bezogen auf das Gewicht des Kerns mit dem Wirkstoff und den inneren Überzug betragen.
US 2005/0191352 beschreibt die Herstellung von pharmazeutische Wirkstoffe enthaltenden Extrudaten mit kontrollierter Wirkstofffreisetzung mittels Schmelzextrusion. Die zu extrudierenden Mischungen können neben dem Wirkstoffe Polymere wie z. B. Eudragit® RS, Eudragit® NE oder Mischungen dieser Polymere enthalten. Die Extrusion erfolgt bevorzugt in einem Doppelschneckenextruder. Die aufgetragenen Extrudate können im heißen Zustand mittels rotierender Messer zu zylindrischen oder auch zu sphärischen, elipsoiden oder linsenförmigen Partikeln zerkleinert und geformt werden. Die so erhaltenen wirkstoffhaltigen Partikel können, z. B. durch Verfüllen in Kapseln, zu multipartikulären Arzneiformen weiterverarbeitet werden.
EP 1 563 897 A1 beschreibt eine Vorrichtung zur Herstellung gerundeter Pellets (Pelletizer). Die Vorrichtung besteht aus einer vorgeschalteten Zufuhreinrichtung, für insbesondere aus einem Extruder zugeführtes verformbares Material und einem Gehäuse mit einer rotierenden Schneideinheit zum Schneiden des Materials in
Materialabschnitte, sowie Mitteln zur Erzeugung eines Gasstroms im Gehäuse, durch dessen Wirkung die Materialabschnitte gegen eine Gehäusewand prallen, wobei sie eine Abrundung erfahren. Bevorzugt ist die Gehäusewand gekühlt, um den Materialabtrag zu verringern. Die Vorrichtung eignet sich insbesondere zur Herstellung von Pellets für den Pharmabereich, indem pharmazeutische Hilfsstoffe, wie z.B. Polymere, mit mindestens einem pharmazeutischen Wirkstoff im Extruder vermengt, das Extrudat im Schneidgehäuse durch eine Düse austritt und durch Heißabschlag unter Gaskühlung zu Pellets zerteilt und ausgerundet wird.
Aufgabe und Lösung
Die Erfindung geht von überzogenen Arzneiformen aus, wie sie z. B. aus WO 01/68058 bekannt sind. In der WO 01/68058 werden für den äußeren Polymerüberzug Auftragsmengen allgemein von 10 bis 50 Gew.-% und in den Beispielen 14 bis 30 Gew.-% bezogen auf das Gewicht des Kerns mit dem Wirkstoff und dem inneren Überzug, angegeben. Die in WO 01/68058 beschriebene Arzneiform ist durch ihren Schichtaufbau aufwendig in der Herstellung. Es sollte daher eine leichter und kostengünstiger herstellbare technische Alternative bereitgestellt werden.
Die Aufgabe wir insbesondere gelöst durch
wirkstoffhaltige Pellets mit Polymerüberzug und einer mittleren Teilchengröße im Bereich von 300 bis 1100 μm, enthaltend eine pharmazeutisch wirksame Substanz, eingebettet im einer Polymermatrix aus einem oder mehreren Polymeren, dadurch gekennzeichnet, dass die Pellets eine Friabilität von höchstens 0,1 %, gemessen mit 200 g Pellets auf einer Siebmaschine mit 200 μm Sieb, einem Siebdurchmesser von 20 cm und 1 ,5 mm Schüttel-Amplitude bei einer Schüttel-Frequenz von 50 1/sec für 10 min in Gegenwart von sechs Gummiwürfeln mit 1 ,8 cm Kantenlänge, aufweisen und mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen sind mit der Maßgabe, dass die Pellets im Freisetzungstest nach USP in künstlichem Magensaft bei pH 1 ,2 nach 120 min nicht mehr als 10 % des enthaltenen Wirkstoffs freisetzen
Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung der Pellets.
Die Erfindung betrifft weiterhin multipartikuläre Arzneiformen, enthaltend ein oder mehrere der erfindungsgemäßen Pellets.
Ausführung der Erfindung
Die Erfindung betrifft
wirkstoffhaltige Pellets mit Polymerüberzug und einer mittleren Teilchengröße im Bereich von 300 bis 1100 μm, enthaltend eine pharmazeutisch wirksame Substanz, eingebettet im einer Polymermatrix aus einem oder mehreren Polymeren, dadurch gekennzeichnet, dass die Pellets eine Friabilität von höchstens 0,1 %, gemessen mit 200 g Pellets auf einer Siebmaschine mit 200 μm Sieb, einem Siebdurchmesser von 20 cm und 1 ,5 mm Schüttel-Amplitude bei einer Schüttel-Frequenz von 50 1/sec (50 Hertz) für 10 min in Gegenwart von sechs Gummiwürfeln mit 1 ,8 cm Kantenlänge, aufweisen und mit einem Polymerüberzug aus einem anionischen (Meth)acrylat- Copolymer überzogen sind mit der Maßgabe, dass die Pellets im Freisetzungstest nach USP in künstlichem Magensaft bei pH 1 ,2 nach 120 min nicht mehr als 10 %, bevorzugt nicht mehr als 7 %, besonders bevorzugt nicht mehr als 5 %, insbesondere nicht mehr als 3 % des enthaltenen Wirkstoffs freisetzen.
Wirkstofffreisetzung nach USP
Die Wirkstofffreisetzung kann nach USP, insbesondere USP 28-NF23, General Chapter <711 >, Dissolution, Apparatus 2 (Paddle), Method <724> „Delayed Release (Enteric Coated) Articles-General General Drug Release Standard", Method B (100 Upm, 37 0C) bestimmt werden: Die Pellets werden zunächst für 120 min in künstlichen Magensaft (USP) bei pH 1 ,2 auf Magensaftresistenz geprüft. Die Wirkstoffkonzentration im Testmedium kann abhängig vom Wirkstoff z. B. photometrisch bestimmt werden.
Mittlere Teilchengröße
Die mittlere Teilchengröße der Pellets kann im Bereich von 300 bis 1100, bevorzugt von 400 bis 1000 μm liegen.
Wirkstoffanteil
Der Wirkstoffanteil kann bezogen auf ein Pellet ohne Polymerüberzug kann 0,1 bis 70, bevorzugt 10 bis 60 Gew.-% betragen.
Friabilität
Die erfindungsgemäßen Pellets weisen eine außerordentlich hohe Abriebfestigkeit bzw. Friabilität auf. Die Abriebfestigkeit ist deutlicher höher als bei üblichen Arzneiformen bzw. Pellets und ist im Standardtest nach Ph. Eur. (5. Ausgabe, Abschnitt 2.9.7 kaum noch feststellbar oder keiner 0,001 %, also praktisch null. Der Standardtest ist daher zur Differenzierung der Friabilität zwischen Pellets des Standes der Technik und den erfindungsgemäßen Pellets kaum geeignet.
Die Friabilität der erfindungsgemäßen Pellets wird daher durch einen modifizierten Test mit gegenüber dem Standardtest verschärften Bedingungen beschrieben.
Die Pellets weisen eine Friabilität von höchstens 0,1 , bevorzugt höchstens 0,05 %, gemessen mit 200 g Pellets auf einer Siebmaschine mit 200 μm Sieb, einem Siebdurchmesser von 20 cm und 1 ,5 mm Schüttel-Amplitude bei einer Schüttelfrequenz von 50 1/s (50 Hertz) für 10 min in Gegenwart von sechs Gummiwürfeln mit 1 ,8 cm Kantenlänge (Gewicht der Gummiwürfel je 8,3 g), aufweisen.
Die Gummiwürfel haben eine Kantenlänge von 1 ,8 cm und ein Gewicht von je 8,3 g, die Dichte des Gummis beträgt demnach ca. 1 ,42 g/cm3. Bevorzugt sind Gummiwürfel aus Hartgummi.
Die Friabilität in % wird bestimmt, indem der aufgefangene Abrieb gewogen und ins Verhältnis zum Ausgangsgewicht der Pellets gesetzt wird.
Zur Messung der Friabilität kann z. B. eine Siebmaschine Retsch AS 200 Control verwendet werden. Siebmaschinen anderer Hersteller, mit denen sich die angegebenen Schüttelbedingungen herstellen lassen sind gleichermaßen geeignet.
Polymermatrix
Die erfindungsgemäßen Pellets enthalten eine pharmazeutisch wirksame Substanz, eingebettet in einer Polymermatrix aus einem oder mehreren Polymeren, bevorzugt (Meth)acrylat-Copolymeren.
Der Anteil der Polymermatrix bezogen auf ein Pellet ohne Polymerüberzug kann z. B. 20 bis 99,9 Gew.-%, bevorzugt 30 bis 60 Gew.-% betragen.
Die Polymermatrix kann pharmazeutisch übliche Hilfsstoffe, z. B. Bindemittel, enthalten.
(Meth)acrylat-Copolymere in der Polymermatrix
Die Polymermatrix kann kationische (Meth)acrylat-Copolymere, insbesondere (Meth)acrylat-Copolymere mit quaternären Ammoniumgruppen oder (Meth)acrylat- Copolymeren mit tertiären Ammoniumgruppen enthalten.
Die Polymermatrix kann ganz oder teilweise neutrale oder im Wesentlichen neutrale Methacrylat-Copolymere enthalten oder daraus bestehen.
(Meth)acrylatcopolymere mit quaternären Ammoniumgruppen (Typ EUDRAGIT® RS/RL)
Geeignete Polymere für die Polymermatrix sind z. B. (Meth)acrylat-Copolymere mit quaternären Ammoniumgruppen. (Meth)acrylat-Copolymere mit quaternären Ammoniumgruppen sind z. B. aus EP-A 181 515 oder aus DE-PS 1 617 751 bekannt. Es handelt sich um unabhängig vom pH-Wert wasserunlösliche bzw. in Wasser lediglich quellbare Polymerisate, die für Arzneimittelϋberzϋge geeignet sind. Als mögliches Herstellungsverfahren ist die Substanzpolymeriation in Gegenwart eines im Monomerengemisch gelösten radikalbildenden Initiators zu nennen. Ebenso kann das Polymerisat auch mittels Lösungs- oder Fällungspolymerisation hergestellt werden. Das Polymerisat kann auf diese Weise in Form eines feinen Pulvers erhalten werden, was bei der Subtanzpolymerisation durch Mahlen, bei Lösungs- und Fällungspolymerisation z. B. durch Sprühtrocknung erreichbar ist.
Die Polymermathx kann ein Polymerisat aus 98 bis 85 Gew.-% d- bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 2 bis 15 Gew.-% (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe oder eine Mischung mehrerer Polymer dieser Substanzklasse enthalten.
Bevorzugte d- bis C4-Al kylestern der Acryl- oder der Methacrylsäure sind Methylacrylat, Ethylacrylat, Butylacrylat, Butylmethacrylat und Methylmethacrylat.
Als (Meth)acrylat Monomer mit quaternären Ammoniumgruppen wird 2-Trimethylammoniumethylmethacrylat-Chlohd besonders bevorzugt.
Die Polymermathx kann ein Polymerisat aus 93 bis 88 Gew.-% Cr bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 7 bis 12 Gew.-% (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe enthalten (Typ Eudragit® RL).
Ein konkret geeignetes Copolymer enthält z. B. 60 Gew.-% Methylmethacrylat, 30 Gew.-% Ethylacrylat und 10 Gew.-%
2-Trimethylammoniumethlymethacrylat-Chlorid (EUDRAGIT® RL).
Die Polymermatrix kann ein Polymerisat aus 97 bis mehr als 93 Gew.-% d- bis C4- Alkylestern der Acryl- oder der Methacrylsäure und 3 bis weniger als 7 Gew.-% (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe enthalten (Typ Eudragit® RS).
Ein konkret geeignetes Copolymer enthält 65 Gew.-% Methylmethacrylat, 30 Gew.-% Ethylacrylat und 5 Gew.-% 2-Thmethylammoniumethylmethacrylat-Chlohd aufgebaut sein (EUDRAGIT® RS).
Insbesondere kommen auch Mischungen der genannten (Meth)acrylatcopolymere von Typ Eudragit® RL und Eudragit® RS in Frage, z. B. im Verhältnis von 20 zu 1 bis 1 zu 20, bevorzugt von 9 zu 1 bis 1 zu 9 Gewichtsteilen. Insbesondere bevorzugt sind Mischungen aus EUDRAGIT® RS und EUDRAGIT® RL.
Neutrale (Meth)acrylatcopolymere (Typ EUDRAGIT® NE oder Typ Eudragit® NM)
Weitere geeignete Polymere für die Polymermatrix sind z. B. neutrale oder im wesentlichen neutrale Methacrylat-Copolymere. Neutrale oder im wesentlichen neutrale Methacrylat-Copolymere können mindestens zu 95, insbesondere zu mindestens 98, bevorzugt zu mindestens 99, insbesondere zu mindestens 99, besonders bevorzugt zu 100 Gew.-% aus (Meth)acrylat-Monomeren mit neutralen Resten, insbesondere d- bis C4-Al kylresten bestehen.
Geeignete (Meth)acrylat-Monomere mit neutralen Resten sind z. B. Methylmethacrylat, Ethylmethacrylat, Butylmethacrylat, Methylacrylat, Ethylacrylat, Butylacrylat. Bevorzugt sind Methylmethacrylat, Ethylacrylat und Methylacrylat.
In geringen Anteilen, zu weniger als 5, bevorzugt höchstens 2, besonders bevorzugt höchstens 1 oder 0,05 bis 1 Gew.-% können Methacrylatmonomere mit anionischen Resten, z. B. Acrylsäure und/oder Methacrylsäure, enthalten sein.
Geeignet sind z. B. neutrale oder nahezu neutrale (Meth)acrylat Copolymere aus 20 bis 40 Gew.-% Ethylacrylat, 60 bis 80 Gew.-% Methylmethacrylat und 0 bis weniger als 5, bevorzugt 0 bis 2 oder 0,05 bis 1 Gew.-% (Typ EUDRAGIT® NE).
EUDRAGIT® NE und Eudragit® NM sind Copolymere aus 30 Gew.-% Ethylacrylat und 70 Gew.-% Methylmethacrylat.
Bevorzugt sind neutrale oder im Wesentlichen neutrale Methylacrylatcopolymere, die gemäß der WO 01/68767 als Dispersionen unter Verwendung von 1 - 10 Gew.-% eines nichtionischen Emulgators mit einem HLB-Wert von 15, 2 bis 17,3 hergestellt wurden. Letztere bieten den Vorteil, dass eine Phasenseparation unter Ausbildung von Kristallstrukturen durch den Emulgator unterbleibt (Eudragit® NM).
Gemäß EP 1 571 164 A2 können entsprechende, nahezu neutrale (Meth)acrylatcopolymere, mit geringen Anteilen, 0,05 bis 1 Gew.-% an monoolefinisch ungesättigten C3-C8-Carbonsäuren jedoch auch durch Emulsionspolymerisation in Gegenwart vergleichsweise geringer Mengen anionischer Emulgatoren, z. B. 0,001 bis 1 Gew.-% hergestellt werden.
(Meth)acrylat-Copolymere mit tertiären Aminogruppen
Weitere geeignete Polymere für die Polymermatrix sind z. B auch (Meth)acrylatcopolymere mit tertiären Aminogruppen. (Meth)acrylat-Copolymere mit tertiären Aminogruppen können sich teilweise oder ganz aus Alkylacrylaten und/oder Alkylmethacrylaten mit einer tertiären Aminogruppe im Alkylrest zusammensetzen. Geeignete (Meth)acrylatcopolymere sind bekannt z. B. aus EP 0 058 765 B1.
(Meth)acrylat-Copolymere mit tertären Aminogruppen sind nur im einem pH-Bereich in etwa unter pH 5 löslich. Sie werden daher oft zur Geschmacksisolierung von
Arzneiformen oder für im Magensaft rasch lösliche Arzneiformen verwendet. Im Rahmen der vorliegenden Erfindung ist die Polymermatrix jedoch mit einem anionischen (Meth)acrylat-Copolymeren überzogen, das sich erst im Darmsaft je nach Typ ab etwa 5,5 oder darüber auflöst. In diesem pH-Bereich sich (Meth)acrylat- Copolymere mit tertären Aminogruppen unlöslich bzw. lediglich quellend. Sie verhalten sich als Matrrixpolymere daher ähnlich retardierend, wie die oben beschriebenen (Meth)acrylat-Copolymere mit quaternären Ammoniumgruppen und stellen somit eine weitere Alternativ für die Formulierung erfindungsgemäßen Pellets und Arzneiformen dar.
Ein entsprechendes (Meth)acrylat-Copolymer kann sich z.B. aus 30 bis 80 Gew.-% radikalisch polymerisierten C1 - bis C4-Alkylestern der Acryl- oder der Methacrylsäure und 70 bis 20 Gew.-% (Meth)acrylat-Monomeren mit einer tertiären Aminogruppe im Alkylrest zusammensetzen (Typ EUDRAGIT® E).
Geeignete Monomere mit funktionellen tertiären Aminogruppen sind in US 4 705 695, Spalte 3, Zeile 64 bis Spalte 4, Zeile 13 aufgeführt. Insbesondere zu nennen sind Dimethylaminoethylacrylat, 2-Dimethylaminopropylacrylat, Dimethylaminopropylmethacrylat, Dimethylaminobenzylacrylat, Dimethylaminobenzylmethacrylat, (3-Dimethylamino-2,2-dimethly)propylacrylat, Dimethylamino-2,2-dimethly)propylmethacrylat, (3-Diethylamino-2,2- dimethly)propylacrylat und Diethylamino-2,2-dimethly)propylmethacrylat. Besonders bevorzugt ist Dimethylaminoethylmethacrylat.
Der Gehalt der Monomere mit tertiären Aminogruppen im Copolymeren kann vorteilhafterweise zwischen 20 und 70 Gew.-%, bevorzugt zwischen 40 und 60 Gew.- % liegen. Der Anteile der d- bis C4-Al kylestern der Acryl- oder Methacrylsäure beträgt 70 - 30 Gew.-%. Zu nennen sind Methylmethacrylat, Ethylmethacrylat, Butylmethacrylat, Methylacrylat, Ethylacrylat und Butylacrylat.
Ein geeignetes (Meth)acrylatcopolymer mit tertiären Aminogruppen kann z. B. aus 20 - 30 Gew.-% Methylmethacrylat, 20 - 30 Gew.-% Butylmethacrylat und 60 - 40 Gew.- % Dimethylaminoethylmethacrylat aufgebaut sein.
Ein konkret geeignetes handelsübliches (Meth)acrylatcopolymer mit tertiären Aminogruppen ist z. B. aus 25 Gew.-% Methylmethacrylat, 25 Gew.-% Butylmethacrylat und 50 Gew.-% Dimethylaminoethylmethacrylat aufgebaut (EUDRAGIT® E100 bzw. EUDRAGIT® E PO (Pulverform)). EUDRAGIT® E100 bzw. EUDRAGIT® E PO sind unterhalb von ca. pH 5,0 wasserlöslich und somit auch magensaftlöslich.
Polyvinylacetat/Polyvinylacetat-Copolymere, Ethyl- und Methylcellulose
Die Polymermatrix kann weiterhin auch ein Polyvinylacetat, ein Polyvinylacetat- Copolymer (z. B. Typ Kollicoat® SR 3OD oder Kollidon® SR), eine Ethylcellulose oder eine Methylcellulose enthalten.
Polymerüberzug
Die wirkstoffhaltigen Pellets sind mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen.
Der Polymerüberzug kann bezogen auf das Pelletgewicht 1 bis 15, 1 bis weniger als 14, bevorzugt 1 bis 13, besonders bevorzugt 1 bis weniger als 10, insbesondere 4 bis 9 Gew.-% betragen.
Der Polymerüberzug kann pharmazeutisch übliche Hilfsstoffe, z. B. Weichmacher, enthalten.
Der Polvmerüberzuq kann ein Polymerisat aus 25 bis 95, Gew.-% d- bis C4- Alkylestern der Acryl- oder der Methacrylsäure und 5 bis 75 Gew.-% (Meth)acrylat- Monomeren mit einer anionischen Gruppe enthalten.
Der Polymerüberzug kann ein Polymerisat aus 40 bis 60, Gew.-% Methacrylsäure und 60 bis 40 Gew.-% Methylmethacrylat oder 60 bis 40 Gew.-% Ethylacrylat enthalten.
Der Polymerüberzug kann ein Polymerisat aus 20 bis 40 Gew.-% Methacrylsäure und 80 bis 60 Gew.-% Methylmethacrylat enthalten.
Der Polymerüberzug kann ein Polymerisat aus 10 bis 30 Gew.-%, Methylmethacrylat, 50 bis 70 Gew.-% Methylacrylat und 5 bis 15 Gew.-% Methacrylsäure enthalten.
(Meth)acrylatcopolymere mit anionischen Gruppen (Typen EUDRAGIT® L, L100 55, S und FS)
Geeignete anionische (Meth)acrylatcopolymere sind Polymerisate aus 25 bis 95, Gew.-% Cr bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 5 bis 75 Gew.- % (Meth)acrylat-Monomeren mit einer anionischen Gruppe. Entsprechende Polymere sind je nach Gehalt an anionischen Gruppen und dem Charakter der weiteren Monomere bei pH-Werten oberhalb von pH 5,0 wasserlöslich und somit auch darmsaftlöslich.
In der Regel addieren sich die genannten Anteile zu 100 Gew.-%. Es können jedoch zusätzlich, ohne daß dies zu einer Beeinträchtigung oder Veränderung der wesentlichen Eigenschaften führt, geringe Mengen im Bereich von 0 bis 10, z. B. 1 bis 5 Gew.-% weiterer vinylisch copolymehsierbarer Monomere, wie z. B. Hydroxyethylmethacrylat oder Hydroxyethylacrylat enthalten sein.
Cr bis C4-Al kylestern der Acryl- oder Methacrylsäure sind insbesondere Methylmethacrylat, Ethylmethacrylat, Butylmethacrylat, Methylacrylat, Ethylacrylat und Butylacrylat.
Ein (Meth)acrylat-Monomer mit einer anionischen Gruppe kann z. B. Acrylsäure, bevorzugt jedoch Methacrylsäure sein.
Weiterhin geeignet sind anionische (Meth)acrylat Copolymere aus 40 bis 60, Gew.-% Methacrylsäure und 60 bis 40 Gew.-% Methylmethacrylat oder 60 bis 40 Gew.-% Ethylacrylat (Typen EUDRAGIT® L oder EUDRAGIT® L100-55).
EUDRAGIT® L ist ein Copolymer aus 50 Gew.-% Methylmethacrylat und 50 Gew.-% Methacrylsäure.
EUDRAGIT® L100-55 ist ein Copolymer aus 50 Gew.-% Ethylacrylat und 50 Gew.-% Methacrylsäure. EUDRAGIT® L 30D-55 ist eine Dispersion enthaltend 30 Gew.-% EUDRAGIT® L 100-55.
Ebenso geeignet sind anionische (Meth)acrylat Copolymere aus 20 bis 40 Gew.-% Methacrylsäure und 80 bis 60 Gew.-% Methylmethacrylat (Typ EUDRAGIT® S).
Besonders gut geeignet sind (Meth)acrylat Copolymere, bestehend aus 10 bis 30 Gew.-%, Methylmethacrylat, 50 bis 70 Gew.-% Methylacrylat und 5 bis 15 Gew.-% Methacrylsäure (Typ EUDRAGIT® FS).
EUDRAGIT® FS ist ein Copolymer aus 25 Gew.-%, Methylmethacrylat, 65 Gew.-% Methylacrylat und 10 Gew.-% Methacrylsäure. EUDRAGIT® FS 30 D ist eine Dispersion enthaltend 30 Gew.-% EUDRAGIT® FS.
Weiterhin geeignet für die Zwecke der Erfindung ist ein Copolymer (s. WO 2003/072087) welches sich aus
20 bis 34 Gew.-% Methacrylsäure und/oder Acrylsäure, 20 bis 69 Gew.-% Methylacrylat und 0 bis 40 Gew.-% Ethylacrylat und/oder gegebenenfalls 0 bis 10 Gew.-% weiteren vinylisch copolymehsierbarer Monomeren
zusammensetzt, mit der Maßgabe, daß die Glastemperatur des Copolymers nach ISO 11357-2, Punkt 3.3.3, höchstens 60 0C beträgt. Dieses (Meth)acrylatcopolymer ist wegen seiner guten Reißdehungseigenschaften insbesondere zum Verpressen von Pellets zu Tabletten geeignet.
Das Copolymer setzt sich insbesondere zusammen aus radikalisch polymerisierten Einheiten von
20 bis 34, bevorzugt 25 bis 33, besonders bevorzugt 28 bis 32 Gew.-% Methacrylsäure oder Acrylsäure, bevorzugt ist Methacrylsäure,
20 bis 69, bevorzugt 35 bis 65, besonders bevorzugt 35 bis 55 Gew.-% Methylacrylat und gegebenenfalls
0 bis 40, bevorzugt 5 bis 35, besonders bevorzugt 15 bis 35 Gew.-% Ethylacrylat zusammen, mit der Maßgabe, daß die Glastemperatur des Copolymers (Messung ohne Weichmacherzusatz bei einem Restmonomergehalt (REMO) von weniger als 100 ppm, Aufheizrate 10 °C/min, Stickstoffatmosphare) nach ISO 11357-2, Punkt 3.3.3 (Tmg), höchstens 60, bevorzugt 40 bis 60, besonders bevorzugt 45 bis 55 0C beträgt.
Das Copolymer besteht bevorzugt in wesentlichen bis ausschließlich aus den Monomeren Methacrylsäure, Methylacrylat und Ethylacrylat in den oben angegegebenen Mengenanteilen.
Es können jedoch zusätzlich, ohne daß dies zu einer Beeinträchtigung der wesentlichen Eigenschaften führt, geringe Mengen im Bereich von 0 bis 10, z. B. 1 bis 5 Gew.-% weiterer vinylisch copolymehsierbarer Monomere, wie z. B. Methylmethacrylat, Butylmethacrylat, Butylacrylat oder Hydroxyethylmethacrylat enthalten sein.
Unter Glastemperatur wird hier insbesondere die midpoint temperature Tmg nach ISO 11357-2, Punkt 3.3.3, verstanden. Die Messung erfolgt ohne Weichmacherzusatz, bei Restmonomergehalten (REMO) von weniger als 100 ppm, bei einer Aufheizrate von 10 °C/min und unter Stickstoffatmosphäre.
Die Copolymere werden in an sich bekannter Weise durch radikalische Substanz-, Lösungs-, Perl- oder Emulsionspolymerisation erhalten. Sie müssen vor der Verarbeitung durch geeignete Mahl-, Trocken- oder Sprühprozesse in den erfindungsgemäßen Teilchengrößenbereich gebracht werden. Dies kann durch einfaches Brechen extrudierter und abgekühlter Granulatstränge oder Heißabschlag erfolgen.
Insbesondere bei Mischung mit weiteren Pulvern oder Flüssigkeiten kann der Einsatz von Pulvern vorteilhaft sein. Geeignete Gerätschaften zur Herstellung der Pulver sind dem Fachmann geläufig, z. B. Luftstrahlmühlen, Stiftmühlen, Fächermühlen. Gegebenenfalls können entsprechende Siebungsschritte einbezogen werden. Eine geeignete Mühle für industrielle Großmengen ist zum Beispiel eine Gegenstrahlmühle (Multi Nr. 4200), die mit ca. 6 bar Überdruck betrieben wird.
Weiterhin geeignet für die Zwecke der Erfindung sind Copolymere (s. WO 2004/096185) aus
20 bis 33 Gew.-% Methacrylsäure und/oder Acrylsäure,
5 bis 30 Gew.-% Methylacrylat und
20 bis 40 Gew.-% Ethylacrylat und größer 10 bis 30 Gew.-% Butylmethacrylat und gegebenenfalls
0 bis 10 Gew.-% weiteren vinylisch copolymehsierbarer Monomeren, wobei sich die Anteile der Monomeren zu 100 Gew.-% addieren,
zusammensetzt, mit der Maßgabe, dass die Glastemperatur des Copolymers (glass transition temperature) nach ISO 11357-2, Punkt 3.3.3 (midpoint temperature Tmg), 55 bis 70 0C beträgt. Copolymere dieses Typs sind wegen seiner guten mechanischen Eigenschaften insbesondere zum Verpressen von Pellets zu Tabletten geeignet.
Das oben genannte Copolymer setzt sich insbesondere zusammen aus radikalisch polymerisierten Einheiten von
20 bis 33, bevorzugt 25 bis 32, besonders bevorzugt 28 bis 31 Gew.-% Methacrylsäure oder Acrylsäure, bevorzugt ist Methacrylsäure,
5 bis 30, bevorzugt 10 bis 28, besonders bevorzugt 15 bis 25 Gew.-% Methylacrylat, 20 bis 40,
bevorzugt 25 bis 35, besonders bevorzugt 28 bis 32 Gew.-% Ethylacrylat,
sowie größer 10 bis 30, bevorzugt 15 bis 25, besonders bevorzugt 18 bis 22 Gew.-% Butylmethacrylat
zusammen, wobei die Monomerzusammensetzung so gewählt wird, dass die Glastemperatur des Copolymers 55 bis 70 0C, bevorzugt 59 bis 66, besonders bevorzugt 60 bis 65 0C beträgt. Unter Glastemperatur wird hier insbesondere die midpoint temperature Tmg nach ISO 11357-2, Punkt 3.3.3, verstanden. Die Messung erfolgt ohne Weichmacherzusatz, bei Restmonomergehalten (REMO) von weniger als 100 ppm, bei einer Aufheizrate von 10 °C/min und unter Stickstoffatmosphäre.
Das Copolymer besteht bevorzugt in wesentlichen bis ausschließlich, zu 90, 95 oder 99 bis 100 Gew.-%, aus den Monomeren Methacrylsäure, Methylacrylat, Ethylacrylat und Butylmethacrylat in den oben angegebenen Mengenbereichen.
Es können jedoch zusätzlich, ohne daß dies zu einer Beeinträchtigung der wesentlichen Eigenschaften führen muß, geringe Mengen im Bereich von 0 bis 10, z. B. 1 bis 5 Gew.-% weiterer vinylisch copolymehsierbarer Monomere, wie z. B. Methylmethacrylat, Butylacrylat, Hydroxyethylmethacrylat, Vinylpyrrolidon, Vinylmalonsäure, Styrol, Vinylalkohol, Vinylacetat und/oder deren Derivate enthalten sein.
Die Copolymere werden in an sich bekannter Weise durch radikalische Substanz-, Lösungs-, Perl- oder Emulsionspolymerisation erhalten. Sie müssen vor der Verarbeitung durch geeignete Mahl-, Trocken- oder Sprühprozesse in den erfindungsgemäßen Teilchengrößenbereich gebracht werden. Dies kann durch einfaches Brechen extrudierter und abgekühlter Granulatstränge oder Heißabschlag erfolgen.
Insbesondere bei Mischung mit weiteren Pulvern oder Flüssigkeiten kann der Einsatz von Pulvern vorteilhaft sein. Geeignete Gerätschaften zur Herstellung der Pulver sind
dem Fachmann geläufig, z. B. Luftstrahlmühlen, Stiftmühlen, Fächermühlen. Gegebenenfalls können entsprechende Siebungsschritte einbezogen werden. Eine geeignete Mühle für industrielle Großmengen ist zum Beispiel eine Gegenstrahlmühle (Multi Nr. 4200), die mit ca. 6 bar Überdruck betrieben wird.
Die Herstellung der anionischen (Meth)acrylatcopolymere mit Anteilen anionischer Monomere von über 5 Gew.-% im Polymerisat kann in an sich bekannter Weise durch radikalische Polymerisation der Monomeren erfolgen (siehe z. B. EP 0 704 207 A2, EP 0 704 208 A2, WO 2003/072087, WO 2004/096185). Die Copolymere sind in an sich bekannter Weise durch radikalische Emulsionspolymerisation in wäßriger Phase in Gegenwart von vorzugsweise anionischen Emulgatoren herstellbar, beispielsweise nach dem in DE-C 2 135 073 beschriebenen Verfahren.
Die genannten Copolymehsate können nach gängigen Verfahren der radikalischen Polymerisation kontinuierlich oder diskontinuierlich (Batch-Verfahren) in Gegenwart radikalbildender Initiatoren und gegebenenfalls Reglern zur Einstellung des Molekulargewicht in Substanz, in Lösung, durch Perlpolymerisation oder in Emulsion hergestellt werden. Das mittlere Molekulargewicht Mw (Gewichtsmittel, bestimmt z. B. durch Messung der Lösungsviskosität) kann z. B. im Bereich von 80.000 bis 1.000.000 (g/mol) liegen. Bevorzugt ist die Emulsionspolymerisation in wäßriger Phase in Gegenwart wassergelöster Initiatoren und (vorzugsweise anionischer) Emulgatoren. Im Falle der Substanzpolymerisation kann das Copolymer in fester Form durch Brechen, Extrusion, Granulieren oder Heißabschlag erhalten werden.
Zur Einstellung spezieller Freisetzungsprofile bzw. Freisetzungsorte können auch Mischungen der genannten anionischen (Meth)acrylat-Copolymere zum Einsatz kommen. Gegebenenfalls können auch Mischungen der anionischen (Meth)acrylat- Copolymere mit nicht mehr als 50, bevorzugt 10 bis 30 Gew.-% der bereits genannten, neutralen oder im wesentlichen neutralen Methacrylat-Copolymeren vorliegen. Bevorzugt enthalten die Überzüge jedoch höchstens 10 Gew.-%, bevorzugt 0 - 5 Gew.-%, insbesondere keine neutralen oder im wesentlichen neutralen Methacrylat-Copolymeren .
Wirkstoffe
Die enthaltene pharmazeutisch wirksame Substanz kann ein pharmazeutischer Wirkstoff oder ein Nahrungsergänzungsmittel sein.

Verfahren zur Herstellung der erfindungsgemäßer Pellets
Die Erfindung betrifft ein Verfahren zur Herstellung von wirkstoffhaltigen Pellets mit Polymerüberzug mittels Schmelzverarbeitung, wobei die pharmazeutisch wirksame Substanz, das oder die Polymere für die Polymermatrix vermischt werden und für mindestens 10 sec, bevorzugt für mindestens 20 sec eine Temperatur von mindestens 5 0C oberhalb der Glastemperatur des Polymeren oder im Falle einer Polymermischung bezogen auf das Polymer mit der höchsten Glasübergangstemperatur einwirkt, die Mischung in einem Extruder bevorzugt einem Doppelschneckenextruder, extrudiert, ausgetragen und durch Heißabschlag mit anschließender Ausrundung zu Pellets mit einer mittleren Teilchengröße im Bereich von 300 bis 1100, bevorzugt von 400 bis 1000 μm μm ausgerundet wird, und die Pellets mittels Spühauftrag mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen werden.
Der Polymermatrix können bei der Herstellung der Pellets pharmazeutisch übliche Hilfsstoffe zugesetzt werden.
Auf die zu verarbeitende Mischung soll als Mindestanforderung für mindestens 10, bevorzugt für mindestens 20 sec eine Temperatur von mindestens 5, bevorzugt von mindestens 10 0C oberhalb der Glastemperatur des Polymeren mit der höchsten Glasübergangstemperatur einwirken. Dies bewirkt die Ausbildung einer einheitlichen Schmelzephase.
Unter Glastemperatur wird hier insbesondere die midpoint temperature Tmg nach ISO 11357-2, Punkt 3.3.3, verstanden. Die Messung erfolgt ohne Weichmacherzusatz, bei Restmonomergehalten (REMO) von weniger als 100 ppm, bei einer Aufheizrate von 10 °C/min und unter Stickstoffatmosphäre. Die Glastemperatur von Eudragit® RS beträgt etwa 50 0C.
Unter Praxisbedingungen werden in vielen Fällen entsprechende Mindesttemperaturen meist leicht erreicht oder übertroffen und auch über längere Zeiträume gehalten, ohne dass die für die pharmazeutisch wirksame Substanz oder die enthaltenen Polymere kritisch wäre. Typische Verarbeitungstemperaturen im Extruder können je nach Polymerzusammensetzung der Mischung z. B. 50 bis 200, bevorzugt 100 bis 180 0C betragen.
Je nach Mischungsbestandteilen, insbesondere den enthaltenen pharmazeutisch wirksame Substanzen, ist jedoch darauf zu achten, dass Temperaturen und Verweildauern so zu bemessen sind, dass thermische Schädigungen oder Beeinträchtigungen möglichst vermieden werden. In der Regel wird man versuchen die Verarbeitungstemperaturen und die Verweildauern eher möglichst gering einzustellen. Ein Fachmann kann dies in Kenntnis der Erfindung leicht auf den Einzelfall abstellen und entsprechend verfahren.
Aufbringen des Polymerüberzugs
Die Polymerüberzüge auf die wirkstoffhaltige Pellets können z. B. durch Sprühauftrag bevorzugt in Wirbelschichtgeräten aufgebracht werden. Der Polymerüberzug wird üblicherweise mit Weichmachern und Trennmitteln nach geeigneten Verfahren gemischt. Hierbei kann das Polymer als Lösung oder Suspension vorliegen. Die Hilfsstoffe können ebenfalls gelöst oder suspendiert sein. Organische oder wässrige Löse- oder Dispergiermittel können verwendet werden. Zur Stabilisierung der Dispersion können zusätzlich Stabilisatoren verwendet werden (Beispiel: Polysorbat 80 oder andere geeignete Emulgatoren bzw. Stabilisatoren).
Beispiele für Trennmittel sind Glycerolmonostearat oder andere geeignete Fettsäurederivate, Kieselsäurederivate oder Talkum. Beispiele für Weichmacher sind Propylenglykol, Phthalate, Polyethylenglykole, Sebacate oder Citrate, sowie andere in der Literatur erwähnte Substanzen.
Multipartikuläre Arzneiform
Die erfindungsgemäßen Pellets können in einer multipartikulären Arzneiform, insbesondere in Tabletten, Minitabletten, Kapseln, Sachets oder Trockensäften enthalten sein.
Eine multipartikulare Arzneiform kann als Einzeldosis, z. B. eine Kapsel, zweckmäßigerweise z. B. 20 bis 1000 einzelne Pellets enthalten. Die enthaltenen Pellets können untereinander gleich sein und aus einer einheitliche Pelletpopulation stammen. Es können auch mehrere, von einander verschiedene Pelletpopulationen mit unterschiedlichen Formulierungen nebeneinander in einer multipartikulären Arzneiform enthalten sein.
Die erfindungsgemäßen Pellets können somit zur Herstellung von Arzneiformen, insbesondere multipartikulären Arzneiform verwendet werden.
Hilfsstoffe
Die Polymermatrix und/der Polymerüberzug pharmazeutisch übliche Hilfsstoffe enthält.
Grundsätzlich müssen natürlich alle eingesetzten Hilfsstoffe toxikologisch unbedenklich und für den Verwendungszweck, insbesondere in Nahrungsergänzungsmitteln oder Arzneiformen geeignet sein und ohne Risiko für den Konsumenten oder Patienten zu verwenden sein.
Einsatzmengen und Verwendung der üblichen Zuschlagstoffe in Arzneimittelüberzügen oder Beschichtungen sind dem Fachmann geläufig. Übliche Zuschlagstoffe können z. B. Trennmittel, Pigmente, Stabilisatoren, Antioxidantien, Porenbildner, Penetrationsförderer, Glanzmittel, Aromastoffe oder Geschmacksmittel sein. Sie dienen als Verarbeitungshilfsmittel und sollen ein sicheres und reproduzierbares Herstellungsverfahren sowie gute Langzeitlagerstabilität gewährleisten oder sie erreichen in der Arzneiform zusätzliche vorteilhafte
Eigenschaften. Sie werden den Polymerzubereitungen vor der Verarbeitung zugesetzt und können die Permeabilität der Überzüge beeinflussen, was ggf. als zusätzlicher Steuerparameter genutzt werden kann.
• Trennmittel:
Trennmittel besitzen in der Regel lipophile Eigenschaften und werden in der Regel den Sprühsuspensionen zugesetzt. Sie verhindern eine Agglomeration der Kerne während der Befilmung. Bevorzugt werden Talkum, Mg- oder Ca - Stearat, gemahlene Kieselsäure, Kaolin oder nicht ionische Emulgatoren mit einem HLB - Wert zwischen 3 und 8 eingesetzt. Übliche Einsatzmengen für Trennmittel liegen zwischen 0,5 bis 100 Gew.-% bezogen auf die Summe aus Wirkstoff, wasserlöslichem (Meth)acrylatcopolymer und wasserunlöslichem Polymer.
• Pigmente:
Die zu verwendenden Pigmente nicht toxisch und für pharmazeutische Zwecke geeignet. Siehe dazu z. Geeigente Pigmente sind z. B. Aluminiumoxidpigmente oder Gelborange , Cochenillerotlack, Farbpigmente auf Basis von Aluminiumoxid bzw Azofarbstoffen, Sulfonsäurefarbstoffe, Gelborange S (E110, Cl. 15985, FD&C Yellow 6), Indigocarmin (E132, Cl. 73015, FD&C Blue 2), Tartrazin (E 102, Cl. 19140, FD&C Yellow 5), Ponceau 4R (E 125, Cl. 16255, FD&C Cochineal Red A), Chinolingleb (E 104, Cl. 47005, FD&C Yellow 10), Erythrosin (E127, Cl. 45430, FD&C Red 3), Azorubin (E 122, Cl. 14720, FD&C Carmoisine), Amaranth (E 123, C. I. 16185, FD&C Red 2), Brilliantsäuregrün (E 142, Cl. 44090, FD&C Green S).

• Weichmacher
Weitere Zuschlagstoffe können auch Weichmacher sein. Übliche Mengen liegen zwischen 0 und 50, bevorzugt 5 bis 20 Gew.-%.
Weichmacher können je nach Typ (lipophil oder hydrophil) und zugesetzter Menge die Funktionalität der Polymerschicht beeinflussen. Weichmacher erreichen durch physikalische Wechselwirkung mit dem Polymeren eine Absenkung der Glasübergangstemperatur und fördern in Abhängigkeit von der zugesetzten Menge die Verfilmung. Geeignete Stoffe haben in der Regel ein Molekulargewicht zwischen 100 und 20.000 und enthalten eine oder mehrere hydrophile Gruppen im Molekül, z. B. Hydroxyl-, Ester- oder Aminogruppen.
Beispiele geeigneter Weichmacher sind Citronensäurealkylester, Glycehnester, Phthalsäurealkylester, Sebacinsäurealkylester, Succroseester, Sorbitanester, Diethylsebacat, Dibutylsebacat und Polyethylenglykole 200 bis 12.000. Bevorzugte Weichmacher sind Thethylcitrat (TEC), Acetyltriethylcitrat (ATEC) und Dibutylsebacat (DBS). Weiterhin zu nennen sind in der Regel bei Raumtemperatur flüssige Ester wie Citrate, Phthalate, Sebacate oder Rizinusöl. Bevorzugt werden Zitronensäure- und Sebacinsäureester verwendet.
Die Zugabe der Weichmacher zur Formulierung kann in bekannter Weise, direkt, in wässriger Lösung oder nach thermischer Vorbehandlung einer Mischung vorgenommen werden. Auch können Mischungen von Weichmachern eingesetzt werden.
Überziehen der Pellets
Die Filmüberzüge auf die wirkstoffhaltige Pellets werden üblicherweise in Wirbelschichtgeräten aufgebracht. Der Polymerüberzug wird üblicherweise mit Weichmachern und Trennmitteln nach geeigneten Verfahren gemischt. Hierbei kann das Polymer als Lösung oder Suspension vorliegen. Die Hilfsstoffe können ebenfalls gelöst oder suspendiert sein. Organische oder wässrige Löse- oder Dispergiermittel können verwendet werden. Zur Stabilisierung der Dispersion können zusätzlich Stabilisatoren verwendet werden (Beispiel: Polysorbat 80 oder andere geeignete Emulgatoren bzw. Stabilisatoren).
Beispiele für Trennmittel sind Glycerolmonostearat oder andere geeignete Fettsäurederivate, Kieselsäurederivate oder Talkum. Beispiele für Weichmacher sind Propylenglykol, Phthalate, Polyethylenglykole, Sebacate oder Citrate, sowie andere in der Literatur erwähnte Substanzen.
BEISPIELE
Alle Beispiele wurden auf einem gleichläufigen 18 mm Doppelschneckenextruder mit einer funktionellen Länge des Verfahrensteils von 4OD extrudiert. Die Extrusionstemperatur lag im Einzugsbereich bei 10 0C - 100 0C, in den folgenden Zylindern des Extruders wurde die Temperatur auf bis zu 160 0C erhöht. Die Schmelze wurde bei 160 0C ausgetragen und in einem luftgekühlten Heißabschlagprozeß zu Pellets geschnitten. Für den Heißabschlagprozeß wird die Schmelze am Ende des Extrusionsverfahrensteils in einen kegelförmigen Schmelzekanal geführt, welcher am Ende mehrere Austrittsöffnungen am Fuß in Form eines Rings hat. Über diesen Ring rotiert ein oder mehrere Messer, welche(s) die Schmelze in heißem Zustand abschneidet. Die Pellet werden in einem Luftstrom gekühlt und abtransportiert. Die Ausrundung der Pellets erfolgt durch die in der Schmelze noch vorhandene Oberflächenspannung und nicht oder nur in einem sehr geringen Maße während des Transports der Pellets direkt nach dem Abschlagprozeß. Der Wirkstoff und die Polymere wurden dem Extruder über gravimethsche Dosierungen zugeführt.
Die Befilmung der Pellets erfolgte in einem Wirbelschichtgerät ausgerüstet als Bottomspray. Die Ansatzgröße betrug 100 g Pellets.
Das Beispiel V1 ist ein Vergleichsbeispiel
Die Pellets des Beispiels V1 enthalten das wasserunlösliche Polymer EUDRAGIT®
RL sowie EUDRAGIT® RS. Die Pellets wurden mit 1 Gew.-%, bezogen auf
Polymertrockensubstanz, einer Suspension, enthaltend das magensaftresistente
Polymer EUDRAGIT® L 30 D-55 befilmt. Die Formulierung zeigt keine
Magensaftresistenz, da nach 120 min bei pH 1 ,2 bereits 16,8 % des anfangs enthaltenen Wirkstoffs freigesetzt sind.
Die Sprühsuspension zur Befilmung wurde als 30 %ige Suspension hergestellt, enthaltend EUDRAGIT® L 30 D-55, 10 % Triethylcitrat bezogen auf 100 %
Polymerfeststoff, 3 % Glycerolmonostearat bezogen auf 100 % Polymerfeststoff sowie 40 % Polysorbat 80 bezogen auf 100 % Glycerolmonostearat.
Die Beispiele 2 bis 5 sind erfindungsgemäße Beispiele:
Die Pellets der Beispiele 2 bis 5 enthalten das wasserunlösliche Polymer EUDRAGIT® RL sowie EUDRAGIT® RS. Die Pellets wurden mit 2% bis 6 % (Gew.- %) einer Suspension enthaltend das magensaftresistente Polymer EUDRAGIT® L 30 D-55 befilmt. Die Formulierung zeigen Magensaftresistenz, da nach 120 min bei pH 1 ,2 weniger als 10 % des anfangs enthaltenen Wirkstoffs freigesetzt sind. Die Sprühsuspension zur Befilmung wurde als 30 %ige Suspension hergestellt, enthaltend EUDRAGIT® L 30 D-55, 10 % Triethylcitrat bezogen auf 100 % Polymerfeststoff, 3 % Glycerolmonostearat bezogen auf 100 % Polymerfeststoff sowie 40 % Polysorbat 80 bezogen auf 100 % Glycerolmonostearat.
Claims
1. Wirkstoffhaltige Pellets mit Polymerüberzug und einer mittleren Teilchengröße im Bereich von 300 bis 1100 μm, enthaltend eine pharmazeutisch wirksame Substanz, eingebettet im einer Polymermatrix aus einem oder mehreren Polymeren, dadurch gekennzeichnet, dass die Pellets eine Friabilität von höchstens 0,1 %, gemessen mit 200 g Pellets auf einer Siebmaschine mit 200 μm Sieb, einem Siebdurchmesser von 20 cm und 1 ,5 mm Schüttel-Amplitude bei einer Schüttel-Frequenz von 50 1/sec für 10 min in Gegenwart von sechs Gummiwürfeln mit 1 ,8 cm Kantenlänge, aufweisen und mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen sind mit der Maßgabe, dass die Pellets im Freisetzungstest nach USP in künstlichem Magensaft bei pH 1 ,2 nach 120 min nicht mehr als 10 % des enthaltenen Wirkstoffs freisetzen.
2. Pellets nach Anspruch 1 , dadurch gekennzeichnet, dass der Polymerüberzug 1 bis15 Gew.-% bezogen auf das Pelletgewicht beträgt.
3. Pellets nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass der Wirkstoffanteil bezogen auf ein Pellet ohne Polymerüberzug 0,1 bis 70 Gew.- % beträgt.
4. Pellets nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Anteil der Polymermatrix bezogen auf ein Pellet ohne Polymerüberzug 20 bis 99,9 Gew.-% beträgt.
5. Pellets nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Polymermatrix und/der Polymerüberzug pharmazeutisch übliche Hilfsstoffe enthält.
6. Pellets nach einem oder mehreren der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass die Polymermatrix ein Polymerisat aus 98 bis 85 Gew.- % Cr bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 2 bis 15 Gew.-% (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe oder eine Mischung mehrerer Polymer dieser Substanzklasse enthält.
7. Pellets nach Anspruch 5, dadurch gekennzeichnet, dass die Polymermatrix ein Polymerisat aus 93 bis 88 Gew.-% d- bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 7 bis 12 Gew.-% (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe enthält.
8. Pellets nach Anspruch 5, dadurch gekennzeichnet, dass das die Polymermatrix ein Polymerisat aus 97 bis mehr als 93 Gew.-% Cr bis C4- Alkylestern der Acryl- oder der Methacrylsäure und 3 bis weniger als 7 Gew.- % (Meth)acrylat Monomeren mit einer quaternären Ammoniumgruppe enthält
9. Pellets nach einem oder mehreren der Ansprüche 6 bis 8, dadurch gekennzeichnet, dass in der Polymermatrix eine Mischung der Polymere nach Anspruch 6 und 7 im Verhältnis 20 zu 1 bis 1 zu 20 vorliegt.
10. Pellets nach einem oder mehreren der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass die Polymermatrix ein Copolymer aus 20 bis 40 Gew.-% Ethylacrylat und 60 bis 80 Gew.-% Methylmethacrylat und 0 bis weniger als 5 Gew.-% Acrylsäure und/oder Methacrylsäure enthält.
11. Pellets nach einem oder mehreren der Ansprüche 1 bis 10, dadurch gekennzeichnet, dass die Polymermatrix ein Polymerisat aus 30 bis 80 Gew.- % Cr bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 70 bis 20 Gew.-% (Meth)acrylat-Monomeren mit einer tertiären Aminogruppe im Alkylrest enthält.
12. Pellets nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Polymermatrix ein Polyvinylacetat, ein Polyvinylacetat-Copolymer, eine Ethylcellulose oder eine Methylcellulose enthält..
13. Pellets nach einem oder mehreren der Ansprüche 1 bis 12, dadurch gekennzeichnet, dass der Polymerüberzug ein Polymerisat aus 25 bis 95, Gew.-% Cr bis C4-Al kylestern der Acryl- oder der Methacrylsäure und 5 bis 75 Gew.-% (Meth)acrylat-Monomeren mit einer anionischen Gruppe enthält.
14. Pellets nach Anspruch 13, dadurch gekennzeichnet, dass der Polymerüberzug ein Polymerisat aus 40 bis 60, Gew.-% Methacrylsäure und 60 bis 40 Gew.-% Methylmethacrylat oder 60 bis 40 Gew.-% Ethylacrylat enthält.
15. Pellets nach Anspruch 13, dadurch gekennzeichnet, dass der Polymerüberzug ein Polymerisat aus 20 bis 40 Gew.-% Methacrylsäure und 80 bis 60 Gew.-% Methylmethacrylat enthält.
16. Pellets nach Anspruch 13, dadurch gekennzeichnet, dass der Polymerüberzug ein Polymerisat aus 10 bis 30 Gew.-%, Methylmethacrylat, 50 bis 70 Gew.-% Methylacrylat und 5 bis 15 Gew.-% Methacrylsäure enthält.
17. Pellets nach einem oder mehreren der Ansprüche 1 bis 16, dadurch gekennzeichnet, dass die enthaltene pharmazeutisch wirksame Substanz ein pharmazeutischer Wirkstoff oder ein Nahrungsergänzungsmittel ist.

19. Pellets nach einem oder mehreren der Ansprüche 1 bis 18, dadurch gekennzeichnet, dass sie in einer multipartikulären Arzneiform, insbesondere in Tabletten, Minitabletten, Kapseln, Sachets oder Trockensäften enthalten sind.
20. Verfahren zur Herstellung von wirkstoffhaltigen Pellets mit Polymerüberzug nach einem oder mehreren der Ansprüche 1 bis 19 mittels Schmelzverarbeitung, dadurch gekennzeichnet, dass die pharmazeutisch wirksame Substanz, das oder die Polymere für die Polymermatrix vermischt werden und für mindestens 10 sec eine Temperatur von mindestens 5 0C oberhalb der Glastemperatur des Polymeren oder im Falle einer Polymermischung bezogen auf das Polymer mit der höchsten Glasübergangstemperatur einwirkt, die Mischung in einem Extruder extrudiert, ausgetragen und durch Heißabschlag mit anschließender Ausrundung zu Pellets mit einer mittleren Teilchengröße im Bereich von 300 bis 1100 μm ausgerundet wird, und die Pellets mittels Spühauftrag mit einem Polymerüberzug aus einem anionischen (Meth)acrylat-Copolymer überzogen werden.
21. Verfahren nach Anspruch 20, dadurch gekennzeichnet, dass der Polymermatrix und/oder dem Polymerüberzug bei der Herstellung der Pellets pharmazeutisch übliche Hilfsstoffe zugesetzt werden.
22. Verfahren nach Anspruch 20 oder 21 dadurch gekennzeichnet, dass die Verarbeitungstemperatur im Extruder 50 bis 200 0C beträgt.
23. Multipartikuläre Arzneiform, enthaltend ein oder mehrere Pellets nach einem oder mehreren der Ansprüche 1 bis 19.
24. Verwendung von Pellets nach einem oder mehreren der Ansprüche 1 bis 19 zur Herstellung einer multipartikulären Arzneiform.
