Abstract

Die Erfindung betrifft ein Verfahren und ein Kit zur Kalibrierung eines Photolumineszenzmesssystems, insbesondere eines Fluoreszenzmesssystems. Das Kit umfasst eine Mehrzahl von Fluoreszenzstandards i und deren korrigierte und zertifizierte Fluoreszenzspektren Ii(λ), wobei die Fluoreszenzstandards i so gewählt sind, dass ihre spektral korrigierten Fluoreszenzspektren Ii(λ) einen breiten Spektralbereich mit hoher Intensität abdecken. Die Standards zeichnen sich zudem durch hohe Halbwertsbreiten FWHMi ihrer Banden von zumindest 1400 cm-1 aus. Gemäß dem erfindungsgemäßen Verfahren werden Teilkorrekturfunktionen Fi(λ) durch Quotientenbildung aus gemessenen Fluoreszenzspektren Ji(λ) und den entsprechenden korrigierten Fluoreszenzspektren Ii(λ) gebildet und zu einer Gesamtkorrekturfunktion F(λ) für einen breiten Spektralbereich miteinander verknüpft. Hierfür erfolgt eine Berechnung von Verknüpfungsfaktoren αi durch statistische Mittelung aufeinander folgender Teilkorrekturfunktionen Fi(λ) lediglich über einen vorgegebenen, begrenzten Überlappungsbereich λi/i+1 ±ΔλOL um die gegenseitige Überschneidungswellenlänge λi/i+1.
