Abstract
La présente invention concerne de nouveaux dérivés de chalcone de formule (I) : dans laquelle X, Y, Z, W, R1, R2, R3 R4 et R5 sont tels que définis, lesdits dérivés présentant une activité antimitotique, ainsi que les compositions pharmaceutiques contenant de tels composés et leur utilisation pour la fabrication de médicaments.
Description
NOUVEAUX DERIVES DE CHALCONE A ACTIVITE ANTIMITOTIQUE
La présente invention concerne de nouveaux dérivés de chalcone présentant une activité antimitotique, ainsi que les compositions pharmaceutiques contenant de tels composés et leur utilisation pour la fabrication de médicaments.
De nombreux travaux ont déjà été réalisés sur des dérivés de chalcone. A titre illustratif, on peut citer la demande US 6462075 qui décrit des dérivés de chalcone présentant une activité d'inhibition de l'angiogénèse. Ces composés sont présentés comme pouvant être utilisés comme agents anti- tumoraux, anti-cancéreux, pour traiter les maladies angiogénique de la peau et les maladies inflammatoires chroniques.
Selon la présente invention, les inventeurs ont développé une nouvelle série de chalcones présentant des propriétés dose-réponse en cytométrie en flux sur plusieurs lignées tumorales et en cytotoxicité améliorées par rapport aux composés de l'art antérieur de structure les plus proches.
La présente invention a, en particulier, pour objet des nouveaux dérivés de chalcone de formule (I) :
- X représente un atome d'hydrogène ou un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl, -NH2, -NHCOR3 avec Ra qui est un groupe alkyle linéaire ou ramifié de 1 à 5 atomes de carbone, un groupe aryl qui peut être mono ou polysubstituté par un substituant choisi parmi les atomes d'halogène, les groupes OH, OMe et
-NRbRc avec Rb et R0 qui représentent indépendamment l'un de l'autre une chaîne alkyle linéaire ou ramifiée de 1 à 6 atomes de carbone,
- Y représente un atome d'hydrogène ou un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl, étant entendu qu'un au moins des groupes X et Y est différent de l'hydrogène, - Z représente un atome d'hydrogène ou un groupe méthyl, éthyl, propyl, isopropyl, benzyl,
- W représente un atome d'hydrogène, ou un groupe -OH, -O-méthyl, -O-éthyl, ou -O-benzyl,
- Rl, R2, R3, R4 et R5 représentent, indépendamment les uns des autres, un atome d'hydrogène ou d'halogène ou un groupe -OH, méthyl,
-O-méthyl, éthyl, -O-éthyl, propyl, -O-propyl, benzyl, -O-benzyl, -NH2, -NHCH3, -N(CH3)2, étant entendu qu'au moins deux substituants Rl, R2, R3, R4 et R5 sont différents de H, et que lorsque X ou Y représente un atome d'hydrogène, au moins trois substituants Rl, R2, R3, R4 et R5 sont différents de H, ainsi que leurs hydrates, solvats ou sels pharmaceutiquement acceptables, à l'exception du 2'-hydroxy-4',6',2,4,5-pentaméthoxychalcone, et du 2',4',6',2,4,5- pentaméthoxychalcone.
Dans tous les composés de formule (I), la configuration autour de la double liaison α,β est trans comme indiqué sur la formule développée (I).
Dans la définition ci-dessus, par halogène, on entend un atome de chlore, brome, iode ou fluor.
Par alkyle, on entend une chaîne hydrocarbonée saturée.
Par aryl, on entend un groupe phényl, naphtyl ou cinnamyl. Selon des aspects préférés de l'invention, les composés de formule (I) présentent l'une quelconque des caractéristiques ci-après ou une combinaison de plusieurs de ces caractéristiques, lorsqu'elles ne s'excluent pas l'une l'autre :
- au moins trois des substituants Rl, R2, R3, R4 et R5 sont différents de l'hydrogène,
- X représente un atome d'hydrogène ou un groupe -OMe, -OEt, -NH2, le groupe OMe étant préféré, étant entendu que, quand X représente un atome d'hydrogène, Y représente un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl,
- Y représente un atome d'hydrogène ou un groupe -OMe, -OEt, le groupe OMe étant préféré, étant entendu que, quand Y représente un atome d'hydrogène, X représente un groupe -OH, -O-méthylf -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl,
- Z représente un atome d'hydrogène ou un groupe méthyl ou éthyl,
- W représente un groupe -O-méthyl ou de préférence un atome d'hydrogène,
- Rl, R2, R3, R4 et R5 représentent, indépendamment les uns des autres, un atome d'hydrogène ou de fluor, un groupe méthyl, -O-méthyl, -O-éthyl, étant entendu qu'au moins deux substituants Rl, R2, R3, R4 et R5 sont différents de H, et que lorsque X ou Y représente un atome d'hydrogène, au moins trois substituants Rl, R2, R3, R4 et R5 sont différents de H,

L'étude de relation structure-activité indique que la nature des substituants des groupes phényl et leur position sont déterminantes pour l'activité antimitotique.
Sur le noyau A de la chalcone, un groupe méthoxy semble être le plus avantageux parmi tous les substituants étudiés et les positions 2,4 et 6 sont les plus importantes pour la substitution.
En conclusion, une di ou triméthoxylation sur les carbones 2,4,6 du noyau A et 2',4',6' du noyau B sont particulièrement avantageux pour l'activité antimitotique des chalcones décrites dans cette invention.
Les sels des composés selon l'invention sont préparés selon des techniques bien connues de l'homme de l'art. Les sels des composés de formule (I) selon la présente invention comprennent ceux avec des acides minéraux ou organiques qui permettent une séparation ou une cristallisation convenable des composés de formule (I), ainsi que des sels pharmaceutiquement acceptables. En tant qu'acide approprié, on peut citer: l'acide picrique, l'acide oxalique ou un acide optiquement actif, par exemple un acide tartrique, un acide dibenzoyltartrique, un acide mandélique ou un acide camphosulfonique, et ceux qui forment des sels physiologiquement acceptables, tels que le chlorhydrate, le bromhydrate, le sulfate, l'hydrogénosulfate, le dihydrogénophosphate, le maléate, le fumarate, le 2- naphtalènesulfonate, le paratoluènesulfonate.
Lorsqu'un composé selon l'invention présente un ou plusieurs carbones asymétriques, les isomères optiques de ce composé font partie intégrante de l'invention. La présente invention comprend les composés de formule (I) sous forme d'isomères purs mais également sous forme de mélange d'isomères en proportion quelconque. Les composés (I) sont isolés sous forme d'isomères purs par les techniques classiques de séparation: on pourra utiliser, par exemple des recristallisations fractionnées d'un sel du racémique avec un acide ou une base optiquement active dont le principe est bien connu ou les techniques classiques de chromatographies sur phase chirale ou non chirale.
Les groupes fonctionnels éventuellement présents dans la molécule des composés de formule (I) et dans les intermédiaires réactionnels peuvent être protégés, soit sous forme permanente soit sous forme temporaire, par des groupes protecteurs qui assurent une synthèse univoque des composés attendus. Les réactions de protection et déprotection sont effectuées selon des techniques bien connues de l'homme de l'art. Par groupe protecteur

Les composés de formule (I) selon l'invention dans lesquels W = H sont obtenus par condensation aldolique d'une acétophénone de formule (II) et d'un benzaldéhyde de formule (III) comme illustré sur le Schéma 1 ci-après dans lequel X, Y, Z, Rl, R2, R3 R4 et R5 sont tels que définis pour (I) :

Une telle condensation aldolique est réalisée en milieu basique, par exemple en présence de potasse, de préférence dans un solvant polaire comme le méthanol.
Les composés de formule (II) sont des composés commerciaux ou préparés selon des techniques bien connues de l'homme de l'art. Par exemple, dans le cas où OZ, X, Y = OMe, l'acétophénone (II) est préparée par méthylation de l'acétophénone hydroxylée en utilisant le sulfate de méthyle ou l'iodure de méthyle comme agents de méthylation selon des techniques bien connues de l'homme de l'art.
Dans le cas où X = NH2, l'acétophénone (II) pourra être préparée selon la méthode décrite dans par N. Deka; M. Hadjeri; M. Lawson; C. Beney; A-M. Mariotte et A. Boumendjel dans "Acetylated dimethoxyaniline as a key

Pour les composés dans lesquels W est un méthoxy, l'acétophénone est obtenue à partir de la phloroglucinol et du méthoxyacétonitrile selon la méthode décrite par K. Wâhâla et T. A. Hase dans "Expédient synthesis of polyhydroxyisoflavones ", J. Chem. Soc, Perkin Trans. 1. 1991, 3005-3008. Les composés dans lesquels W est un hydroxy, O-éthyl ou O-benzyl, l'acétophénone est obtenue à partir de la phloroglucinol d'une part et de l'acide hydroxyacétique, de l'acide 2-éthyloxyacétique ou de l'acide 2-benzyloxyacétique respectivement, selon la méthode décrite par Wahala et al.
Les composés de formule (III) sont des composés commerciaux. Aucun signe de toxicité n'est observé avec ces composés aux doses pharmacologiquement actives et leur toxicité est donc compatible avec leur utilisation comme médicaments.
En particulier, les composés de formule générale (I) définis précédemment, ou leurs sels pharmaceutiquement acceptables, pourront être utilisés pour préparer un médicament destiné à traiter les maladies / désordres / phénomènes naturels suivants : la prolifération et la dissémination tumorale, la prolifération excessive des cellules normales, l'angiogénèse pathologique, l'activité excessive du système immunitaire. Les composés selon l'invention sont particulièrement intéressants pour leur activité anti-mitotique et pourront être utilisés comme anti-tumoraux ou anticancéreux notamment. De façon générale, les composés selon l'invention pourront être utilisés pour préparer un médicament destiné au traitement, à titre curatif ou préventif, de tout type de cancer.
En particulier, les composés de formule (I), ainsi que leurs sels pharmaceutiquement acceptables, pourront être utilisés pour la fabrication de médicaments destinés au traitement ou à la prévention de lésions tumorales bénignes, de troubles de l'immunité se caractérisant par une activité excessive du système immunitaire, de pathologies caractérisées par
une activité angiogénique excessive, de cancers, incluant les hémopathies malignes et les tumeurs solides, y compris les tumeurs d'origine glandulaire, mésenchymateuse, génitale, cutanée et neurologique.
A titre d'exemples de telles pathologies, on peut citer les lésions tumorales bénignes telles que les naevi cutanés, les polypes aérodigestifs et muqueux, les adénomes ; les troubles de l'immunité se caractérisant par une activité excessive du système immunitaire tels que les maladies auto- immunes avec auto-anticorps, représentées de façon non exhaustive par la polyarthrite rhumatoide, le lupus, la sclérodermie, les thyroïdites autoimmunes, le vitiligo, les cirrhoses autoimmunes, les pneumonies autoimmunes ; les manifestations secondaires à une hypersensibilité, par exemple rhinite, conjonctive et asthme allergiques, eczéma, allergies médicamenteuses ; les pathologies caractérisées par une activité angiogénique augmentée représentées de façon non exhausive par la rétinopathie diabétique, les angiomes.
En particulier, on pourra citer comme cancers pouvant être traités par les composés de la présente invention, les carcinomes, les hémopathies malignes des lignées myéloïdes et lymphoïdes, les tumeurs d'origine mésenchymateuse, les sarcomes, les tumeurs du système nerveux central et périphérique, les mélanomes, les séminomes, tératocarci nomes, ostéosarcomes, le xéroderma pigmentosum, le chératoaquantum, les néoplasies endocrines, et le sarcome de Kaposi. La présente invention a donc également pour objet les composés de formule (I), ainsi que leurs sels pharmaceutiquement, ou éventuellement solvats ou hydrates, en tant que médicaments, des compositions pharmaceutiques contenant une dose efficace d'un composé selon l'invention ou d'un sel, d'un solvat ou d'un hydrate pharmaceutiquement acceptable de celui-ci, et des excipients convenables.
Lesdits excipients sont choisis selon la forme pharmaceutique et le mode d'administration souhaité.
Dans les compositions pharmaceutiques de la présente invention pour l'administration orale, sublinguale, sous-cutanée, intramusculaire, intraveineuse, topique, intratrachéale, intranasale, transdermique, rectale ou intraoculaire, les principes actifs de formule (I) ci-dessus, ou leurs sels, solvats et hydrates éventuels, peuvent être administrés sous formes unitaires d'administration, en mélange avec des supports pharmaceutiques classiques, aux animaux et aux êtres humains pour la prophylaxie ou le traitement des troubles ou des maladies ci-dessus. Les formes unitaires d'administration appropriées comprennent les formes par voie orale telles que les comprimés, les gélules, les poudres, les granules et les solutions ou suspensions orales, les formes d'administration sublinguale, buccale, intratrachéale, intranasale, les formes d'administration sous-cutanée, intramusculaire ou intraveineuse et les formes d'administration rectale. Pour l'application topique, on peut utiliser les composés selon l'invention dans des crèmes, pommades, lotions ou collyres.
Afin d'obtenir l'effet prophylactique ou thérapeutique désiré, chaque dose unitaire peut contenir de 0,1 à 10000 mg, de composé selon l'invention en combinaison avec un support pharmaceutique. Cette dose unitaire peut être administrée 1 à 5 fois par jour de façon à administrer un dosage journalier permettant d'obtenir l'effet souhaité.
Lorsqu'on prépare une composition solide sous forme de comprimés, on mélange l'ingrédient actif principal avec un véhicule pharmaceutique, tel que la gélatine, l'amidon, le lactose, le stéarate de magnésium, le talc, la gomme arabique ou analogues. On peut enrober les comprimés de saccharose, d'un dérivé cellulosique, ou d'autres matières appropriées ou encore on peut les traiter de telle sorte qu'ils aient une activité prolongée ou retardée et qu'ils libèrent d'une façon continue une quantité prédéterminée de principe actif.
On obtient une préparation en gélules en mélangeant l'ingrédient actif avec un diluant et en versant le mélange obtenu dans des gélules molles ou dures.
Les compositions pharmaceutiques contenant un composé de l'invention peuvent aussi se présenter sous forme liquide, par exemple, des solutions, des émulsions, des suspensions ou des sirops. Les supports liquides appropriés peuvent être, par exemple, l'eau, les solvants organiques tels que le glycérol ou les glycols, de même que leurs mélanges, dans des proportions variées, dans l'eau.
Une préparation sous forme de sirop ou d'élixir ou pour l'administration sous forme de gouttes peut contenir l'ingrédient actif conjointement avec un édulcorant, acalorique de préférence, du méthylparaben et du propylparaben comme antiseptique, ainsi qu'un agent donnant du goût et un colorant approprié. Les poudres ou les granules dispersibles dans l'eau peuvent contenir l'ingrédient actif en mélange avec des agents de dispersion ou des agents mouillants, ou des agents de mise en suspension, comme la polyvinylpyrrolidone, de même qu'avec des édulcorants ou des correcteurs dé goût.
Pour une administration rectale, on recourt à des suppositoires qui sont préparés avec des liants fondant à la température rectale, par exemple du beurre de cacao ou des polyéthylèneglycols. Pour une administration parentérale, on utilise des suspensions aqueuses, des solutions salines isotoniques ou des solutions stériles et injectables qui contiennent des agents de dispersion et/ou des mouillants pharmacologiquement compatibles, par exemple le propylèneglycol ou le butylèneglycol. Le principe actif peut être formulé également sous forme de microcapsules, éventuellement ave un ou plusieurs supports ou additifs, ou bien avec des matrices telles qu'un polymère ou une cyclodextrine (patch, formes à libération prolongée).
Les compositions de la présente invention peuvent contenir, à côté des produits de formule (I) ci-dessus ou de leurs sels, solvats et hydrates pharmaceutiquement acceptables, par exemple des principes actifs qui peuvent être utiles dans le traitement des troubles ou maladies indiquées ci- dessus.
Ainsi, la présente invention a également pour objet des compositions pharmaceutiques contenant plusieurs principes actifs en association dont l'un est un composé selon l'invention.
L'invention a donc pour objet leur utilisation comme médicaments pouvant être utilisés seuls ou en combinaison avec des traitements tels que la chimiothérapie, la radiothérapie ou les traitements anti-angiogéniques mettant éventuellement en œuvre d'autres substances actives.
Par ailleurs, d'une façon générale, les mêmes préférences que celles indiquées précédemment pour les composés de formule générale (I) sont applicables mυtatis mutandis aux médicaments, compositions pharmaceutiques et utilisation mettant en œuvre les composés selon l'invention.
Les exemples ci-après, en référence à la Figure 1, sont donnés pour illustrer l'invention, mais n'ont aucun caractère limitatif. La Figure 1 montre l'effet dose réponse des composés des exemples 1 et 2 et du composé A sur le blocage en cycle des cellules K562.
Préparation des composés de formule (I) :
Exemple 1
2,2',4,6,6'-pentaméthoxychalcone de formule :
1 nriM de 2,6-méthoxyacétophénone et ImM de 2,4,6- triméthoxybenzaldéhyde dans 10 ml de méthanol, en présence de 1 ml de KOH à 25% sont chauffés à 700C pendant 3 heures. Le solvant est ensuite évaporé sous pression réduite, de l'eau est ajoutée et le mélange est extrait
avec du CH2CI2. La phase organique est séchée, évaporée. Les produits obtenus sont solides. La purification s'effectue par chromatographie en éluant avec un mélange d'acétate d'éthyle et de cyclohexane (1 :5). La chalcone est obtenue avec un rendement de 24 %. F = 1700C
Les exemples 2 à 22 donnés dans le TABLEAU 1 ci-aprés ont été préparés d'une façon analogue, en présence de 1 ml de KOH à 50%. La purification s'effectue soit par lavage à l'éther (a), soit par chromatographie (b).

Pour l'exemple 14, on utilise ImI de potasse (KOH) diluée à 25% comme pour l'exemple 1.
Dans le cas de l'exemple 9, l'acétophénone utilisée est préparée selon la méthode décrite dans Heterocydes 2002, 57, 123-128.
La α,2,2',4,4',6,6'-heptaméthoxychalcone est préparée d'une façon analogue à l'exemple 1. L'acétophénone utilisée (2,4,6,05- tétraméthoxyacétophénone) est préparée selon la méthode décrite dans J. Chem. Soc, Perkin Trans. 1. 1991, 3005-3008. Pour l'ensemble de ces composés, il a été démontré par résonance magnétique nucléaire que la double liaison α,β est trans.
A. Test de cycle
LE TEST DE CYCLE CORRESPOND A L'ANALYSE DU BLOCAGE EN PHASE G2/M D'UNE
LIGNEE DE CELLULES LEUCEMIQUES HUMAINES TEL QU'IL EST EVALUE PAR CYTOMETRIE EN FLUX.
Les cellules K562 sont incubées pendant 24 heures en absence ou en présence de composés selon l'invention et/ou de composés de référence tels que la vincristine. Les composés sont utilisés à une concentration de 10 μM dans un premier temps, des études de type dose-réponse pouvant être effectués secondairement. Au bout de 24 heures, les cellules sont exposées au iodure de propidium, agent intercalant de l'ADN, et analysées en
cytométrie de flux. Les résultats sont exprimés en pourcentage de cellules bloquées en phase G2/M du cycle cellulaire.
Les résultats présentés dans le TABLEAU 2 ci-après sont obtenus avec des composés de formule (I) de l'invention et sont présentés en comparaison avec le % d'inhibition d'un composé décrit dans l'art antérieur US 6462075 (nommé composé A) :

Tests MTT
Le test MTT est un test métabolique utilisé pour évaluer l'inhibition de prolifération de lignées tumorales par un composé selon l'invention. Plusieurs lignées tumorales représentatives de différents types tumoraux tels que le cancer du sein, le cancer du colon, les tumeurs ORL, le cancer du poumon, sont incubées en plaque 96 puits à des concentrations variables d'un composé selon l'invention, pendant 72 heures puis le MTT est rajouté pendant une période de 1 heure. Le MTT est un substrat d'enzymes mitochondriales qui est dégradé en bleu de formazan insoluble. Les cristaux sont ensuite resolubilisés dans de l'isopropanol-HCI O,1N et les plaques sont analysées sur un lecteur de plaque ELISA. La survie relative est calculée par rapport aux cellules n'ayant pas été exposées au composé selon l'invention. L'ICso ou concentration inhibitrice 50 est la valeur de concentration du composé selon l'invention étudié qui induit une diminution de prolifération de 50% par rapport au contrôle.
La comparaison des composés des exemples 1 et 2 au composé A ont donc été effectuée en réalisant des études dose-réponse en cytométrie en flux et des études de cytotoxicité (« tests MTT »). Ce dernier test reflète la capacité des composés à détruire les cellules tumorales. Les résultats présentés Figure 1 et TABLEAU 3 montrent que les composés des exemples 1 et 2 possèdent une meilleure activité que le composé A, à la fois en test de cytométrie en flux et en test de cytotoxicité :
- nous observons sur la Figure 1 un blocage en cycle en G2/M significatif dès la concentration de l'ordre du micromolaire pour les composés des exemples 1 et 2 alors que le composé A n'a pas d'activité à cette même concentration ;
- les concentrations inhibitrices IC50 en cytotoxicité sont environ 10 fois plus faibles pour les composés des exemples 1 et 2 que pour le composé A dans plusieurs lignées tumorales représentatives de différents types de cancers comme le montrent les résultats des tests de cytotoxicité (MTT) in vitro présentés dans le TABLEAU 3.

L'étude de relation structure-activité avec l'ensemble des composés revendiqués indique que la nature des substituants des groupes phényl et leur position sont déterminantes pour l'activité antimitotique.
Sur le noyau A de la chalcone, un groupe méthoxy semble être le plus avantageux parmi tous les substituants étudiés et les positions 2,4 et 6 sont les plus importantes pour la substitution. La taille du substituant et donc l'encombrement stérique autour de cette région de la molécule a également une influence, de sorte que les groupes méthoxy sont préférés.
Les groupes plus hydrophobes tels que le méthoxy semblent également plus avantageux. Enfin, il faut souligner que la triméthoxylation sur les trois positions (2,4 et 6) est la plus avantageuse comparée à une diméthoxylation ou monométhoxylation sur les mêmes positions. Le rôle joué par l'oxygène du méthoxy est essentiel car la substitution d'un méthoxy par un méthyl provoque une diminution de l'effet antimitotique. Il est à souligner que le maintien d'une triméthoxylation en 2,4 et 6 et une perturbation électronique par l'introduction de deux atomes de fluorés en 3 et 5 aboutissent également à une baisse sensible d'activité.
Sur le noyau B, on remarque là aussi le rôle supérieur du groupe méthoxy. Une méthoxylation de préférence sur les positions 2,4 et 6 induit un bon effet antimitotique. La taille du substituant et donc l'encombrement stérique autour de cette région de la molécule a également une influence, de sorte que les groupes méthoxy sont préférés.
Les groupes plus hydrophobes tels que le méthoxy semblent également plus avantageux.
Le maintien de deux méthoxy en 2' et 6' et la présence d'un groupe NH2 (amino) en 4' n'induit q'une légère perte d'activité. La présence du NH2 devrait permettre d'accéder rapidement à d'autres dérivés de chalcones par une simple alkylation ou acylation de l'azote.
En conclusion, la présence d'une di ou tri méthoxylation sur les carbones 2,4,6 du noyau A et 2',4',6' du noyau B sont particulièrement avantageux pour l'activité antimitotique des chalcones décrites dans cette invention.
Claims
REVENDICATIONS
1 - Nouveaux dérivés de chalcone de formule (I) :
- X représente un atome d'hydrogène ou un groupe -OH, -O-méthyl,
-O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl, -NH2, -NHCOR3 avec R3 qui est un groupe alkyle linéaire ou ramifié de 1 à 5 atomes de carbone, un groupe aryl qui peut être mono ou polysubstituté par un substituant choisi parmi les atomes d'halogène, les groupes OH, OMe et -NRbR0 avec Rb et Rc qui représentent indépendamment l'un de l'autre une chaîne alkyle linéaire ou ramifiée de 1 à 6 atomes de carbone,
- Y représente un atome d'hydrogène ou un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl, étant entendu qu'un au moins des groupes X et Y est différent de l'hydrogène,
- Z représente un atome d'hydrogène ou un groupe méthyl, éthyl, propyl, isopropyl, benzyl,
- W représente un atome d'hydrogène, ou un groupe -OH, -O-méthyl, -O-éthyl, ou -O-benzyl, - Rl, R2, R3, R4 et R5 représentent, indépendamment les uns des autres, un atome d'hydrogène ou d'halogène ou un groupe -OH, méthyl, -O-méthyl, éthyl, -O-éthyl, propyl, -O-propyl, benzyl, -O-benzyl, -NH2, -NHCH3, -N(CH3)2, étant entendu qu'au moins deux substituants Rl, R2, R3, R4 et R5 sont différents de H, et que lorsque X ou Y représente un atome d'hydrogène, au moins trois substituants Rl, R2, R3, R4 et R5 sont différents de H,
ainsi que leurs hydrates, solvats ou sels pharmaceutiquement acceptables, à l'exception du 2'-hydroxy-4',6',2,4,5-pentaméthoxychalcone, et du 2',4',6',2,4,5- pentaméthoxychalcone.
2 - Nouveaux dérivés de chalcone selon la revendication 1 caractérisés en ce qu'au moins trois des substituants Rl, R2, R3, R4 et R5 sont différents de l'hydrogène.
3 - Nouveaux dérivés de chalcone selon la revendication 1 ou 2 caractérisés en ce que X représente un atome d'hydrogène ou un groupe -OMe, -OEt, -NH2, le groupe OMe étant préféré, étant entendu que, quand X représente un atome d'hydrogène, Y représente un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl.
4 - Nouveaux dérivés de chalcone selon l'une des revendications 1 à 3 caractérisés en ce que Y représente un atome d'hydrogène ou un groupe -OMe, -OEt, le groupe OMe étant préféré, étant entendu que, quand Y représente un atome d'hydrogène, X représente un groupe -OH, -O-méthyl, -O-éthyl, -O-propyl, -O-isopropyl, -O-benzyl.
5 - Nouveaux dérivés de chalcone selon l'une des revendications 1 à 4 caractérisés en ce que Z représente un atome d'hydrogène ou un groupe méthyl ou éthyl. 6 - Nouveaux dérivés de chalcone selon l'une des revendications 1 à 5 caractérisés en ce que W représente un groupe -O-méthyl ou de préférence un atome d'hydrogène.
7 - Nouveaux dérivés de chalcone selon l'une des revendications 1 à 6 caractérisés en ce que Rl, R2, R3, R4 et R5 représentent, indépendamment les uns des autres, un atome d'hydrogène ou de fluor, un méthyl, -O-méthyl, -O-éthyl, étant entendu qu'au moins deux substituants Rl, R2, R3, R4 et R5 sont différents de H, et que lorsque X ou Y représente un atome d'hydrogène, au moins trois substituants Rl, R2, R3, R4 et R5 sont différents de H. 8 - Nouveaux dérivés de chalcone selon l'une des revendications 1 à 7 caractérisés en ce que ils répondent à la formule (Ia) :

12 - Utilisation d'un des composés selon l'une des revendications 1 à 9 pour la fabrication d'un médicament destiné au traitement ou à la prévention de cancers, incluant les hémopathies malignes et les tumeurs solides, y compris les tumeurs d'origine glandulaire, mésenchymateuse, génitale, cutanée et neurologique.
13 - Utilisation d'un des composés selon l'une des revendications 1 à 9 pour la fabrication d'un médicament destiné au traitement ou à la prévention de lésions tumorales bénignes. 14 - Utilisation d'un des composés selon l'une des revendications 1 à 9 pour la fabrication d'un médicament destiné au traitement ou à la prévention troubles de l'immunité se caractérisant par une activité excessive du système immunitaire.
15 - Utilisation d'un des composés selon l'une des revendications 1 à 9 pour la fabrication d'un médicament destiné au traitement ou à la prévention de pathologies caractérisées par une activité angiogénique excessive.
