Abstract
Die Erfindung betrifft ein alternatives Verfahren zur Herstellung von substituierten 5-Aminopyrazolen, in dem eine Carbonylverbindung mit einem Hydrazinderivat und Chlorcyan zu einem N-Cyanohydrazon umgesetzt werden, welches in Gegenwart einer starken Base cyclisiert wird.
Description
Verfahren zur Herstellung von substituierten Pyrazolen
Die Erfindung betrifft ein alternatives Verfahren zur Herstellung von substituierten 5-Aminopyrazolen.
Verbindungen der Formel I,
wie jenes worin R Cyclopropyl, R Wasserstoff und R Methyl ist, sind synthetische Bausteine, die beispielsweise verwendet werden können für die Herstellung von Azetidinen. Azetidine sind gemäss WO-A-2003/077907 und WO-A-2005/026113 geeignet als CCR-3 Rezeptor- Antagonisten in der Behandlung von Entzündungen und allergischen Krankheiten.
Mit dem bekannten Verfahren von Höhn, H., Z. Chem. 1970, 10, 386-388, zitiert in US-A-3894005, lassen sich 5-Aminopyrazole der Formel I aus einem Acrylsäurenitril der Formel CHR1=CR2CN herstellen, welches zunächst mit Hydrazin, einem Aldehyd oder Keton zum Hydrazon umgesetzt wird und anschliessend durch Behandlung mit Natrium- butanolat cyclisiert werden kann.
Aus Ryckmans et al., Tetrahedron, 1997, 53, 1729-1734, ist ein Verfahren bekannt, in dem aktivierte enolisierbare Ketone mit Hydrazinen zu l-Cyano-2-vinyl-hydrazonen umgesetzt werden, welche dann entweder thermisch oder in Gegenwart einer Base in 4-substituierte 5-Aminopyrazole der Formel I umgesetzt werden. Die aktivierende Gruppe R2 an der 4-Position des Pyrazolringes bleibt hierbei erhalten. Vorzugsweise ist die aktivierende Gruppe R2 eine Acylgruppe. R2 = Phenyl ist bereits weitaus weniger begünstigt. Bei der Cyclisierung eines 5:1 E/Z-Hydrazon-Gemisches aus des iV-Cyano-N-methylhydrazons von Benzylmethylketon (R2 = Phenyl) wird l,3-Dimethyl-4-phenyl-5-aminopyrazol in 66% Ausbeute gebildet. Die Bildung von l,4-Dimethyl-3-phenyl-5-aminopyrazol wird nicht berichtet. Ohne eine aktivierende Gruppe, also beispielsweise mit R2 = Wasserstoff
oder Ci-6-Alkyl konnte nach dem Verfahren von Ryckmans kein 5-Aminopyrazol erhalten werden.
Andere Verfahren für die Herstellung von 5-Amino-l,3-dimethylpyrazol werden in WO-A-94/13661 und WO-A-95/34563 offenbart.
Aufgabe der Erfindung war es ein alternatives Verfahren zur Herstellung von substituierten 5-Aminopyrazolen zur Verfügung zu stellen. Zur Steigerung der Verfahrensökonomie sollten darüber hinaus die Substituenten R1 bis R3 bereits beim Ringaufbau ins Molekül eingeführt werden können um eine nachträgliche Substitution zu vermeiden.
Diese Aufgabe wird entsprechend Anspruch 1 gelöst.
Beansprucht wird ein Verfahren zur Herstellung von substituierten Pyrazolen der Formel 3

worin R1 ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Ci-6-Alkyl,
Ci-6-Alkoxy, C3.6-Cycloalkyl, Aryl und Heteroaryl, worin ausser Wasserstoff jeder Substituent R1 gegebenenfalls einen oder mehrere Substituenten aus der Gruppe bestehend aus Ci-6-Alkyl, C1-6-Alkoxy, Halogen, Nitro tragen kann, und R2 ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Cyano, Halogen, Ci-6-Alkyl, C1-6-Alkoxy, Ci-6-Alkoxycarbonyl, C3-6-Cycloalkyl, Aryl und Heteroaryl, worin ausser Wasserstoff, Cyano und Halogen jeder Substituent R2 gegebenenfalls einen oder mehrere Substituenten aus der Gruppe bestehend aus Ci-6-Alkyl, C1-6-Alkoxy, Halogen, Nitro tragen kann, oder worin R und R gemeinsam eine -(CH2)n- Gruppe mit n = 3, 4 oder 5 repräsentieren, die gegebenenfalls ein oder mehrere Halogenatome enthalten kann, und worin R3 ausgewählt ist aus der Gruppe bestehend aus Ci-6-Alkyl, C3.6-Cycloalkyl, Aryl und Heteroaryl, worin jeder Substituent R3 gegebenenfalls mit einem oder mehreren Halogenatomen substituiert ist,
indem in einer ersten Stufe eine Verbindung der Formel

worin R1, R und R3 wie oben definiert sind, welche in einer letzten Stufe in Gegenwart einer starken Base zu einer Verbindung der Formel I umgesetzt wird.
Die Verbindungen der Formel II können Aldehyde oder Ketone sein. Im Falle von unsym- metrischen Ketonen können zwei unterschiedliche Verbindungen der Formel I entstehen. Welches Produkt überwiegt, hängt von den sterischen und elektronischen Eigenschaften der Substituenten R1 und R2 sowie den Reaktionsbedingungen ab. In jedem Fall unterscheiden sich die beiden Produkte jedoch wesentlich voneinander, so dass sie leicht
getrennt werden können. Falls R1 und R2 gemeinsam eine -(CH2)n- Gruppe mit n = 3, 4 oder 5 repräsentieren, ist die Verbindung der Formel II ein cyclisches Keton. Beispiele für cyclische Ketone der Formel II sind Cyclopentanon, Cyclohexanon oder Cycloheptanon.
In einem bevorzugten Verfahren ist die Verbindung der Formel II aus der entsprechenden Spalte der Tabelle 1 ausgewählt. In der Spalte „Verbindung der Formel I" ist jeweils angegeben, welche Verbindung bei der Reaktion überwiegend entsteht. Der Rest R3 ergibt sich darin durch das jeweils verwendete Hydrazinderivat der Formel III.

Hier und im Folgenden bedeutet der Ausdruck „Ci-n-Alkyl" eine unverzweigte oder verzweigte Alkylgruppe mit 1 bis n Kohlenstoffatomen. So repräsentiert Ci-7-Alkyl beispiels-
weise Gruppen wie Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, sec-Buty\, tert-Butyl, Pentyl, Hexyl, Heptyl oder 1,4-Dimethyl-pentyl.
Hier und im Folgenden bedeutet der Ausdruck „Ci-n-Alkoxy" eine unverzweigte oder ver- zweigte Alkoxygruppe mit 1 bis n Kohlenstoffatomen. So repräsentiert Ci-7-Alkoxy beispielsweise Gruppen wie Methoxy, Ethoxy, Propoxy, Isopropoxy, Butoxy, Isobutoxy, sec- Butoxy, tert-Butoxy, Pentyloxy, Hexyloxy, Heptyloxy oder 1,4-Dimethyl-pentyloxy.
Hier und im Folgenden bedeutet der Ausdruck „C3-6-Cycloalkyl" eine Cycloalkylgruppe mit 3 bis 6 Kohlenstoffatomen und repräsentiert Cyclopropyl, Cyclobutyl, Cyclopentyl und Cyclohexyl.
Unter Aryl wird hier und im Folgenden insbesondere eine aromatische Gruppe mit 6 bis 10 Kohlenstoffatomen verstanden, wie beispielsweise Phenyl, p-Tolyl oder Naphthyl.
Unter Aralkyl wird hier und im Folgenden insbesondere eine mit einer Arylgruppe substituierte Alkylgruppe wie beispielsweise Phenylethyl verstanden, wobei die Alkylgruppe 1 bis 4 Kohlenstoffatome enthält, und die Arylgruppe 6 bis 10 Kohlenstoffatome enthält, wie oben definiert.
Unter Heteroaryl wird hier und im Folgenden insbesondere eine heteroaromatische Gruppe mit 4 bis 8 Kohlenstoffatomen verstanden, wie beispielsweise 2- oder 3-Furanyl, 2- oder 3-Thiophenyl oder 2-, 3- oder 4-Pyridinyl.
Hier und im Folgenden bedeutet der Ausdruck „Halogen" Fluor, Chlor, Brom oder Iod.
In einer bevorzugten Verfahrensvariante ist die in der zweiten Stufe verwendete Base eine anorganische Base, vorzugsweise ausgewählt aus der Gruppe bestehend aus Alkali- und Erdalkalihydroxiden, Alkali- und Erdalkalicarbonaten, Trinatriumphosphat und Gemischen davon.
Vorzugsweise wird die erste Stufe bei der Rückflusstemperatur des gewählten Lösungsmittels durchgeführt. Der Reaktionsfortschritt kann sehr einfach über Dünnschicht-
Chromatographie oder Gaschromatographie bestimmt werden.
Das Produkt der ersten Stufe muss nicht isoliert werden und kann direkt weiter umgesetzt werden.
In der zweiten Stufe wird das in der ersten Stufe gebildete Hydrazinderivat in Gegenwart einer Base mit Chlorcyan umgesetzt. Insbesondere anorganische Basen sind für die zweite Stufe geeignet und können ausgewählt werden aus der Gruppe bestehend aus Alkali- und Erdalkalihydroxiden, Alkali- und Erdalkalicarbonaten, Trinatriumphosphat und Gemischen davon. Besonders bevorzugt wird als Base ein Alkalicarbonat und hierbei besonders Kaliumcarbonat verwendet.
In einer bevorzugten Verfahrensvariante wird das Chlorcyan in der zweiten Stufe als Gas oder gelöst in einem Lösungsmittel verwendet. Es kommt im erfindungsgemässen Ver- fahren nicht darauf an, ob die Reaktionsmischung der ersten Stufe zum Chlorcyan oder das Chlorcyan zur Reaktionsmischung gegeben wird.
In einer besonders bevorzugten Verfahrensvariante wird die zweite Stufe bei einer Temperatur zwischen -100 und 0 0C durchgeführt, besonders bevorzugt zwischen -70 und -20 0C.
Da das Produkt der ersten Stufe in der zweiten Stufe direkt weiter mit Chlorcyan umgesetzt werden kann, ist es besonders vorteilhaft, die Reaktionen der ersten und zweiten Stufe als „Eintopfreaktion" durchzuführen.
Insbesondere können Lösungsmittel für die erste und zweite Stufe ausgewählt werden aus der Gruppe bestehend aus Cyclohexan, Hexan, Heptan, Petrolether, Ethanol, Diethylether, Methyl-/ert-butylether (MTBE), Tetrahydrofuran (THF), Toluol, Xylol und Gemischen davon. Besonders bevorzugt sind MTBE, THF und Toluol.
Unter Petrolether werden allgemein technische Alkangemische mit einem relativ breiten Siedebereich verstanden, insbesondere aber auch Isomerengemische beispielsweise von Hexan und Heptan.
Die in der dritten Stufe verwendete starke Base muss in der Lage sein, das unmittelbar an die Gruppe R gebundene Kohlenstoffatom zu deprotonieren. Vorzugsweise wird die starke Base ausgewählt aus der Gruppe bestehend aus Metallhydriden, Metallamiden, Metallalkoxiden und metallorganischen Verbindungen. Als Metallhydride werden vorzugs- weise NaH oder KH verwendet. Metallamide werden vorzugsweise aus der Gruppe bestehend aus Natriumamid, Lithiumdiisopropylamid (LDA) und dem Lithiumamid von Tetramethylpiperidin (Li-TMP) ausgewählt. Als Metallalkoxide werden vorzugsweise Natriumethanolat und Kalium-ter/-butoxid verwendet. Metallorganische Verbindungen werden vorzugsweise ausgewählt aus der Gruppe bestehend aus «-Butyllithium, sec-Buty\- lithium und tert-Butyllithium.
In einer besonders bevorzugten Verfahrensvariante ist die Base ausgewählt aus der Gruppe bestehend aus Lithium-diisopropylamid, Kalium-ter/-butoxid, «-Butyllithium, sec-Butyl- lithium und /ert-Butyllithium.
In einer besonders bevorzugten Verfahrensvariante wird die dritte Stufe bei einer Temperatur zwischen -100 und 0 0C durchgeführt, besonders bevorzugt zwischen -70 und -20 0C.
Für die dritte Stufe muss das Lösungsmittel inert gegen die verwendete starke Base sein. In Beispiel 3 wird ein Lösungsmittelwechsel durchgeführt, welcher aber nicht zwingend ist. In einer bevorzugten Verfahrensvariante findet zwischen der zweiten und dritten Stufe kein Lösungsmittelwechsel statt.
Ein Lösungsmittel, welches gegenüber allen Reagentien der drei Stufen inert ist, kann verwendet werden um die Reaktionen der ersten, zweiten und dritten Stufe als „Eintopfreaktion" durchzuführen.
Für die dritte Stufe wird das Lösungsmittel vorzugsweise ausgewählt aus der Gruppe bestehend aus Cyclohexan, Hexan, Heptan, Petrolether, Diethylether, MTBE, THF, Toluol, Xylol und Gemischen davon. Besonders bevorzugt sind MTBE, THF und Toluol.
In der weiteren bevorzugten Verfahrensvariante einer „Eintopfreaktion", welche alle drei Stufen umfasst, wird das Lösungsmittel ausgewählt aus der Gruppe bestehend aus Cyclo-
hexan, Hexan, Heptan, Diethylether, MTBE, THF, Toluol, Xylol und Gemischen davon, besonders bevorzugt aus sind MTBE, THF und Toluol.
Nach Reaktionsende, wird die starke Base gequencht, beispielsweise durch Wasserzugabe, und das Produkt kann isoliert werden. Vorzugsweise wird bei mit Wasser wenigstens teilweise mischbaren Lösungsmitteln ein Salz wie beispielsweise Ammoniumchlorid zur Phasentrennung dazugegeben.
Beispiele:
Herstellung einer Lithium-diisopropylamid-Lösung (LDA-Lösung):
1,6 M H-Butyllithium-Lösung (108 mL, 173 mmol) in Hexan (BuLi) wurde bei -60 °C zu einer Mischung aus Diisopropylamin (19,0 g, 189 mmol) in 200 mL THF gegeben und die Mischung 1 h gerührt. Die erhaltene LDA-Lösung wurde direkt in Beispiel 3 verwendet. Alternativ kann aber auch eine kommerziell erhältliche LDA-Lösung oder festes LDA verwendet werden, welches beispielsweise von Fluka erhältlich ist. Festes LDA ist gegebenenfalls vor Gebrauch in einem geeigneten Lösungsmittel zu lösen oder zu suspendieren, beispielsweise in THF, MTBE oder Hexan.
Beispiel 1: (£I/Z)-N-(l-Cyclopropyl-ethyliden)-N'-methylhydrazin (Mischung)
Eine Mischung aus Cyclopropylmethylketon (12,6 g, 150 mmol) und Methylhydrazin (11,0 g, 240 mmol) wurden in 100 mL Toluol bei einer Temperatur von 93 °C unter Rück- fluss für 11 h unter Rühren erhitzt. Nach Abschluss der Reaktion wurde die Reaktionsmischung auf 0 0C abgekühlt. Es wurde eine ((£/Z)-./V-(l-Cyclopropyl-ethyliden)- iV-methylhydrazin)-Mischung mit einer EIZ- Verteilung von ca. 3:1 erhalten, von der ein Aliquot für die Charakterisierung entnommen und gereinigt wurde. 1H-NMR (CDCl3): δ = 4,40 (br, IH), 3,92 (s, 3H2), 3,90 (s, 3H£), 1,72 (s, 3H2), 1,58 (m, IHf), 1,52 (s, 3H£), 1,50 (m, IH2), 0,80 (dt, 2H2), 0,70 (m, 2H2), 0,68 (m, 2H£), 0,62 ppm (m, 2H£).
Beispiel 2: (£"/Z)-iV-Cyan-iV-(l-cyclopropyl-etliyliden)-N-methylhydrazin (Mischung) Die Hauptmenge der Reaktionsmischung aus Beispiel 1 wurde nach dem Abkühlen mit einer wässrigen K2CO3-Lösung (27,6 g, 200 mmol in 55 mL Wasser) versetzt. Zu dieser
Mischung wurde bei 0 0C über 90 min Chlorcyan (14,0 g, 230 mmol) eingeleitet. An- schliessend wurde die Mischung noch 2 h bei 0 °C gerührt. Nach Reaktionsende wurde die organische Phase abgetrennt und Toluol abgedampft. Der ölige Rückstand (22,8 g Rohprodukt) wurde in 100 mL Tetrahydrofuran (THF) aufgenommen. Es wurde eine ((£/Z)-N-Cyan-Λp-(l-cyclopropyl-ethyliden)-N-methylhydrazin)-Mischung mit einer E/Z- Verteilung von ca. 3 : 1 erhalten, von der ein Aliquot für die Charakterisierung entnommen und gereinigt wurde.
1H-NMR (CDCl3): 6 = 3,18 (s, 3HZ), 3,16 (s, 3H£), 2,18 (m, 1HZ), 1,92 (s, 3H£), 1,70 (s, 3HZ), 1,68 (m, 1H£), 1,80 (dt, 2HZ), 0,86 (m, 2HZ), 0,82 ppm (m, 4H£).
Beispiel 3: 5-Cyclopropyl-2-methyl-2//-pyrazol-3-ylamin
Ca. 300 mL einer frisch bereiteten ca. 0,6 M LDA-Lösung (siehe oben) wurde bei -60 bis -65 °C mit 113 g der Rohproduktlösung aus Beispiel 2 innerhalb von 1 h versetzt. Die Kontrolle per Dünnschichtchromatographie ergab vollständigen Umsatz nach 1 h. Nach Reaktionsende konnte sich die Reaktionsmischung auf -10 0C erwärmen wurde dann mit gesättigter NH4C1-Lösung (30 mL) versetzt. Nach Phasentrennung wurde die organische Phase abgetrennt und die wässrige Phase erneut mit THF extrahiert (2x20 mL). Die vereinigten organischen Phasen wurden über MgSO4 getrocknet und bis zur Trockene eingedampft. Das Rohprodukt (18,9 g) wurde als hellgelber Feststoff mit einer Ausbeute von 92% erhalten, bezogen auf die ursprüngliche Menge von Cyclopropylethanon in Beispiel 1.
1H-NMR (CDCl3): δ = 5,20 (s, IH), 3,58 (s, 3H), 3,45 (br, 2H), 1,80 (m, IH), 0,82 (m, 2H), 0,62 ppm (m, 2H).
Beispiel 4: Umkristallisation von 5-Cyclopropyl-2-methyl-2H-pyrazol-3-ylamin
18,9 g des Produkts aus Beispiel 2 wurden in einer Mischung aus Diisopropylether (35 mL) und Ethylacetat (70 mL) bei 65 0C gelöst. Anschliessend wurde Hexan (25 mL) zugegeben und die Temperatur langsam auf 10 °C gesenkt. Der abgeschiedene Feststoff abfiltriert und das in der Mutterlauge verbliebene Produkt einmal erneut umkristallisiert. Insgesamt konnten 13,4 g 5-Cyclopropyl-2-methyl-2H-pyrazol-3-ylamin (65% bezüglich Cyclopropylethanon) als hellgelber Feststoff isoliert werden.
Claims
1. Verfahren zur Herstellung von substituierten 5-Aminopyrazolen der Formel
worin R1 ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Ci-6-Alkyl, C1-6-AIkOXy, C3-6-Cycloalkyl, Aryl und Heteroaryl, worin ausser Wasserstoff jeder
Substituent R1 gegebenenfalls einen oder mehrere Substituenten aus der Gruppe bestehend aus Ci-6-Alkyl, Ci-6-Alkoxy, Halogen, Nitro tragen kann, und R2 ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Cyano, Halogen, Ci-6-Alkyl, Ci-6-Alkoxy, Ci-6-Alkoxycarbonyl, C3-6-Cycloalkyl, Aryl und Heteroaryl, worin ausser Wasserstoff, Cyano und Halogen jeder Substituent R2 gegebenenfalls einen oder mehrere Substituenten aus der Gruppe bestehend aus Ci-6-Alkyl, Ci-6-Alkoxy, Halogen, Nitro tragen kann, oder worin R1 und R2 gemeinsam eine -(CH2)n- Gruppe mit n = 3, 4 oder 5 repräsentieren, die gegebenenfalls ein oder mehrere Halogenatome enthalten kann, und worin R3 ausgewählt ist aus der Gruppe bestehend aus Ci-6-Alkyl, C3-6-Cycloalkyl,
Aryl und Heteroaryl, worin jeder Substituent R3 gegebenenfalls mit einem oder mehreren Halogenatomen substituiert ist, indem in einer ersten Stufe eine Verbindung der Formel
worin R1 und R2 wie oben definiert sind, mit einer Verbindung der Formel
NH2NHR3 III
worin R3 wie oben definiert ist, umgesetzt wird zu einer Verbindung der Formel
worin R1, R2 und R3 wie oben definiert sind, welche dann in einer zweiten Stufe mit Chlorcyan in Gegenwart einer Base umgesetzt wird zu einer Verbindung der Formel
worin R1, R2 und R3 wie oben definiert sind, welche in einer letzten Stufe in Gegenwart einer starken Base zu einer Verbindung der Formel I umgesetzt wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die in der zweiten Stufe verwendete Base eine anorganische Base ist, vorzugsweise ausgewählt aus der
Gruppe bestehend aus Alkali- und Erdalkalihydroxiden, Alkali- und Erdalkali- carbonaten, Trinatriumphosphat und Gemischen davon.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Reaktionen der ersten und zweiten Stufe als „Eintopfreaktion" durchgeführt wird.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass die in der dritten Stufe verwendete starke Base ausgewählt ist aus der Gruppe bestehend aus Metallhydriden, Metallamiden, Metallalkoxiden und metallorganischen Verbindungen.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass die starke Base ausgewählt ist aus der Gruppe bestehend aus Lithium-diisopropylamid, Kalium-tert-but- oxid, «-Butyllithium, sec-Butyllithium und ter/-Butyllithium.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass zwischen der zweiten und dritten Stufe kein Lösungsmittelwechsel stattfindet.
7. Verfahren nach Anspruch 5 oder 6, dadurch gekennzeichnet, dass die Reaktionen der ersten, zweiten und dritten Stufe als „Eintopfreaktion" durchgeführt werden.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass das Lösungsmittel ausgewählt ist aus der Gruppe bestehend Cyclohexan, Hexan, Heptan, Petrolether, Diethylether, Methyl-tert-butylether (MTBE), Tetrahydrofuran (THF), Toluol, Xylol und Gemischen davon.
