Bei der Herstellung homogener Zubereitungen biologisch aktiven Substanzen hat die Solubilisierung von hydrophoben, also in Wasser schwerlöslichen Stoffen, eine sehr große praktische Bedeutung erlangt.
Unter Solubilisierung ist das Löslichmachen von in einem bestimmtem Lösungsmittel, insbesondere wasser, schwer- oder unlöslichen Substanzen durch grenzflächenaktive Verbindungen, die Solubilisatoren, zu verstehen. Solche Solublisatoren sind in der Lage, schlecht wasserlösliche oder wasserunlösliche Stoffe in klare, höchstens opaleszie- rende wässrige Lösungen zu überführen, ohne dass hierbei die chemische Struktur dieser Stoffe eine Veränderung erfährt (Vgl. Römpp Chemie Lexikon, 9. Auflage, Bd.5. S. 4203, Thieme Verlag, Stuttgart, 1992).
Die hergestellten Solubilisate sind dadurch gekennzeichnet, dass der schlecht wasser- lösliche oder wasserunlösliche Stoff in den Molekülassoziaten der oberflächenaktiven Verbindungen, die sich in wässriger Lösung bilden - den sogenannten Mizellen - kolloidal gelöst vorliegt. Die resultierenden Lösungen sind stabile einphasige Systeme, die optisch klar bis opaleszent erscheinen und ohne Energieeintrag hergestellt werden können.
Solubilisatoren können beispielsweise das Aussehen von kosmetischen Formulierungen sowie von Lebensmittelzubereitungen verbessern, indem sie die Formulierungen transparent machen. Außerdem kann im Falle von pharmazeutischen Zubereitungen auch die Bioverfügbarkeit und damit die Wirkung von Arzneistoffen durch die Verwen- düng von Solubilisatoren gesteigert werden.
Als Solubilisatoren für pharmazeutische Arzneistoffe und kosmetische Wirkstoffe werden hauptsächlich Tenside wie ethoxiliertes (hydriertes) Ricinusöl, ethoxilierte Sorbitanfettsäureester oder ethoxilierte Hydroxystearinsäure eingesetzt,.
Die oben beschriebenen, bisher eingesetzten Solubilisatoren zeigen jedoch eine Reihe anwendungstechnischer Nachteile.
So ist z. B. deren parenterale Applikation mit einer Freisetzung von Histamin und einem daraus resultierenden Blutdruckabfall verbunden (Lorenz et al., Agents and Actions, Vol. 12, 1/2, 1982).
Die bekannten Solubilisatoren besitzen für einige schwerlösliche Arzneistoffe wie z.B. Clotrimazol nur eine geringe lösungsvermittelnde Wirkung.
Grenzflächenaktive Verbindungen besitzen häufig eine hohe hämolytische Aktivität, die einer Anwendung auf dem Gebiet der Pharmazie, insbesondere in Parenteralia entgegensteht.
Eine weitere wünschenswerte Anforderung an Solubilisatoren ist die Fähigkeit, mit schwerlöslichen Substanzen sogenannte „feste Lösungen" auszubilden. Der Begriff feste Lösungen bezeichnet einen Zustand, in dem eine Substanz molekulardispers in einer festen Matrix, beispielsweise einer Polymermatrix, verteilt ist. Solche festen Lösungen führen zum Beispiel bei Verwendung in festen pharmazeutischen Darreichungsformen eines schwerlöslichen Wirkstoffs zu einer verbesserten Freisetzung des Wirkstoffs. Eine wichtige Anforderung an solche feste Lösungen ist, dass sie auch bei Lagerung über längere Zeit stabil sind, d.h., dass der Wirkstoff nicht auskristallisieren soll.
Bei der Ausbildung von festen Lösungen spielt neben der grundsätzlichen Fähigkeit der Solublisatoren zur Bildung von festen Lösungen auch die Hygroskopizität der SoIu- bilisatoren eine bedeutende Rolle. Solubilisatoren, die aus der Umgebungsluft zuviel Wasser aufnehmen, führen zu einem Zerfliessen der festen Lösung und der unerwünschten Kristallisation der Wirkstoffe. Auch bei der Verarbeitung zu Darreichungsformen kann eine zu große Hygroskopizität Probleme bereiten.
US 4,432,881 beschreibt hydrophob modifizierte Polyacrylsäure mit einem Molekulargewicht zwischen 200000 und 5000000, die durch Copolymerisation von Acrylsäure mit den entsprechenden N-Alkylacrylamiden oder Acrylaten erhalten werden. Verwendet werden die erhaltenen Polymere als dispergierbare hydrophobe Verdicker.
In der US 4,395,524 wird die Copolymerisation von hydrophilen Komponenten (z.B. Acrylamid, Acrylsäure, N-Vinylpyrrolidon u.a.) mit N-Alkylacrylamiden beschrieben. Die so erhaltenen Polymere mit einem Molekulargewicht von 30000 bis 2000000 werden als Verdicker, Sedimentationsstabilisatoren oder Dispergiermittel verwendet.
EP-A-O 268 164 beschreibt die Verwendung von Copolymerisaten von monoolefinisch ungesättigten Säuren und Alkylester monoolefinisch ungesättigter Säuren zur Stabilisierung von O/W-Emulsionen.
In der EP-A 876 819 ist die Verwendung von Copolymeren aus mindestens 60 Gew.-% N-Vinylpyrrolidon und Amiden oder Estern mit langkettigen Alkylgruppen beschrieben.
In der EP-A 948 957 ist die Verwendung von Copolymerisaten aus monoethylenisch ungesättigten Carbonsäuren wie beispielsweise Acrylsäure und hydrophob modifizierten Comonomeren wie beispielsweise N-Alkyl- oder N, N-Dialkyl- Amiden ungesättigter Carbonsäuren mit Ce-C3o-Alkylresten beschrieben.
Die bisher bekannten polymeren Solubilisatoren weisen die Nachteile auf, dass sie entweder keine stabilen festen Lösungen ausbilden oder zu hygroskopisch sind. Aus- serdem lassen sie noch Raum für Verbesserungen, was die Solubilisierung in wässri- gen Systemen betrifft.
Es bestand die Aufgabe, neue Solubilisatoren für pharmazeutische, kosmetische, lebensmitteltechnische sowie agrotechnische Anwendungen bereitzustellen.
Diese Aufgabe wurde gelöst durch Copolymere, enthaltend
a) 60 bis 99 Gew.-% mindestens eines Monomeren ausgewählt aus der Gruppe der N-Vinyllactame und N-Vinylamide,
b) 1 bis 40 Gew.-% mindestens eines Monomeren ausgewählt aus der Gruppe der Vinylester von aliphatischen verzweigten C8-C3o-Carbonsäuren,
c) 0 bis 30 Gew.-% Vinylacetat,
d) 0 bis 39 Gew.-% mindestens eines weiteren radikalisch copolymerisierbaren Monomeren,
wobei sich die Gew.-% Angaben der Einzelkomponenten zu 100 Gew.-% addieren, und mit der Maßgabe, dass die Summe der Mengen an b) und c) 1 bis 40 Gew.-% der Gesamtmenge beträgt.
Weiterhin betrifft die Erfindung deren Verwendung als Solubilisatoren für in Wasser schwerlösliche Substanzen sowie entsprechende Zubereitungen.
Als Monomere a) eignen sich N-vinyllactame wie N-Vinylpyrrolidon, N-Vinylpiperidon und N-Vinylcaprolactam oder N-Vinylamide wie N-Methyl-N-Vinylacetamid, N-Vinyl- acetamid und N-Vinylformamid
Bevorzugte Monomere a) sind N-Vinylpyrrolidon und N-Vinylcaprolactam.
Der Anteil des Monomeren a) im Copolymerisat liegt bevorzugt im Bereich von 70 bis 95 Gew.-%, besonders bevorzugt im Bereich von 74 bis 94 Gew.-%.
Als hydrophobe Komponenten b) kommen erfindungsgemäß Vinylester von aliphati- schen verzweigten, insbesondere von gesättigten verzweigten, C8-C3o-Carbonsäuren in Betracht. Dabei eignen sich insbesondere die Vinylester der sogenannten Versatic®- Säuren mit mindestens 8 C-Atomen. Bei den Versatic-Säuren handelt es sich um stark verzweigte gesättigte Monocarbonsäuren mit tertiären Carboxylgruppen, wobei die α-Verzweigungsstelle mindestens eine Methylgruppe trägt und die Zahlenangabe sich auf die Gesamtzahl der Kohlenstoffatome bezieht, sodass Versatic 8 beispielsweise für 2,2-Dimethyl-hexansäure steht. Geeignete Säuren sind weiterhin beispielsweise 2,2-Dimethyl-heptansäure, 2-Ethyl-2-methyl-heptansäure, 2,2-Dimethyl-octansäure, 2-Ethyl-2-methyl-octansäure oder 2,2-Dimethylnonansäure, bevorzugt sind Vinylester der Versatic 9 und Versatic 10-Säuren. Solche Vinylester von Versatic-säuren sind kommerziell erhältlich.
Der Anteil der hydrophoben Monomerbausteine b) im Copolymerisat liegt bevorzugt im Bereich von 5 bis 30 Gew.-%, besonders bevorzugt 10 bis 20 Gew.-%.
Als Monomer c) wird Vinylacetat in Mengen von bis zu 30 Gew.-% eingesetzt. Die Summe der Mengen an Monomeren b) und c) beträgt vorzugsweise 8 bis 30, besonders bevorzugt 10 bis 30 Gew.-%.
Besonders bevorzugt beträgt die Summe der Komponenten a) bis c) 100 Gew.-%.
Weiterhin können die Copolymere die folgenden radikalisch copolymerisierbaren Monomere d) enthalten:
N-C8-C3O-AIkVl- oder N,N-C8-C3o-Dialkyl-substituierte Amide von monoethylenisch ungesättigten C3-C8-Carbonsäuren, wobei es sich bei den Alkylresten um geradkettige oder verzweigte aliphatische oder cycloaliphatische Alkylreste mit 8 bis 30, bevorzugt 8 bis 18 Kohlenstoffatomen handelt. Unter den monoethylenisch ungesättigten Carbonsäuren mit 3 bis 8 C-Atomen kommen hierbei Acrylsäure, Methacrylsäure, Dimethac- rylsäure, Ethacrylsäure, Maleinsäure, Citraconsäure, Methylenmalonsäure, Allylessig- säure, Vinylessigsäure, Crotonsäure, Fumarsäure, Mesaconsäure und Itaconsäure in Betracht, bevorzugt Acrylsäure, Methacrylsäure, Maleinsäure oder Mischungen der genannten Carbonsäuren.
Bevorzugte amidierte Comonomere sind beispielsweise N-Stearylacrylamid, N-Stearyl- methacrylamid, N-(1 -Methyl)undecylacrylamid, N-(1 -Methyl)undecylmethacrylamid, N-Dodecylacrylamid, N-Dodecylmethacrylamid, N-Octylacrylamid, N-Octylmethacryl-
amid, N.N-Dioctylacrylamid, N,N-Dioctylmethacrylamid, N-Cetylacrylamid, N-Cetyl- methacrylamid, N-Dodecylacrylamid, N-Dodecylmethacrylamid, N-Myristylacrylamid, N-Myristylmethacrylamid, N-(2-Ethyl)-hexylacrylamid, N-(2-Ethyl)-hexylmethacrylamid.
Im Falle von Maleinsäureanhydrid als Comonomer kann diese polymeranalog mit N-Alkylaminen durch Ringöffnung zu den entsprechenden Amiden umgesetzt werden.
Als weitere Comonomere d) können monoethylenisch ungesättigte C3-Ce-Carbon- säureester mit einem C8-C3O-AIkOhOl, bevorzugt einem C8-Ci8-AIkOhOl, eingesetzt wer- den.
Besondere Bedeutung kommt hierbei den Acryl- bzw. Methacrylsäureestem mit Fettalkoholen einer Kettenlänge von 8 bis 18 Kohlenstoffatomen, wobei die Alkylreste verzeigt oder unverzweigt sein können, zu.
Insbesondere seien hier genannt: Octylacrylat, 2-Ethylhexylacrylat, Nonylacrylat, Decy- lacrylat, Laurylacrylat, Myristylacrylat, Cetylacrylat, Stearylacrylat, Oleylacrylat, Behenylacrylat, Octylmethacrylat, 2-Ethylhexylmethacrylat, Nonylmethacrylat, Decylmethacrylat, Laurylmethacrylat, Myristylmethacrylat, Cetylmethacrylat, Stearylmethacrylat, Oleylmethacrylat, Behenylmethacrylat, tert.-Butylcyclohexylacrylat.
Als weitere zusätzliche Komponente d) können Vinylester langkettiger aliphatischer, gesättigter oder ungesättigter, unverzweigter C8-C3o-Carbonsäuren, wie z.B. Capryl- säure, Caprinsäure, Laurinsäure, Myristinsäure, Palmitinsäure, Stearinsäure, Arachin- säure, Behensäure, Lignocerinsäure, Cerotinsäure sowie Melissensäure eingesetzt werden.
Ferner können als Monomere d) C8-C3o-Alkyl-Vinylether, bevorzugt C8-Ci8-Alkyl- Vinylether einpolymerisiert werden.Als bevorzugte Alkylreste der Vinylether seien ver- zweigte oder unverzweigte Ce-Cie-Alkylketten wie z.B. n-Octyl, 2-Ethylhexyl, n-Nonyl, n-Decyl, n-Undecyl, n-Dodecyl, n-Tridecyl, n-Tetradecyl, n-Pentadecyl, n-Hexadecyl, n-Heptadecyl sowie n-Octadecyl genannt.
Als zusätzliche radikalisch copolymerisierbare Monomere d) kommen in Betracht:
Monoethylenisch ungesättigte Carbonsäuren mit 3 bis 8 C-Atomen, wie z.B. Acrylsäu- re, Methacrylsäure, Dimethacrylsäure, Ethacrylsäure, Maleinsäure, Citraconsäure, Me- thylenmalonsäure, Allylessigsäure, Crotonsäure, Fumarsäure, Mesaconsäure und Ita- consäure.
Aus dieser Gruppe von Monomeren werden bevorzugt Acrylsäure, Methacrylsäure oder Mischungen der genannten Carbonsäuren verwendet.
Die monoethylenisch ungesättigten Carbonsäuren können als freie Säure, als Anhydride sowie in partiell oder vollständig neutralisierter Form bei der Copolymerisation eingesetzt werden.
Für die Neutralisation der oben genannten Carbonsäuren verwendet man vorzugsweise Alkalimetall- oder Erdalkalimetallbasen, Ammoniak oder Amine, bevorzugt Natronlauge, Kalilauge, Soda, Pottasche, Natriumhydrogencarbonat, Magnesiumoxid, Calci um hydroxid, Calciumoxid, gasförmiges oder wässriges Ammoniak, Triethylamin, Ethanolamin, Diethanolamin, Triethanolamin, Morpholin, Diethylentriamin oder Tetra- ethylenpentamin.
Weitere geeignete Comonomere d) sind beispielsweise monoethylenisch ungesättigte C3-Ce-Carbonsäureester von kurzkettigen Ci-C4-Alkoholen oder Nitrile in Anteilen von 0 bis 5 Mol-% für die Polymerisation zu verwenden.
Beispielhaft seien genannt: Acrylsäuremethylester, Acrylsäureethylester, Methacrylsäu- remethylester, Methacrylsäureethylester, Hydroxyethylacrylat, Hydroxypropylacrylat, Hydroxybutylacrylat, Hydroxyethylmethacrylat, Hydroxypropylmethacrylat, Hydroxyiso- butylacrylat, Hydroxyisobutylmethacrylat, Maleinsäuremonomethylester, Maleinsäure- dimethylester, Maleinsäuremonoethylester, Maleinsäurediethylester, 2-Ethylhexyl- acrylat,2-Ethylhexylmethacrylat, N-Dimethylacrylamid, N-tert.-butylacrylamid, Acrylnitril, Methacrylnitril, Dimethylaminoethylacrylat, Diethyl-aminoethylacrylat, Dimethylami- noethylmethacrylat, Diethylaminoethylmethacrylat sowie die Salze der zuletzt genann- ten Monomeren mit Carbonsäuren oder Mineralsäuren sowie die quaternierten Produkte.
Weitere geeignete Monomere d) sind beispielsweise:
- Acrylamidoglycolsäure, Vinylsulfonsäure, Allylsulfonsäure, Methallylsulfonsäure, Styrolsulfonsäure, Acrylsäure-(3-sulfo-propyl)ester, Methacrylsäure(3-sulfopro- pyl)ester und Acrylamidomethylpropansulfonsäure;
Phosphonsäuregruppen enthaltende Monomere, wie Vinylphosphonsäure, Allylphosphonsäure und Acrylamidomethanpropanphosphonsäure;
Es ist selbstverständlich auch möglich, Mischungen der genannten Monomeren einzusetzen.
Bevorzugte Monomere d) sind Acrylsäure, Methacrylsäure, Methylmethacrylat, Ethyl- acrylat, Vinyllaurat, Stearylmethacrylat und Laurylacrylat.
Der Anteil der Monomerbausteine d) im Copolymer liegt bevorzugt im Bereich von 0 bis 5 Gew.-%, besonders bevorzugt beträgt er 0 Gew.-%.
Die erfindungsgemäß verwendeten Copolymere können K-Werte nach Fikentscher, gemessen 1 Gew.-%ig in Ethanol, von 10 bis 200, bevorzugt 15 bis 100, besonders bevorzugt 20 bis 50, aufweisen.
Die Copolymere werden dadurch hergestellt, dass man die entsprechenden Monomeren radikalisch polymerisiert.
Die Herstellung erfolgt nach bekannten Verfahren, z.B. der Lösungs-, Fällungs-, oder durch umgekehrte Suspensions- Polymerisation unter Verwendung von Verbindungen, die unter den Polymerisationsbedingungen Radikale bilden.
Die Polymerisationstemperaturen liegen üblicherweise in dem Bereich von 30 bis 200, vorzugsweise 40 bis 1100C. Geeignete Initiatoren sind beispielsweise Azo- und Pero- xyverbindungen sowie die üblichen Redoxinitiatorsysteme, wie Kombinationen aus Wasserstoffperoxid und reduzierend wirkenden Verbindungen, z.B. Natriumsulfit, Natriumbisulfit, Natriumformaldehydsulfoxilat und Hydrazin.
Als Reaktionsmedium finden alle üblichen Lösungsmittel Verwendung, in denen die Monomere löslich sind. Vorzugsweise werden Wasser oder alkoholische Lösungsmittel wie z.B. Methanol, Ethanol, n-Propanol oder Isopropanol oder Gemische solcher Alkohole mit Wasser, eingesetzt.
Um zu gewährleisten, dass die Reaktion zu homogenen Produkten führen, ist es vorteilhaft, die Monomere und den Starter separat der Reaktionslösung zuzuführen. Dies kann beispielsweise in Form von getrennten Zuläufen für die einzelnen Reaktionspartner erfolgen.
Die Polymerisation kann auch in Gegenwart üblicher Regler durchgeführt werden, falls relativ niedrigere Molekulargewichte eingestellt werden sollen.
Der Feststoffgehalt der erhaltenen organischen Lösung beträgt üblicherweise 20 bis 60 Gew.-%, insbesondere 25 bis 40 Gew.-%.
Ein für die Polymerisation verwendetes nicht-wässriges Lösungsmittel kann anschließend mittels Wasserdampfdestillation entfernt und gegen Wasser ausgetauscht werden.
Die wässrigen Lösungen der Copolymere können durch verschiedene Trocknungsverfahren wie z.B. Sprühtrocknung, Fluidized Spray Drying, Walzentrocknung oder
Gefriertrocknung in Pulverform überführt werden, aus der sich durch Redispergieren in Wasser erneut eine wässrige Lösung herstellen lässt.
Anwendungen:
Die erfindungsgemäß zu verwendenden Copolymere lassen sich grundsätzlich auf allen Gebieten einsetzen, bei denen in Wasser nur schwerlösliche oder unlösliche Substanzen entweder in wässrigen Zubereitungen zum Einsatz kommen sollen oder ihre Wirkung in wässrigem Milieu entfalten sollen. Die Copolymere finden demgemäss Ver- wendung als Solubilisatoren von in Wasser schwerlöslichen Substanzen, insbesondere biologisch aktiven Substanzen.
Der Begriff „in Wasser schwerlöslich" umfasst erfindungsgemäß auch praktisch unlösliche Substanzen und bedeutet, dass für eine Lösung der Substanz in Wasser bei 200C mindestens 30 bis 100 g Wasser pro g Substanz benötigt wird. Bei praktisch unlöslichen Substanzen werden mindestens 10.000 g Wasser pro g Substanz benötigt.
Im Sinne der vorliegenden Erfindung sind unter in Wasser schwerlösliche biologisch aktive Substanzen pharmazeutische Wirkstoffe für Mensch und Tier, kosmetische oder agrochemische Wirkstoffe oder Nahrungsergänzungsmittel oder diätetische Wirkstoffe zu verstehen.
Weiterhin kommen als zu solubilisierende schwerlösliche Substanzen auch Farbstoffe wie anorganische oder organische Pigmente in Betracht.
Durch die vorliegende Erfindung werden insbesondere amphiphile Verbindungen für die Anwendung als Lösungsvermittler für pharmazeutische und kosmetische Zubereitungen sowie für Lebensmittelzubereitungen zur Verfügung gestellt. Sie besitzen die Eigenschaft, schwer lösliche Wirkstoffe auf dem Gebiet der Pharmazie und Kosmetik, schwerlösliche Nahrungsergänzungsmittel, beispielsweise Vitamine und Carotinoide aber auch schwerlösliche Wirkstoffe für den Einsatz in Pflanzenschutzmitteln sowie veterinärmedizinische Wirkstoffe zu solubilisieren.
Überraschend wurde bei den beanspruchten Verbindungen ein gutes Solubilisations- vermögen für pharmazeutische und kosmetische Wirkstoffe gefunden. Ferner werden mit den beanspruchten Verbindungen Anwendungen erhalten, die sich durch eine sehr geringe Hämolyserate, einer nebenwirkungsfreien Verträglichkeit nach parenteraler, oraler und topischer Applikation auf Haut- und Schleimhaut auszeichnen. Die Verbindungen besitzen insbesondere keine Nebenwirkungen durch Wechselwirkungen mit Blutkörperchenmembranen. Nach parenteraler Applikation findet keine bzw. nur eine geringe Histaminfreisetzung statt. Das Molekulargewicht der Solubilisatoren kann ge- wünschtenfalls so eingestellt werden, dass die Solubilisatoren nierengängig sind.
Solubilisatoren für Kosmetik:
Erfindungsgemäßen können die Copolymere als Solubilisatoren in kosmetischen For- mulierungen eingesetzt werden. Beispielsweise eignen sie sich als Solubilisatoren für kosmetische Öle. Sie besitzen ein gutes Solubilisiervermögen für Fette und Öle, wie Erdnußöl, Jojobaöl, Kokosnußöl, Mandelöl, Olivenöl, Palmöl, Ricinusöl, Sojaöl oder Weizenkeimöl oder für etherische Öle wie Latschenkiefernöl, Lavendelöl, Rosmarinöl, Fichtennadelöl, Kiefemnadelöl, Eukalyptusöl, Pfefferminzöl, Salbeiöl, Bergamottöl, Terpentinöl, Melissenöl, Salbeiöl, Wacholderöl, Zitronenöl, Anisöl, Kardamonöl; Pfefferminzöl, Campheröl etc. oder für Mischungen aus diesen Ölen.
Weiterhin können die erfindungsgemäßen Polymere als Solubilisatoren für in Wasser schwerlösliche oder unlösliche UV-Absorber wie beispielsweise 2-Hydroxy-4-methoxy- benzophenon (Uvinul® M 40, Fa. BASF), 2,2',4,4'-Tetrahydroxybenzophenon (Uvinul® D 50), 2,2'-Dihydroxy-4,4'-dimethoxybenzophenon (Uvinul® D49), 2,4-Dihydroxybenzo- phenon (Uvinul® 400), 2-Cyano-3,3-diphenylacrylsäure-2'-ethylhexylester (Uvinul® N 539), 2,4,6-Trianilino-p-(carbo-2'-ethylhexyl-1 '-oxi)-1 , 3,5-triazin (Uvinul® T 150), 3-(4- Methoxybenzyliden)-campher (Eusolex® 6300, Fa. Merck), N,N-Dimethyl-4-amino- benzoesäure-2-ethylhexylester (Eusolex® 6007), Salicylsäure-3,3,5-trimethylcyclo- hexylester, 4-lsopropyl-dibenzoylmethan (Eusolex® 8020), p-Methoxyzimtsäure-2- ethylhexylester und p-Methoxyzimtsäure-2-isoamylester sowie Mischungen davon verwendet werden.
Gegenstand der vorliegenden Erfindung sind daher auch kosmetische Zubereitungen, die mindestens einen der erfindungsgemäßen Copolymere der eingangs genannten Zusammensetzung als Solubilisatoren enthalten. Bevorzugt sind solche Zubereitungen, die neben dem Solubilisator einen oder mehrere schwerlösliche kosmetische Wirkstoffe, beispielsweise die oben genannten Öle oder UV-Absorber enthalten.
Bei diesen Formulierungen handelt es sich um Solubilisate auf Wasser oder Wasser/Alkohol Basis. Die erfindungsgemäßen Solubilisatoren werden im Verhältnis von 0,2:1 bis 20:1 , bevorzugt 1 :1 bis 15:1 , besonders bevorzugt 2:1 bis 12:1 zum schwerlöslichen kosmetischen Wirkstoff eingesetzt.
Der Gehalt an erfindungsgemäßem Solubilisator in der kosmetischen Zubereitung liegt, abhängig vom Wirkstoff, im Bereich von 1 bis 50 Gew.-%, bevorzugt 3 bis 40 Gew.-%, besonders bevorzugt 5 bis 30 Gew.-%.
Zusätzlich können dieser Formulierung weitere Hilfsstoffe zugesetzt werden, beispielsweise nichtionische, kationische oder anionische Tenside wie Alkylpolyglycoside, Fettalkoholsulfate, Fettalkoholethersulfate, Alkansulfonate, Fettalkoholethoxilate, Fettalko-
holphosphate, Alkylbetaine, Sorbitanester, POE-Sorbitanester, Zuckerfettsäureester, Fettsäurepolyglycerinester, Fettsäurepartialglyceride, Fettsäurecarboxylate, Fettalko- holsulfosuccinate, Fettsäuresarcosinate, Fettsäureisethionate, Fettsäuretaurinate, Zitronensäureester, Silikon-Copolymere, Fettsäurepolyglykolester, Fettsäureamide, Fett- säurealkanolamide, quartäre Ammoniumverbindungen, Alkylphenoloxethylate, Fett- aminoxethylate, Cosolventien wie Ethylenglykol, Propylenglykol, Glycerin u.a..
Als weitere Bestandteile können natürliche oder synthetische Verbindungen, z.B. Lanolinderivate, Cholesterinderivate, Isopropylmyristat, Isopropylpalmitat, Elektrolyte, Farb- Stoffe, Konservierungsmittel, Säuren (z.B. Milchsäure, Zitronensäure) zugesetzt werden.
Diese Formulierungen finden beispielsweise in Badezusatzpräparaten wie Badeölen, Rasierwässern, Gesichtswässern, Haarwässern, Eau de Cologne, Eau de Toilette so- wie in Sonnenschutzmitteln Verwendung. Weiterhin finden sie Anwendung im Bereich Oral Care, beispielsweise in Zahnpasten, Mundwässern oder Mundcremes.
Beschreibung der Solubilisierungsmethode:
Bei der Herstellung der Solubilisate für kosmetische Formulierugen können die erfindungsgemäßen Copolymere als 100 %ige Substanz oder bevorzugt als wässrige Lösung eingesetzt werden.
Üblicherweise wird der Solubilisator in Wasser gelöst und mit dem jeweils zu verwen- denden schwerlöslichen kosmetischen Wirkstoff intensiv vermischt.
Es kann aber auch der Solubilisator mit dem jeweils zu verwendenden schwerlöslichen kosmetischen Wirkstoff intensiv vermischt werden und anschließend unter ständigem Rühren mit demineralisiertem Wasser versetzt werden.
Solubilisatoren für pharmazeutische Anwendungen:
Die beanspruchten Copolymerisate eignen sich ebenso für die Verwendung als Solubilisator in pharmazeutischen Zubereitungen jeder Art, die dadurch gekennzeichnet sind, dass sie einen oder mehrere in Wasser schwer lösliche oder wasserunlösliche Arzneistoffe sowie Vitamine und/oder Carotinoide enthalten können. Insbesondere handelt es sich dabei um wässrige Lösungen bzw. Solubilisate zur oralen oder besonders bevorzugt zur parenteralen Applikation, wie z.B. Injektionslösungen zur intravenösen, intramuskulären oder subkutaner oder intraperitonealer Applikation.
Des weiteren eignen sich die beanspruchten Copolymere zum Einsatz in oralen Darreichungsformen wie Tabletten, Kapseln, Pulvern, Lösungen. Hier können Sie den schwerlöslichen Arzneistoff mit einer erhöhten Bioverfügbarkeit zur Verfügung stellen.
Bei der parenteralen Applikation können neben Solubilisaten auch Emulsionen, beispielsweise Fettemulsionen eingesetzt werden. Auch für diesen Zweck eignen sich die beanspruchten Copolymere um einen schwerlöslichen Arzneistoff zu verarbeiten.
Pharmazeutische Formulierungen der oben genannten Art können durch Verarbeiten der beanspruchten Copolymere mit pharmazeutischen Wirkstoffen nach herkömmlichen Methoden und unter Einsatz bekannter und neuer Wirkstoffe erhalten werden.
Die erfindungsgemäße Anwendung kann zusätzlich pharmazeutische Hilfsstoffe und/oder Verdünnungsmittel enthalten. Als Hilfsstoffe werden Cosolventien, Stabilisa- toren, Konservierungsmittel besonders aufgeführt.
Die verwendeten pharmazeutischen Wirkstoffe sind in Wasser unlösliche bzw. wenig lösliche Substanzen. Gemäß DAB 9 (Deutsches Arzneimittelbuch) erfolgt die Einstufung der Löslichkeit pharmazeutischer Wirkstoffe wie folgt: wenig löslich (löslich in 30 bis 100 Teilen Lösungsmittel); schwer löslich (löslich in 100 bis 1000 Teilen Lösungsmittel); praktisch unlöslich (löslich in mehr als 10000 Teilen Lösungsmittel). Die Wirkstoffe können dabei aus jedem Indikationsbereich kommen.
Als Beispiele seien hier Benzodiazepine, Antihypertensiva, Vitamine, Cytostatika - ins- besondere Taxol, Anästhetika, Neuroleptika, Antidepressiva, Antibiotika, Antimykotika, Fungizide, Chemotherapeutika, Urologika, Thrombozytenaggregationshemmer, Sulfonamide, Spasmolytika, Hormone, Immunglobuline, Sera, Schilddrüsentherapeutika, Psychopharmaka, Parkinsonmittel und andere Antihyperkinetika, Ophthalmika, Neuropathiepräparate, Calciumstoffwechselregulatoren, Muskelrelaxantia, Narkosemittel, Lipidsenker, Lebertherapeutika, Koronarmittel, Kardiaka, Immuntherapeutika, regulatorische Peptide und ihre Hemmstoffe, Hypnotika, Sedativa, Gynäkologika, Gichtmittel, Fibrinolytika, Enzympräparate und Transportproteine, Enzyminhibitoren, Emetika, Durchblutungsfördernde Mittel, Diuretika, Diagnostika, Corticoide, Cholinergika, GaI- lenwegstherapeutika, Antiasthmatika, Broncholytika, Betarezeptorenblocker, Calcium- antagonisten, ACE-Hemmer, Arteriosklerosemittel, Antiphlogistika, Antikoagulantia, Antihypotonika, Antihypoglykämika, Antihypertonika, Antifibrinolytika, Antiepileptika, Antiemetika, Antidota, Antidiabetika, Antiarrhythmika, Antianämika, Antiallergika, Anthelmintika, Analgetika, Analeptika, Aldosteronantagonisten, Abmagerungsmittel genannt.
Eine mögliche Herstellvariante ist das Auflösen des Solubilisators in der wässrigen Phase, gegebenenfalls unter leichtem Erwärmen und das anschließende Lösen des
Wirkstoffs in der wässrigen Solubilisatorlösung. Das gleichzeitige Auflösen von Solubi- lisator und Wirkstoff in der wässrigen Phase ist ebenfalls möglich.
Die Verwendung der erfindungsgemäßen Copolymere als Lösungsvermittler kann bei- spielsweise auch in der Weise erfolgen, dass der Wirkstoff in dem Solubilisator, gegebenenfalls unter Erwärmen, dispergiert wird und unter Rühren mit Wasser vermischt wird.
Gegenstand der Erfindung sind daher auch pharmazeutische Zubereitungen, die min- destens einen der erfindungsgemäßen Copolymere als Solubilisator enthalten. Bevorzugt sind solche Zubereitungen, die neben dem Solubilisator einen in Wasser schwerlöslichen oder wasserunlöslichen pharmazeutischen Wirkstoff, beispielsweise aus den oben genannten Indikationsgebieten enthalten.
Besonders bevorzugt sind von den oben genannten pharmazeutischen Zubereitungen solche, bei denen es sich um parenteral applizierbare Formulierungen handelt.
Der Gehalt an erfindungsgemäßem Solubilisator in der pharmazeutischen Zubereitung liegt, abhängig vom Wirkstoff, im Bereich von 1 bis 50 Gew.-%, bevorzugt 3 bis 40 Gew.-%, besonders bevorzugt 5 bis 30 Gew.-%.
Solubilisatoren für Lebensmittelzubereitungen:
Neben der Anwendung in der Kosmetik und Pharmazie eignen sich die erfindungsge- mäßen Copolymeren auch als Solubilisatoren im Lebensmittelbereich für schwer wasserlösliche oder wasserunlösliche Nähr-, Hilfs- oder Zusatzstoffe, wie z.B. fettlösliche Vitamine oder Carotinoide. Als Beispiele seien klare, mit Carotinoiden gefärbte Getränke genannt.
Solubilisatoren für Pflanzenschutzzubereitungen:
Die Anwendung der erfindungsgemäßen Copolymere als Solubilisatoren in der Agrochemie kann u.a. Formulierungen umfassen, die Pestizide, Herbizide, Fungizide oder Insectizide enthalten, vor allem auch solche Zubereitungen von Pflanzenschutzmitteln, die als Spritz- oder Gießbrühen zum Einsatz kommen.
Die erfindungsgemäßen wasserlöslichen Copolymere zeichnen sich durch eine besonders gute solubilisierende Wirkung aus. Sie eignen sich auch hervorragend zur Herstellung von stabilen festen Lösungen.
In den folgenden Beispielen wird die Herstellung und Verwendung der erfindungsgemäßen Copolymere näher erläutert.
Beispiele
Die Abkürzung VEOVA wird in den folgenden Beispielen für Vinylester von Versatic- Säuren verwendet. Die Ziffer hinter der Abkürzung bezeichnet die Anzahl der C-Atome. Die Monomeren sind kommerziell erhältlich.
Zur Herstellung der Polymere wurde die folgende Apparatur verwendet:
2 I HWS Topf mit Wasserbad, Ankerrührer und Thermometer. Der HWS Topf verfügte über Anschlüsse für 3 Zuläufe, einen Rückflusskühler sowie über ein Einleitungsrohr, für die Einleitung von Stickstoff oder Wasserdampf.
Beispiel 1
Herstellung von Copolymeren aus N-Vinylpyrrolidon / Vinylacetat / VEOVA 9 (Gewichtsverhältnis 70 / 15 /15 )
In der Apparatur wurden in der angegebenen Reihenfolge 12 g Isopropanol und 70 g des Zulaufs 1 bei 180 U/min gemischt. Ein leichter Stickstoffstrom wurde ständig durch die Apparatur geleitet. Gleichzeitig wurde auf eine Innentemperatur von 700C geheizt. Bei 68°C wurde die Teilmenge Zulauf 2 (1 ,0 g) zugegeben und 10 Minuten anpolymeri- siert.
Danach wurde der Zulauf 1 und 2 gestartet. Der Zulauf 1 wurde in 4 Stunden zudosiert. 37 g des Zulaufs 2 wurden in 5 Stunden zudosiert. Nach Beendigung des Zulaufs 1 wurde der Zulauf 3 in 1 Stunde zudosiert. Nach Beendigung der Teilmenge des Zulaufs 2 wurde noch eine Stunde bei 700C nachpolymerisiert. Dann wurde auf eine Innentem- peratur von 75°C aufgeheizt. Parallel mit dem Aufheizvorgang wurde der Zulauf 2 (Restemenge 52 g) gestartet und in 2 Stunden zudosiert. Nach Ende von Zulauf 2 wurde noch 2 Stunden bei 75°C nachpolymerisiert.
Anschließend wurden 400 g vollentsalztes Wasser zugegeben und Wasserdampf in die Polymerlösung für 3 Stunden eingeleitet.
Man erhielt eine gelbliche, viskose Lösung mit einem Feststoffgehalt von 32 Gew.-%. Der K-Wert betrug 33 (gemessen 1 gew.-%ig in Ethanol).
Beispiel 2
Herstellung von Copolymeren aus N-Vinylpyrrolidon / VEOVA 9 (Gewichtsverhältnis 80 / 20)
In der Apparatur wurden in der angegebenen Reihenfolge 12 g Isopropanol und 78 g des Zulaufs 1 bei 180 U/min gemischt. Ein leichter Stickstoffstrom wurde ständig durch die Apparatur geleitet. Gleichzeitig wurde auf eine Innentemperatur von 75°C geheizt. Bei 73°C wurde die Teilmenge Zulauf 2 (5,0 g) zugegeben und 10 Minuten anpolymeri- siert.
Danach wurde der Zulauf 1 und 2 gestartet. Der Zulauf 1 wurde in 4 Stunden, der Zulauf 2 in 6 Stunden zudosiert. Dann wurde noch zwei Stunden nachpolymerisiert.
Anschließend wurden 400 g vollentsalztes Wasser zugegeben und Wasserdampf in die Polymerlösung für ca. 3 Stunden eingeleitet.
Man erhält eine gelbliche, viskose Lösung mit einem Feststoffgehalt von 27 Gew.-%. Der K-Wert beträgt 35 (gemessen 1 %ig in Ethanol).
Beispiel 3
Herstellung von Copolymeren aus N-Vinylpyrrolidon / VEOVA 9 (Gewichtsverhältnis 90 / 10)
In der Apparatur wurden in der angegebenen Reihenfolge 12 g Isopropanol und 78 g des Zulaufs 1 bei 180 U/min gemischt. Ein leichter Stickstoffstrom wurde ständig durch die Apparatur geleitet. Gleichzeitig wurde auf eine Innentemperatur von 75°C geheizt. Bei 73°C wurde die Teilmenge Zulauf 2 (5,0 g) zugegeben und 10 Minuten anpolymeri- siert.
Danach wurde der Zulauf 1 und 2 gestartet. Der Zulauf 1 wurde in 4 Stunden, der Zu- lauf 2 in 6 Stunden zudosiert. Dann wurde noch zwei Stunden nachpolymerisiert.
Anschließend wurden 400 g vollentsalztes Wasser zugegeben und Wasserdampf in die Polymerlösung für ca. 3 Stunden eingeleitet.
Man erhält eine gelbliche, viskose Lösung mit einem Feststoffgehalt von 34 Gew.-%.
Der K-Wert betrug 35 (gemessen 1 gew.-%ig in Ethanol).
Beispiel 4
Herstellung von Copolymeren aus N-Vinylpyrrolidon / N-Vinylcaprolactam / VEOVA 10 (Gewichtsverhältnis 70 / 20 / 10)
In der Apparatur wurden in der angegebenen Reihenfolge 12 g Isopropanol und 70 g des Zulaufs 1 bei 180 U/min gemischt. Ein leichter Stickstoffstrom wurde ständig durch die Apparatur geleitet. Gleichzeitig wurde auf eine Innentemperatur von 700C geheizt. Bei 68°C wurde die Teilmenge Zulauf 2 (1 ,0 g) zugegeben und 10 Minuten anpolymeri- siert.

Nach Ende von Zulauf 2 wurde noch 2 Stunden bei 75°C nachpolymerisiert.
Anschließend wurden 400 g vollentsalztes Wasser zugegeben und Wasserdampf in die Polymerlösung für ca. 3 Stunden eingeleitet.
Man erhielt eine gelbliche, viskose Lösung mit einem Feststoffgehalt von 32 Gew.-%. Der K-Wert betrug 47 (gemessen 1 gew.-%ig in Ethanol).

Herstellung von festen Lösungen: Allgemeine Vorschrift
Zur Herstellung des Polymer-Wirkstoff-Gemischs wurden der Wirkstoff und das Polymer im Gewichtsverhältnis 1 :1 in ein geeignetes Glasgefäß eingewogen (jeweils 2g)
und anschließend 16 ml Dimethylfomnamid als Lösungsmittel hinzugefügt. Der Ansatz wurde bei 200C 24 Stunden auf einem Magnetrührer gerührt. Die Lösung wurde anschließend mit Hilfe eines 120 μm-Rakels auf einer Glasplatte ausgezogen. Diese wurde 0,5 Stunden bei RT im Abzug getrocknet und anschließend im Trockenschrank bei 500C und 10 mbar für weitere 0,5 Stunden getrocknet, um das Lösungsmittel quantitativ zu entfernen. Die Proben wurden anschließend visuell begutachtet. Wenn der Wirkstoff nach 7 Tagen nicht auskristallisierte, hatte sich eine stabile feste Lösung ausgebildet.
Herstellung von Solubilisaten
In ein Becherglas wurden 2g des Copolymers eingewogen. Anschließend wurde dem Ansatz jeweils ein Arzneistoff wie folgt zugewogen, um eine übersättigte Lösung zu erhalten. (Falls sich die eingewogene Masse im Medium auflöste, wurde die Einwaage bis zur Ausbildung eines Bodensatzes erhöht).
Anschließend wurde Phosphatpuffer pH 7,0 hinzugegeben, bis Solubilisator und Phosphatpuffer im Gewichtsverhältnis von 1 :9 vorlagen. Mit Hilfe eines Magnetrührers wur- de dieser Ansatz bei 200C 72 Stunden gerührt. Danach erfolgte mindestens eine 1 stündige Ruhezeit. Nach der Filtration des Ansatzes wurde dieser photometrisch vermessen und der Gehalt an Wirkstoff bestimmt.
Ergebnisse: Gehalt an solubilisiertem Wirkstoff [g/100ml]
Claims
1. Copolymere, enthaltend
a) 60 bis 99 Gew.-% mindestens eines Monomeren ausgewählt aus der
Gruppe bestehend aus N-Vinyllactamen und N-Vinylamiden,
b) 1 bis 40 Gew.-% mindestens eines Monomeren ausgewählt aus der Gruppe der Vinylester von aliphatischen verzweigten C8-C3o-Carbonsäuren,
c) 0 bis 30 Gew.-% Vinylacetat,
d) 0 bis 39 Gew.-% mindestens eines weiteren radikalisch copolymerisierba- ren Monomeren,
wobei sich die Gew.-% Angaben der Einzelkomponenten a) bis d) zu 100 % addieren, und mit der Maßgabe, dass die Summe der Mengen von b) und c) 1 bis 40 Gew.-% der Gesamtmenge beträgt.
2. Copolymere nach Anspruch 1, wobei die Copolymeren
a) 70 bis 95 Gew.-% mindestens eines Monomeren ausgewählt aus der Gruppe bestehend aus N-Vinyllactamen und N-Vinylamiden,
b) 5 bis 30 Gew.-% mindestens eines Monomeren ausgewählt aus der Gruppe der Vinylester von aliphatischen verzweigten C8-C3o-Carbonsäuren,
c) 0 bis 25 Gew.-% Vinylacetat,
d) 0 bis 25 Gew.-% mindestens eines weiteren radikalisch copolymerisierba- ren Monomeren,
enthalten.
3. Copolymere nach den Ansprüchen 1 und 2, wobei die Copolymeren einen K- Wert von 10 bis 200 aufweisen.
4. Copolymere nach einem der Ansprüche 1 bis 3, wobei die Monomere a) ausgewählt sind aus der Gruppe bestehend aus N-Vinylpyrrolidon, N-Vinylcaprolactam
5. Copolymere nach einem der Ansprüche 1 bis 3, wobei die Copolymere als Monomere b) Vinylester von Versatic-Säuren enthalten.
6. Copolymere nach einem der Ansprüche 1 bis 4, wobei die Copolymeren als Monomere b)Vinylester von Cg-Cio-Versatic-Säuren enthalten.
7. Copolymere nach einem der Ansprüche 1 bis 5, wobei die Summe von b) und c) 8 bis 30 Gew.-% beträgt.
8. Copolymere nach einem der Ansprüche 1 bis 6, wobei die Copolymeren bis 5 Gew.-% an Monomeren d)enthalten.
9. Copolymere nach einem der Ansprüche 1 bis 7, wobei die Summe an Monomeren a) bis c) 100 Gew.-% beträgt.
10. Copolymere nach einem der Ansprüche 1 bis 8 , wobei die Copolymeren als Mo- nomere a)N-Vinylpyrrolidon oder N-Vinylcaprolactam oder Mischungen davon enthalten.
11. Copolymere nach einem der Ansprüche 1 bis 9, erhalten durch radikalisch initiierte Polymerisation der Monomeren a) bis d).

29. Verfahren nach Anspruch 28, dadurch gekennzeichnet, dass man die Komponenten a) bis c) getrennt der Polymerisationsreaktion zuführt.
