"Ethercarbonsäureester von Sterolen bzw. Stanolen"
Gebiet der Erfindung
Die vorliegende Erfindung befindet sich auf dem Gebiet der Ester von Sterolen und/oder Stanolen und betrifft neue Ethercarbonsäureester, ein Verfahren zu deren Herstellung sowie die Verwendung dieser Ester.
Stand der Technik
In den letzten Jahren wurde den Sterolen, insbesondere den Phytosterolen, verstärkte Aufmerksamkeit zu teil. So werden diese in hohem Maße bei der Fermentation zu Steroid-Zwischenstufen wie Androstendion eingesetzt, das wiederum durch meist chemische Modifikationen in pharmazeutisch wirksame Steroide wie Testeron, Estradiol, Testolacton oder Cortison überführt wird. Weiterhin haben Phytosterole die vorteilhafte Eigenschaft, den Cholesterinspiegel im Blut des Menschen zu senken, weshalb sie und ihre Fettsäureester als Nahrungsmittelzusatzstoff zur Anwendung kommen.
Als eines der wesentlichen Probleme bei der Verwendung der Sterole und der durch Hydrierung erhältlichen Stanole hat sich deren äußerst geringe Löslichkeit in Wasser und nur geringe Löslichkeit in Fetten erwiesen. Ein Weg zur Herstellung wässriger Dispersionen sieht die Partikelgrößenreduzierung der Sterole mittels verschiedenster Hochdruckhomogenisatoren vor. So ist aus der internationalen Anmeldung WO 00/45648 die Herstellung von Mikropartikeln von Phytosterolen unter anderem auch in einem wässrigen Medium durch Einsatz starker Scherkräfte erzeugender Apparaturen wie Kolloidmühlen bekannt.
Eine andere Möglichkeit wässrige Dispersionen von Sterolen herzustellen, ist der Einsatz von Emulga- to en mit meist anschließender Homogenisierung, beispielsweise gemäß dem Europäischen Patent EP- B- 897 671.
Sterolderivate, die eine verbesserte Löslichkeit in Fetten aufweisen, sind aus der Europäischen Patentanmeldung EP-A- 195311 bekannt, die durch eine Veresterung von Phytosterolen und verzweigten aliphatischen Alkoholen mit Fettsäuren oder Fettsäureestern in Gegenwart von Lipasen hergestellt werden können. Die Deutsche Patentanmeldung DE-A- 2 035 069 offenbart klare Brat- und Salatöle, enthaltend Carbonsäureester von Phytosterolen hergestellt durch Acylation mittels Perchlorsäure als Katalysator.
Aus der internationalen Patentanmeldung WO 92/19640 wiederum sind ß-Sitostanolfettsäureester bekannt, die durch Umesterung von ß-Sitostanol mit Fettsäureester in Gegenwart von Umesterungskata- lysatoren hergestellt werden.
Aufgabe der vorliegenden Anmeldung war es, Sterolderivate und/oder Stanolderivate bereitzustellen, die in Wasser in möglichst hohen Mengen und möglichst ohne aufwendige Hochdruckhomogenisierungsapparate dispergierbar sind, um breite Anwendungsmöglichkeiten zu erschließen. Gleichzeitig sollten die Derivate selbstdispergierbar sein, d.h. die Anwesenheit von Emulgatoren zur Herstellung wässriger Dispersionen sollte nicht notwendig, sondern lediglich fakultativ sein. Weiterhin sollten die Derivate möglichst einen niedrigeren Schmelzpunkt als die Sterole und/oder Stanole selber aufweisen, beispielsweise um fermentative Folgereaktionen zu erleichtern. Schließlich galt es Derivate zur Verfügung zu stellen, die in wässrigen Medien bei Bedarf unter kontrollierten Bedingungen die Sterole und/oder Stanole wieder freizusetzen vermögen.
Beschreibung der Erfindung
Gegenstand der Erfindung sind Sterol- und/oder Stanolester von Ethercarbonsäuren der allgemeinen Formel (I)

in der R für einen von einem Alkohol abgeleiteten Alkyl-, Alkenyl- und/oder Alkylphenylrest mit 1 bis 50 Kohlenstoffatomen, OAlk für ringgeöffnete Ethylenoxid-, Propylenoxid- und/oder Butylenoxid-Einheiten, und n für Zahlen von 0 bis 100 steht.
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Herstellung von Sterol- und/oder Stanolestern von Ethercarbonsäuren, dadurch gekennzeichnet, dass Sterole und/oder Stanole mit Ethercarbonsäuren bzw. deren Salze der Formel (II)
R(OAIk)nOCH COOX (II),
in der R für einen von einem Alkohol abgeleiteten Alkyl-, Alkenyl- und/oder Alkylphenylrest mit 1 bis 50 Kohlenstoffatomen, Oalk für ringgeöffnete Ethylenoxid-, Propylenoxid und/oder Butylenoxid-Einheiten, n für Zahlen von 0 bis 100 und X für Wasserstoff oder ein Alkalimetall steht, in Anwesenheit eines Veresterungskatalysators verestert werden. Weitere Gegenstände der vorliegenden Erfindung betreffen die Verwendung von Sterol- und/oder Stanolester von Ethercarbonsäuren nach Formel (I) als Sterol- und/oder Stanol-Quelle, vorzugsweise als Rohstoff zur Herstellung von Steroid Vorstufen, insbesondere zur fermentativen Herstellung von 4- Androsten- 3,17-dion (AD) und/oder 4-Androstadien-3,17-dion (ADD), als Emulgatoren, insbesondere in kosmetischen Zusammensetzungen und in Nahrungsmitteln, als kosmetischer Wirkstoff und schließlich als hypocholesterinänischer Wirkstoff, insbesondere in Nahrungsmitteln und/oder Nahrungsergän- zungsmitteln.
Sterole und Stanole
Sterole, die auch häufig als Sterine bezeichnet werden, sind Steroide mit 27 bis 30 Kohlenstoffatomen mit einer Hydroxylgruppe am dritten Kohlenstoffatom. In der Regel weisen Sterole auch eine Doppelbindung auf. Stanole sind die entsprechenden gesättigten Sterolverbindungen, die keine Doppelbindung aufweisen. Die Sterole sind in der Natur frei, als Ester oder Glykoside sehr verbreitet. Dabei werden tierische Sterole, sogenannte Zoosterine, pflanzliche Sterole, sogenannte Phytosterole und Sterole von Pilzen, sogenannte Mykosterine, unterschieden. Im Sinne der vorliegenden Erfindung sind prinzipiell alle Sterole bzw. Stanole geeignet, vorzugsweise die Phytosterole bzw. deren hydrierte Stanolver- bindungen. Bevorzugt werden Phytosterole, die hauptsächlich eine oder mehrere der folgenden Verbindungen enthalten: ß-Sitosterol, Campesterol, Stigmasterol, Brassicasterol, Stigmastanol und Cam- pestanol.
Insbesondere geeignet sind im Sinne der Erfindung Mischungen von Sterolen und/oder Stanolen, die mindestens zwei der folgenden Verbindungen: ß-Sitosterol, Campesterol, ß-Sitostanol und/oder Cam- pestanol enthalten, die folgende Formel aufweisen:

Ganz besonders bevorzugt im Sinne der vorliegenden Erfindung werden Phytosterole derivatisiert von pflanzlichen Ölen, insbesondere von Rapsöl. Insbesondere geeignet sind die unter dem Handelsnamen General ™100 und General ™122 von der Cognis Corporation vertriebenen Sterolmischungen. General ™ 100 ist eine Mischung, die reich an Beta-Sitosterol, Campesterol und Stigmasterol ist (ca. 80 Gew.%) und zusätzlich Brassicasterol (ca. 5 Gew.%), Stigmastanol (ca. 12 Gew.%) and Campestanol (Rest zu 100 Gew.%) enthält. General ™122 ist ebenfalls eine Mischung, die 40 bis 55 Gew.% beta-Sitosterol, 20 bis 28 Gew.% Campesterol, 14 bis 23 Gew.% Stigmasterol und ggf. zusätzlich 0 bis 8 Gew.% Brassicasterol, 0 bis 5 Gew.% Stigmastanol und 0 bis 2 Gew.% Campestanol enthält. Weiterhin insbesondere geeignet ist das von der Cognis Deutschland GmbH & Co. KG vertriebene General R™, eine Mischung aus 40-60 Gew.% beta-Sitosterol, 30-45 Gew.% Campesterol und 8-18 Gew.% Brassicasterol, 0-5 Gew.% Stigmasterol, 0-5 % Stigmastanol, 0-5 Gew.% Ergostanol und einem Gesamtgehalt von 90- 100 Gew.%.
Ethercarbonsäuren
Im Sinne der Erfindung sind die Sterole und/oder Stanole verestert mit Ethercarbonsäuren der Formel (I), d.h. die Hydroxylgruppe der Sterole und/oder Stanole bildet mit der Ethercarbonsäure eine Estergruppe.
Zur Herstellung der an und für sich bekannten Ethercarbonsäuren geht man von Alkoholen aus, die im ersten Verfahrensschritt ggf. alkoxyliert, und anschließend mit einer Halogencarbonsäure oder deren Salze carboxymethyliert werden. Dieses Verfahren wird beispielsweise in der deutschen Patentschrift DE-C-197 40 954 beschrieben.
Die Alkohole, die der späteren Ethercarbonsäure zugrunde liegen, können aliphatischer, cycloaliphati- scher oder auch aromatischer Natur und sowohl gesättigt oder ungesättigt sein. Bewährt haben sich lineare aliphatische Alkohole mit 1 bis 36 Kohlenstoffatomen, die ggf. eine weitere funktionelle Gruppe tragen (d.h. R in Formel (I) steht für einen von einem Alkohol abgeleiteten linearen ggf. weitere funktionelle Gruppen tragenden Alkylrest mit 1 bis 36 Kohlenstoffatomen). Bei den weiteren funktioneilen Gruppen handelt es sich vorzugsweise um Hydroxylgruppen, insbesondere um endständige Hydroxylgruppen. Mit anderen Worten kann es sich bei den Ethercarbonsäuren zugrunde liegenden Alkoholen um Diole, also difunktionelle Alkohole handeln, die vorzugsweise 2 bis 36 Kohlenstoffatome aufweisen. Für diesen Fall steht in der allgemeinen Formel (I) R für einen von einem difunktionellen Alkohol abgeleiteten linearen endständig Hydroxylgruppe tragenden Alkylrest mit 2 bis 36 Kohlenstoffatomen. Beispiele für derartige difunktionelle Alkohole sind Ethylenglykol, Propylenglykol, 1 ,5- Pentandiol, 1 ,8- Octandiol, 1 ,6-Hexandiol,1 ,10-Decandiol, 1 ,12 Dodecandiol oder die dem Fachmann bekannten Dimer- und/oder Trimeralkohole. Unter Dimerdiol/Trimertriolen sind technische Gemische zu verstehen, die durch Oligomerisierung von ungesättigten Fettsäuren mit 12 bis 22, vorzugsweise 16 bis 18 Kohlenstoffatomen oder deren Methylester und nachfolgende Hochdruckhydrierung erhalten werden.
Bevorzugt im Sinne der Erfindung leiten sich die Ethercarbonsäuren der allgemeinen Formel (I) ab von monofunktionellen aliphatischen Alkoholen. Beispiele für geeignete primäre monofunktionelle geradket- tige Alkohole sind Hexanol, Heptanol, Octanol, Nonanol, Decanol, Undecanol, Dodecanol, Tridecanol, Tetradecanol, Pentadecanol, Hexadecanol, Heptadecanol, Octadecanol, Nonadecanol, Eicosanol, Docosanol, Tetracosanol, 10-Undecen-1-ol, Oleylalkohol, Elaidylalkohol, Ricinolalkohol, Linoleylalkohol, Linolenylalkohol, Gadoleylalkohol, Arachidonalkohol, Erucaalkohol, Brassidylalkohol. Beispiele für geeignete primäre monofunktionelle verzweigte Alkohole sind Isononylalkohol, Isotridecylalkohol oder Guerbetalkohole, die bekanntlich durch Dimerisierung von Fettalkoholen zugänglich sind und sich strukturell dadurch auszeichnen, dass sie in α-Stellung zur endständigen CH2θH-Gruppe einen längeren Alkylrest vorzugsweise mit 2 bis 12 Kohlenstoffatomen aufweisen. Geeignete Guerbetalkohole sind 2-Hexyldecanol, 2-Octyldodecanol und 2-Hexyldecylpalmitat/stearat, 2-Ethylhexanol und Propylhepta- nol.
Bevorzugt im Sinne der Erfindung werden Ethercarbonsäuren, die sich von monofunktionellen gesättigten linearen Alkoholen mit 1 bis 18 Kohlenstoffatomen ableiten, d.h. in der allgemeinen Formel (I) steht R für einen von einem monofunktionellen Alkohol abgeleiteten linearen Alkylrest mit 1 bis 18 Kohlenstoffatomen. Insbesondere geeignet sind Ethercarbonsäuren, die sich von Methanol, Ethanol, Propa- nol, Butanol, Capronalkohol, Caprylalkohol, Caprinalkohol, Laurylalkohol, Myristylalkohol, Cetylalkohol urid/oder Stearylalkohol, sowie deren technische Mischungen, die z.B. bei der Hochdruckhydrierung von technischen Methylestern auf Basis von Fetten und Ölen anfallen ableiten. Insbesondere bevorzugt sind Ethercarbonsäuren von linearen Alkoholen mit 1 bis 6 Kohlenstoffatomen.
Die Ethercarbonsäuren der allgemeinen Formel (I) enthalten gegebenenfalls ringgeöffnete Ethylenoxid-, Propylenoxid- und/oder Butylenoxid-Einheiten (OAlk), vorzugsweise steht OAlk für die Ethylenoxid- Einheit OCH2CH2. Der Alkoxylierungsgrad der Ethercarbonsäuren der allgemeinen Formel (I) ist durch „n" wiedergegeben, wobei es sich selbstverständlich um ein reines statistisches Maß handelt. evorzugt steht n für eine Zahl von 1 bis 20, vorzugsweise von 3 bis 15.
Besonders geeignete Ethercarbonsäuren sind kommerziell erhältlich beispielsweise von der Firm a Kao unter dem Handelsnamen Akypo™, wie Akypo LF 1 ™ (Ethercarbonsäure auf Basis von Octanol, ethoxyliert mit 5 Mol Ethylenoxid) Akypo LF 2 ™ (Ethercarbonsäure auf Basis von Octanol, ethoxyliert mit 8 Mol Ethylenoxid), Akypo RLM 100 ™ (Ethercarbonsäure auf Basis von C12/14-Alkohol, ethoxyliert mit 10 Mol Ethylenoxid).
Verfahren zur Herstellung von Sterol- und/oder Stanolestern von Ethercarbonsäuren
Die erfindungsgemäßen Sterol- und/oder Stanolester werden durch Veresterung der Sterole und/oder Stanole mit Ethercarbonsäuren bzw. deren Salze der Formel (II) in Anwesenheit eines Veresterungskatalysators hergestellt.
In der Regel werden die Sterole und/oder Stanole und Ethercarbonsäuren in molaren Mengen von 2: 1 bis 1 : 2, vorzugsweise von 1 ,5: 1 bis 1 : 1 ,5 und insbesondere von 1 ,3 : 1 bis 1: 1 ,3 eingesetzt. Um eine möglichst vollständige Veresterung zu erhalten, empfiehlt es sich die Ethercarbonsäure in leichtem Überschuß einzusetzen. Die Veresterung wird in Anwesenheit bekannter Veresterungskatalysatoren wie Zinkoxalat, Zinkoxid oder Calciumoxid durchgeführt. Es empfehlen sich Reaktionstemperaturen über den Schmelzpunkt und unterhalb des Zersetzungspunktes der Sterole und/oder Stanole, vorzugsweise zwischen 190 und 230 °C. Bei einem Überschuss an Alkoholkomponente kann anhand der Säurezahl nach DIN 53402 der Grad der Veresterung abgelesen werden, wobei die Veresterung bei einer Restsäurezahl (SZ) unter 10, vorzugsweise unter 5, als praktisch vollständig angesehen werden kann. Bei einem Überschuss an Säurekomponente kann analog verfahren werden.
In der Regel beträgt die Reaktionszeit in etwa 4 bis 6 Stunden, wobei die Einsatzmenge an Katalysator, Reaktionstemperatur und Resthydroxylzahl mitbestimmende Parameter sind. Falls gewünscht kann die Veresterung im Vakuum durchgeführt werden. Weiterhin kann es vorteilhaft sein, vor der eigentlichen Veresterung die Ausgangsstoffe zu trocknen.
Gewerbliche Anwendbarkeit
Die erfindungsgemäßen Ethercarbonsäureester der Sterole und/oder Stanole sind flüssige oder wachsartige Produkte, die leicht zu schmelzen sind. Weiterhin können die erfindungsgemäßen Verbindungen leicht in Wasser emulgiert werden, ohne dass es eines Zusatzes von Emulgatoren bedarf, d.h. sie sind in Wasser selbstemulgierbar. Aufgrund dieser gegenüber den Sterolen und/oder Stanolen vorteilhaften Eigenschaften sind die erfindungsgemäßen Ethercarbonsäureester der Sterole und/oder Stanole eine Sterol- und/oder Stanolquelle, d.h. sie können in den gleichen Anwendungsgebieten wie die Sterole und/oder Stanole eingesetzt werden. Bevorzugt im Sinne der Erfindung werden die Ether- carbonsäureester der Sterole und/oder Stanole einerseits als Rohstoff bei der fermentativen Herstellung von 4-Androsten-3,17-dion (AD) und/oder 4-Androstadien-3, 17-dion (ADD) verwendet, welche wiederum weiter zur Herstellung der verschiedensten Steroidderivate wie Testosteron, Estradiol, Ethi- nylestradiol, Testolactone, Progesterone, Cortison, Cortisol, Prednison oder auch Prednisolon eingesetzt werden können. Bei der fermentativen Herstellung von AD und ADD werden die Ethercarbonsäureester der Sterole und/oder Stanole analog den Bedingungen aus der Deutschen Offenlegungsschrift DE 3147834-A, die die Fermentation von Cholesterin unter Zusatz von ethoxylierten Sterolen beschreibt, umgesetzt und aufgearbeitet. Weitergehende Informationen sind dem Fachmann aus dem Artikel von P. Fernandez et al., „Microbial conversion of steroid compounds: recent developments", aus Enzyme and Microbial Techn. 32 (2003), 688-705 zugänglich. Zur Nutzung der genannten Substanzen als Sterinquelle ist auf die Anwesenheit von Lipasen zur Esterspaltung zu achten.
Aufgrund seiner selbstemulgierenden Eigenschaften können die Ethercarbonsäureester der Sterole und/oder Stanole als Emulgatoren, insbesondere in kosmetischen Zusammensetzungen und in Nahrungsmitteln verwendet werden. Dabei sind sie als Emulgatoren für O/W- und für W/O-Emulsionen geeignet. Die Einsatzmenge richtet sich nach dem Anwendungszweck und liegt in der Regel bei 0,1 bis 10 Gew.%, vorzugsweise 0,5 bis 5 Gew.%, bezogen auf Mittel. Besonders bevorzugt werden Kombinationen mit weiteren aus dem Stand der Technik bekannten Emulgatoren verwendet. Als weitere Emulgatoren eignen sich beispielsweise nichtionogene Tenside aus mindestens einer der folgenden Gruppen:
> Anlagerungsprodukte von 2 bis 30 Mol Ethylenoxid und/ oder 0 bis 5 Mol Propylenoxid an lineare Fettalkohole mit 8 bis 22 C-Atomen, an Fettsäuren mit 12 bis 22 C-Atomen, an Alkylphenole mit 8 bis 15 C-Atomen in der Alkylgruppe sowie Alkylamine mit 8 bis 22 Kohlenstoffatomen im Alkylrest; Alkyl- und/oder Alkenyloligoglykoside mit 8 bis 22 Kohlenstoffatomen im Alk(en)ylrest und deren ethoxylierte Analoga; Anlagerungsprodukte von 1 bis 15 Mol Ethylenoxid an Ricinusöl und/oder gehärtetes Ricinusöl;
> Anlagerungsprodukte von 15 bis 60 Mol Ethylenoxid an Ricinusöl und/oder gehärtetes Ricinusöl; Partialester von Glycerin und/oder Sorbitan mit ungesättigten, linearen oder gesättigten, verzweigten Fettsäuren mit 12 bis 22 Kohlenstoffatomen und/oder Hydroxycarbonsäuren mit 3 bis 18 Kohlenstoffatomen sowie deren Addukte mit 1 bis 30 Mol Ethylenoxid;
> Partialester von Polyglycerin (durchschnittlicher Eigenkondensationsgrad 2 bis 8), Polyethy- lenglycol (Molekulargewicht 400 bis 5000), Trimethylolpropan, Pentaerythrit, Zuckeralkoholen (z.B. Sorbit), Alkylglucosiden (z.B. Methylglucosid, Butylglucosid, Laurylglucosid) sowie Polyglu- cosiden (z.B. Cellulose) mit gesättigten und/oder ungesättigten, linearen oder verzweigten Fettsäuren mit 12 bis 22 Kohlenstoffatomen und/oder Hydroxycarbonsäuren mit 3 bis 18 Kohlenstoffatomen sowie deren Addukte mit 1 bis 30 Mol Ethylenoxid; > Mischester aus Pentaerythrit, Fettsäuren, Citronensäure und Fettalkohol und/oder Mischester von Fettsäuren mit 6 bis 22 Kohlenstoffatomen, Methylglucose und Polyolen, vorzugsweise Gly- cerin oder Polyglycerin.
> Mono-, Di- und Trialkylphosphate sowie Mono-, Di- und/oder Tri-PEG-alkylphosphate und deren Salze;
> Wollwachsalkohole;
> Polysiloxan-Polyalkyl-Polyether-Copolymere bzw. entsprechende Derivate;
> Block-Copolymere z.B. Polyethylenglycol-30 Dipolyhydroxystearate;
> Polymeremulgatoren, z.B. Pemulen-Typen (TR-1 ,TR-2) von Goodrich;
> Polyalkylenglycole sowie
> Glycerincarbonat.
Eine weitere Verwendung der Sterol- und/oder Stanolester von Ethercarbonsäuren betrifft die Verwendung als kosmetischer Wirkstoff.
Weiterhin können die Sterol- und/oder Stanolester der Ethercarbonsäuren nach Anspruch 1 als hypo- cholesterinänischer Wirkstoff, insbesondere in Nahrungsmitteln und/oder Nahrungsergänzungsmitteln verwendet werden. So können beispielsweise die Verbindungen in Nahrungsmitteln wie Margarine, Salatöle und Bratöle gemäß den o.g. Schriften, vorzugsweise in Mengen von 0,1 bis 10 Gew.% bezogen auf Mittel zugesetzt werden. Durch die Zugabe der genannten Ester und die durch Lipasen im Körper bewirkte Spaltung der Ester und die somit erwirkte Freisetzung des Sterins wird eine Reduzierung des Cholesterinspiegels im Blut von Mensch und Tier erzielt.
Beispiele
Generol R™ der Fa. Cognis Deutschland GmbH & Co. KG ist eine Mischung von Phytosterolen auf Basis von 40-60 Gew.% ß-Sitosterol, 30-45 Gew.% Campesterol und 8-18 Gew.% Brassicasterol, 0-5 Gew.% Stigmasterol, 0-5 % Stigmastanol, 0-5 Gew.% Ergostanol und einem Gesamtgehalt von 9O-100 Gew.%. Schmelzbereich 130-145 °C.
Beispiel 1
In einem Reaktionskolben wurden 0,5 mol (212,3 g) Generol R™ der Fa. Cognis Deutschland GmbH & Co. KG (Kenndaten: OHZ 132; mittleres Molekulargewicht berechnet aus der OHZ = 425,1 g /mol) und 0,65 mol (308,1 g) der Ethercarbonsäure Akypo™ LF 1 von Kao (Ethercarbonsäure auf Basis von n- Octanol, ethoxyliert mit 5 Mol Ethylenoxid; Säurezahl 118 und mittleres Molekulargewicht 474,4 g/mol) bei 140°C eine Stunde getrocknet. Anschließend wurden 0,8 g (0,15 Gew.%) Calciumoxid als Veresterungskatalysator zugesetzt und im Wasserstrahlvakuum 5 Stunden bei 200° C umgesetzt. Nach dem Abkühlen auf ca. 60 °C wurde das Produkt über eine Nutsche abfiltriert.
Man erhielt einen bei Raumtemperatur flüssigen Ethercarbonsäureester mit den in Tabelle 1 wiedergegebenen Kenndaten. Aus der Differenz der Verseifungs- und Säurezahl ist klar erkennbar, dass eine Veresterung erfolgte.
Die restliche Säurezahl erklärt sich durch nicht vollständige Umsetzung.
Beispiel 2
Analog Beispiel 1 wurden 0,5 mol Generol R™ der Fa. Cognis Deutschland GmbH & Co. KG und 0,65 mol Akypo LF 2™ (Ethercarbonsäure auf Basis eines Octanols ethoxyliert mit 8 Mol Ethylenoxid; Säurezahl 96,4) umgesetzt.
Man erhielt einen wachsartigen Ethercarbonsäureester mit einem Schmelzbereich von 30 bis 40 °C und den in Tabelle 1 angegebenen Kenndaten.
Beispiel 3
Analog Beispiel 1 wurden 0,5 mol Generol R™ der Fa. Cognis Deutschland GmbH & Co. KG und 0,65 mol Akypo RLM 100™ (Ethercarbonsäure auf Basis eines C12/C14 Alkohols ethoxyliert mit 10 Mol Ethylenoxid; Aktivsubstanzgehalt 85 Gew.%, Säurezahl 74) umgesetzt.
Man erhielt einen wachsartigen Ethercarbonsäureester mit einem Schmelzbereich von 30 bis 40 °C und den in Tabelle 1 angegebenen Kenndaten.
Beispiel 4
10 g der in den Beispielen 1 bis 3 hergestellten Ethercarbonsäureester wurden mit 90 g Wasser versetzt und umgerührt. In allen Fällen erhielt man wässrige beige-farbene Emulsionen mit geringem Bodensatz. Da keine weiteren Emulgatoren zugegeben werden mussten, sind die Ester selbstemulgierend in Wasser.
In der folgenden Tabelle 1 sind die Kenndaten der erhaltenen Ethercarbonsäureester der eingesetzten Sterolmischung angegeben.
Die Verseifungszahl (VZ) wurde bestimmt gemäß DGF C-V 3 Die Hydroxylzahl (OHZ) wurde bestimmt gemäß DGF C-V 17a Die Säurezahl (SZ) wurde bestimmt gemäß DIN 53402

Beispiel 5
Herstellung von O/W- Emulsionen
Es wurden nach einem üblichen Heißprozess O/W- Emulsionen hergestellt, in dem die Ölphase enthaltend Emulgatoren, Fettalkohole und ein Emollient auf 80 °C erhitzt und mittels Rührer homogenisiert wurde. Bei dieser Temperatur wurde unter Rühren eine ebenfalls auf 80°C erhitzte Glycerin/Wasser- π Phase eingebracht. Unter Rühren wurde die Mischung abgekühlt, wobei Formalin zur Konservierung beigegeben wurde.
Die Emulsionen wurden hinsichtlich der Viskosität (Brookfield RVF, Spindel 5 mit 10 Upm) und Stabilität (optische Skala von 1 bis 5, wobei 1 die beste Note darstellt; nach 1-wöchiger Lagerung bei -5 bzw. 40°C) untersucht.
Die Zusammensetzungen der O/W- Emulsionen (Mengenangaben in g) und deren Bewertung sind der folgenden Tabelle 2 zu entnehmen:

Aus Tabelle 2 ist erkennbar, dass die erfindungsgemäßen Ethercarbonsäureester der Sterole alleine Emulgatoreigenschaften aufweisen (Beispiel 5a) und auch in Mischung mit Co-Emulgatoren (Beispiel 5b). Beispiel 6
Biotransformation des Sterol-EO-Ester gemäß Beispiel 1 zu Steroid-Vorstufen
Eingesetzt wurden Hefeextrakt und Kartoffel-Dextrose-Agar der Fa. Difco (USA), Glycerin mit Reinheitsgrad p.a. der Fa. Riedel-de Haen (Deutschland), Tween 20 (Polyethylenglycol-sorbitan- monolaurat) und 4-Androsten-3,17-dion (AD) und 1 ,4-Androstadien-3, 17-dion (ADD) der Fa. Sigma (USA)
Zellkultur: Mycobacterium sp. NRRL B-3805-Zellen wurden in Kartoffel-Dextrose-Agar in einem Schrä- gagarröhrchen (42 gM) gehalten. Inoculum; Mycobacterium sp. NRRL B-3805-Zellen wurden drei Tage lang bei 30°C in einem komplexen Medium A aus Hefeextrakt (10 gM), Glycerin (10 gM), Sterin-Ester (1 ,0 gM, außer wenn anders angegeben), Tween 20 (0,8 gM) in einer Kaliumphosphatpufferlösung mit einem pH-Wert von 7 (20 mM) gezüchtet.
Anschließend wurden die Zellen in 20 ml oder 160 ml des selben Mediums A gegeben. Nach einem Wachstum von 24 Stunden wurde das Medium A zur Beimpfung von 1 ,4 I (2 I Fermenterbehälter (B Braun Biostat MD) eingesetzt. Während der Fermentation wurde mit Luft begast (Begasungsrate 0.5 vvm), und der pH-Wert des Fermentationsmediums wurde durch Zugabe von NaOH (2 N) oder H2SO4 (2 N) auf pH 7 geregelt. Es wurden Proben entnommen, um die gebildete AD-konzentration zu überprüfen.
Analytische Methoden
Routinemäßig wurden periodisch Proben des Fermentationsmediums entnommen und mit dem zweifachen Volumen einer Lösung von Progesteron (0,2 gM ) in n-Heptan als interner Standard extrahiert. Die organische Phase wurde auf ihren Sterol-/Steroidgehalt mittels HPLC analysiert.
Eine Probe des Esters aus Beispiel 1 wurde gemäß der folgenden Prozedur verseift: ca. 1 g jedes Esters wurde zu 25 ml einer 0,5 M ethanolischen Lösung von KOH gegeben und über Nacht bei 28°C inkubiert. 25 ml einer NaCI (5 Gew.%) Lösung wurde danach zugegeben und das Ganze mit 25 ml Diethylether extrahiert. Die organische Phase wurde eingedampft und der trockene Rückstand gewon- nen. Ungefähr 6 mg der verseiften Probe wurde n m er internen tandard-Lösung gelöst unq mittels HPLC quantitativ auf Sterine untersucht.
Während des Fermentationsversuchs wurden wie folgt Proben entnommen und verseift: 2,5 ml einer 0,5 M ethanolischen KOH-Lösung wurden zu 500 ml einer Fermentationsprobe zugegeben und über Nacht bei 28°C inkubiert. Danach wurden 25 ml einer 5 Gew.%-igen NaCI-Lösung zugegeben und das Ganze mit 25 ml Diethylether extrahiert. Die organische Phase wurde eingedampft; der gewon nene trockene Rückstand wurde in 1 ml interner Standard-Lösung gelöst und mittels HPLC quantitativ auf Sterine untersucht.
Die HPLC-Analyse (Lichrospher Si-60 Säule, 10 Om Teilchengröße) wurde mit 1 ml min-1 Flussrate durchgeführt, um die Substrat- und Produktkonzentration zu bestimmen (UV-Detektion bei 215 und 254 nm). Die mobile Phase bestand aus (v/v) n-Heptane (96):Ethanol (4).
Nach 130 Std. Fermentationszeit wurden ungefähr 0,6 mM 4-Androsten-3,17-Dion produziert. Dieses Ergebnis zeigt, dass Mycobacterium sp. NRRL B-3805-Zellen in der Lage sind, die Ester erfindungsgemäß in AD zu konvertieren.

Claims

Patentansprüche
1. Sterol- und/oder Stanolester von Ethercarbonsäuren der allgemeinen Formel (I)
R(OAIk)nOCH2COOH (I),
in der R für einen von einem Alkohol abgeleiteten Alkyl-, Alkenyl- und/oder Alkylphenylrest mit 1 bis 50 Kohlenstoffatomen, OAlk steht für ringgeöffnete Ethylen-, Propylen- und/oder Butenyloxid- Einheiten und n für Zahlen von 0 bis 100 steht.
2. Sterol- und/oder Stanolester nach Anspruch 1 , dadurch gekennzeichnet, dass R für einen von einem Alkohol abgeleiteten linearen ggf. weitere funktioneile Gruppen tragenden Alkylrest mit 1 bis 36 Kohlenstoffatomen steht.
3. Sterol- und/oder Stanolester nach mindestens einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass R für einen von einem monofunktionellen Alkohol abgeleiteten linearen Alkylrest mit 1 bis 18 Kohlenstoffatomen steht.
4. Sterol- und/oder Stanolester nach mindestens einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass R für einen von einem difunktionellen Alkohol abgeleiteten linearen endständig Hydroxylgruppe tragenden Alkylrest mit 2 bis 36 Kohlenstoffatomen steht.
5. Sterol- und/oder Stanolester nach mindestens einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass OAlk für die ringgeöffnete Ethylenoxid-Einheit OCH2CH2 steht.
6. Sterol- und/oder Stanolester nach mindestens einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass n für eine Zahl von 1 bis 20, vorzugsweise von 3 bis 15, steht.
7. Verfahren zur Herstellung von Sterol- und/oder Stanolestern von Ethercarbonsäuren, dadurch gekennzeichnet, dass Sterole und/oder Stanole mit Ethercarbonsäuren bzw. deren Salze der Formel (II)
R(OAIk)nOCH COOX (II),
in der R für einen von einem Alkohol abgeleiteten Alkyl-, Alkenyl- und/oder Alkylphenylrest mit 1 bis 50 Kohlenstoffatomen, OAlk für ringgeöffnete Ethylen-, Propylen- und/oder Butylenoxid- Einheiten, n für Zahlen von 0 bis 100 und X für Wasserstoff oder ein Alkalimetall steht, in Anwesenheit eines Veresterungskatalysators verestert werden.
8. Verwendung von Sterol- und/oder Stanolester von Ethercarbonsäuren nach Anspruch 1 als Sterol- und/oder Stanol-Quelle, vorzugsweise als Rohstoff zur Herstellung von Steroid Vorstufen, insbesondere zur fermentativen Herstellung von 4-Androsten-3,17-dion (AD) und/oder 4-Androsta ien- 3,17-dion (ADD).
9. Verwendung von Sterol- und/oder Stanolestern von Ethercarbonsäuren nach Anspruch 1 als E- mulgatoren, insbesondere in kosmetischen Zusammensetzungen und in Nahrungsmitteln,
10. Verwendung von Sterol- und/oder Stanolestern von Ethercarbonsäuren nach Anspruch 1 als kosmetischer Wirkstoff.
11. Verwendung von Sterol- und/oder Stanolestern von Ethercarbonsäuren nach Anspruch 1 als hy- pocholesterinänischer Wirkstoff, insbesondere in Nahrungsmitteln und/oder Nahrungsergänzungsmitteln.
