Verfahren zur Herstellung von alpha-Bisabolo! aus Nerolidol
Unter Kamillenöl versteht man das ätherische Öl aus den Blütenköpfen der echten Kamille, Chamomilla recutita (L) Rauschert. Als „Oleum chamomillae" ist es im Ergänzungsbuch zum Deutschen Arzneibuch aufgeführt. Die echte Kamille ist eine der gebräuchlichsten Arzneipflanzen. Die Zusammensetzung des Kamillenöls ist von der Provenienz bzw. vom Drogentyp des verwendeten Drogenmaterials abhängig. Sie wird zudem durch die Destillationsbedingungen der Wasserdampfdestillation beeinflußt. Kamillenöl selber enthält eine große Zahl von Mono- und Sesquiterpenen, wobei die therapeutisch relevanten Sesquiterpene quantitativ dominieren. Die wichtigsten Bestandteile des ätherischen Öls sind Chamazulen, das ihm seine tiefblaue Farbe gibt, (-)-alpha-Bisabolol, Bisabololoxid A, Bisaboloxid B, Bisabolonoxid A, eis- und trans-Spiroether und Famesen. Kamillenblüten unterschiedlicher Herkunft weisen dabei auch deutliche Unterschiede in ihrer Zusammensetzung auf. Während Kamille des Bisabolol-Typs in ihrem natürlichen Vorkommen auf Nordostspanien beschränkt ist, ist der

Bisabolooxid-A-Typ über ganz Mittel-, Süd- und Osteuropa sowie Ägypten verbreitet. Der seltenere Bisabolonoxid-A-Typ ist aus Albanien und der Türkei bekannt.
In der Beurteilung der therapeutischen Wirksamkeit von Kamillenextrakt- Präparaten nimmt das (-)-alpha-Bisabolol eine dominierende Stellung ein, da es in seiner antiphlogistischen Wirkung dem (+)-alpha-Bisabolol, dem synthetischen Bisabololracemat sowie den Bisabololoxiden A und B deutlich überlegen ist.
Während der systematische Anbau von Arznei und Gewürzpflanzen aufgrund einer gestiegenen Nachfrage nach „nachwachsenden Rohstoffe" weiterhin an Bedeutung gewinnt, führten die beschränkten natürlichen Ressourcen gleichzeitig zu der Suche und Entwicklung von Verfahren zur Gewinnung synthetischer Produkte.
Synthetisches "alpha-Bisabolol" stellt üblicherweise ein diastereomeres Racemat aus gleichen Anteilen (+/-)-alpha-Bisabolol und (+/-)-epi-alpha- Bisabolol dar. Alle vier Enantiomeren wurden in der Natur gefunden.
(-)-(4S,8R)-alpha-epi-Bisabolol ist ein natürlicher Bestandteil von Citrus bergamia RISSO essential oil [(Ohloff, G.; Giersch, W.; Naf , R.; Delay, F.; Helv. Chim. Acta 1986, 69, 698)] und sein Enantiomer (+)-(4R,8S)-alpha-epi- Bisabolol wurde isoliert aus verschiedenen Abies und Picea Specien [O Donnel, G.W.; Sutherland, M. D.; Aust.J.Chem. 1989, 42, 2021], während (+)-(4R,8R)-alpha-Bisabolol Bestandteil des Atalantia monophylla corren oils [O'Donnel, G.W.; Sutherland, M.D. Aust. J. Chem. 1989, 42, 2021 Babin.D.; Fourneron, J.D.; Julia, M.; Tetrahedron 1981 , 37 (suppl.] und dessen Enantiomer (-)-(4S,8S)-alpha-Bisabolol einer der Hauptbestandteile der deutschen Kamille [ Jellinek, J.S.; Parf. Cosm.Aromes 1984, 57, 55] ist
(-)-(4S,8S)-aipha-Bisabolol wird im großen industriellen Maßstab für zahlreiche Anwendungen im Kosmetik- und Riechstoffbereich hergestellt, z.B. zum Einsatz in Schutzcremes, Lotionen, Deodorants etc., und zwar insbesondere wegen seiner antiinflammatorischen, baktereostatischen und
antimycocitischen Eigenschaften [ C. R.; Fleischhauer, J.; Beyer, J.; Reinhard, E.; Planta Med. 1990, 56, 456 ].
Lange Zeit wurde die absolute Konfiguration der einzelnen Enantiomeren des alpha-Bisabolol nicht eindeutig bestimmt. X.-J. Chen, A.Archelas, R. Forstoss in J. Org. Chem. 1993, 58, 5528 beschreiben dann aber schließlich ein Verfahren zur Herstellung der einzelnen Isomeren durch enantioselektive Hydrolyse, ausgehend von (4S,8RS)- und (4R,8RS)-8,9-Epoxy-p-Menth-1- en.
Aufgrund seiner beschriebenen Wirkungen besteht ein ständiger Bedarf an (+), (-) und (+/-)-alpha-Bisabolol, und/oder (+)-epi , (-)-epi und (+/-)-epi-alpha- Bisabolol, d.h. an Verbindungen der Formel A
A
in der geschlängelte Linien jeweils unabhängig voneinander für eine S- oder R-Konfiguration am zugehörigen C-Atom stehen. So wurden in der Vergangenheit eine Vielzahl von Verfahren und Prozessen zur Herstellung von Bisabolol ausgehend von Nerolidol beschrieben.
Die erste katalytische Cyclisierung von Famesol wurde 1913 beschrieben, als man beobachtete, dass bei einer Reaktionsausführung in Anwesenheit von Kaiiumhydrogensulfat neben den erwarteten Kohlenwasserstoffen auch einige mono- und bicyclische Verbindungen gefunden wurden [F.M.Semmler, K.E.Spomitz, Chem. Ber. 46, 4024 (1913)]. Spätere Arbeiten indentifizierten dann diese cyclischen Verbindungen als Verbindungen der Bisabolen- und Cadalen-Klasse.
1925 wurde erstmals eine sorgfältige Untersuchung durchgeführt, bei der ausgehend von Nerolidol durch Säurekatalyse Produkte wie Farnesen,
Bisabolen und Bisabolol erhalten wurden [L. Ruzicka, E. Capato, Helv. Chim. Acta 8, 259 (1925)]. Es wurde insbesondere gezeigt, daß Nerolidol durch Zusatz von Acetanhydrid und anschließenden Umsatz mit Essig/Schwefelsäure oder Ameisensäure bei Raumtemperatur ein Gemisch liefert, welches Bisabolol umfasst.
1968 . berichtet dann Gutsche [C.D.Gutsche, J.R.Maycock, C.T.Chang, Tetrahedron 24, 859] über die säurekatalysierte Cyclisierung von Farnesol und Nerolidol. Ausgehend von Farnesol oder Nerolidol wurden zunächst durch Umsatz mit Ameisensäure die entsprechenden Formiate erhalten, die dann in einem zweiten Schritt zu den Alkoholen verseift wurden.
Weitere Synthesen von Bisabolol wurden von Ruzicka und Liguari [L. Ruzicka, M. Liguari, Helv. Chim. Acta 15, 3, (1932)] und durch Manjarrez und Guzmann [A. Manjarrez, A. Guzmann, J. Org. Chem. 31 , 348, (1966)] beschrieben. Die säurekatalysierte Cyclisierung in Gegenwart von Ameisensäure in Pentan bzw. AICI3 in Ether [N. H.Andersen, D.D.Syrdal.Tetrahedron Lett, 1972, 2455 ], KHS04 [G. Brieger, T.J. Nestrick, C.McKenna J.Org.Chem.,34, 3789, (1969)] und BF3-Etherat in Methylenchlorid [Y. Ohta, Y. Hirase, Chem. Lett., 1972, 263] wurde ebenfalls beschrieben.
K. Uneyama berichtet dann über eine elektrochemische Darstellungsmethode [K.Uneyama, Y. Masatsugu, T.Ueda, S.Torii, Chem. Lett. , 1984, 529]; dabei wird auch über die Herstellung von DL Bisabolol aus DL-Nerolidol berichtet.
Während die zuvor vorgestellten von Nerolidol ausgehenden Verfahren selten zu Bisabolol-Ausbeuten über 30% führten, wurden mit elektrochemischen
Verfahren Ausbeuten bis zu 52% erhalten. In einer speziellen Elektrolyseapparatur wurde hierbei DL-Nerolidol in einem neutralen
Lösemittel, unter Zusatz von Perchloraten an einer Platinelektrode elektrolysiert. Unter den verschiedenen Metallperchloraten wurden die besten
Ausbeuten mit LiCIO4 erhalten. Als bestes Lösemittel erwies sich Aceton. Da unter den Elektrolysebedingungen in situ eine starke Säure erzeugt wird, wurde auch die direkte Zugabe von Perchlorsäure ohne Elektrolyse getestet.
Die Autoren verweisen auf ein diesbezügliches Experiment, in welchem sie
Perchlorsäure in Aceton mit Nerolidol bei 30°C umsetzten. Die Ausbeute an
Bisabolol verschlechterte sich dabei allerdings deutlich auf 23 %, hingegen stieg der Anteil an Bisabolen auf 54 % an.
Angesichts des referierten Standes der Technik ist es überraschend, dass nunmehr erfindungsgemäß ein Verfahren gefunden werden konnte, in dem je nach Wahl der Reaktionsbedingungen, ausgehend von (+)-, oder (-)- oder racemischem (+/-)-Nerolidol, (+)- oder (-)- oder (+/-)-alpha-Bisabolol und/oder (+)-epi-alpha-Bisabolol,(-)-epi-alpha-Bisabolol (+/-)epi-alpha-Bisabolol der Formel A in deutlich verbesserten Ausbeuten hergestellt werden kann.
Das erfindungsgemäße Verfahren zur Herstellung von alpha-Bisabolol umfasst als wesentlichen Schritt: das Umsetzen von Nerolidol mit einem Gemisch aus einem Keton, einer Sulfonsäure und Perchlorsäure. Die Verwendung der Sulfonsäure als Katalysator/Reaktionspartner bei der Umwandlung von Nerolidol in alpha-Bisabolol ist ein wesentlicher Aspekt der vorliegenden Erfindung.
Der Begriff "alpha-Bisabolol" umfasst dabei im Rahmen dieses Textes (+)- alpha-Bisabolol, (-)-alpha-Bisabolol, (+)-epi-alpha-Bisabolol und (-)-epi-alpha- Bisabolol sowie Mischungen von' zwei, drei oder sämtlichen der genannten Isomeren des alpha-Bisabolol. Insbesondere umfasst der Begriff "alpha- Bisabolol" racemische Gemische von (+/-)-alpha-Bisabolol und/oder (+/-)-epi- alpha-Bisabolol.
Der Begriff "Nerolidol" umfasst S-Nerolidol, R-Nerolidol und Gemische aus S- und R-Nerolidol, insbesondere racemische Gemische.
Die im erfindungsgemäßen Verfahren eingesetzte Sulfonsäure besitzt vorzugsweise die nachfolgende Formel C

Als Beispiele für besonders bevorzugte Sulfonsäuren der Formel C seien diejenigen genannt, für die gilt: R2 = Me, Et, n-Propyl, iso-Propyl, n-Butyl, iso- Butyl, n-Pentyl, n-Hexyl, Benzyl, para-Tolyl, CF3, CF3CH2
Bevorzugte Ketone zur Verwendung im erfindungsgemäßen Verfahren sind Ketone der nachfolgenden Formel B

Das molare Verhältnis von Keton zu Nerolidol kann in weiten Bereichen variiert werden, liegt jedoch vorzugsweise im Bereich zwischen 0,1 :1 und 30:1. Molare Verhältnisse im Bereich zwischen 1 :1 und 10:1 haben sich besonders bewährt.
Auch das molare Verhältnis von Sulfonsäure zu Nerolidol kann in weiten Bereich variiert werden. Bevorzugt sind jedoch molare Verhältnisse zwischen 0,001 :1 und 10:1 , wobei besonders gute Resultate mit molaren Verhältnissen im Bereich zwischen 0,01 :1 und 0,5:1 erzielt wurden.
Es sei darauf hingewiesen, dass die Anwesenheit von Wasser im erfindungsgemäßen Verfahren innerhalb weiter Grenzen tolerabel ist. In manchen Fällen hat sich die Anwesenheit von Wasser sogar als vorteilhaft erwiesen, wobei das molare Verhältnis zwischen Wasser und Nerolidol im Bereich zwischen 0,001 :1 und 10 :1 liegen sollte. Molare Verhältnisse im Bereich zwischen 0,01 :1 und 1 :1 sind bevorzugt.
Wenngleich der Reaktionsmechanismus, der bei Durchführung des erfindungsgemäßen Verfahren, d.h. bei der Umsetzung von Nerolidol zu
alpha-Bisabolol und (als Nebenprodukt) Farnesol abläuft, noch nicht als vollständig geklärt gelten kann, besteht doch eine begründete Annahme, dass sich der Mechanismus wie in den beigefügten Figuren 1 und 2 beschreiben lässt.
Dabei kann die Cyclisierung zum Bisabolylgerüst über einen konzertierten Mechanismus interpretiert werden. Es bildet sich gemäß dem "Reaktionsweg Bisabolol" (Fig. 1 ) zunächst unter Säurekatalyse aus dem Nerolidol und dem Keton der Verbindung B ein Halbacetal welches dann mit der Säure der Formel C verestert wird. Es ist bekannt, dass solche Estergruppierungen gute Abgangsgruppen sind [K.Peter; C.Vollhardt; N.E. Schore Organische. Chemie 2. Auflage S. 194 ISBN 3-527-29097-45], und sie begünstigen dadurch augenscheinlich eine konzertierte Cyclisierung zum 6-gliedrigen Ring gegenüber einer ionischen Zwischenstufe die zum Famesyltyp führt (vgl. "Reaktionsweg Farnesol", Fig. 2).
Das erfindungsgemäße Verfahren führt vorteilhafterweise in einem Schritt direkt vom Nerolidol zum gewünschten Bisabolol der Formel A, so dass nicht, wie zu anderen Verfahren beschrieben, ein Umweg über mehrere Stufen durchgeführt werden muss. Auch sind keine speziellen Apparaturen zur Durchführung des Verfahrens erforderlich. Das erfindungsgemäße Verfahren zeichnet sich auch dadurch aus, dass es zu einem besonders reinen Bisabolol führt und insbesondere das bei den Verfahren aus dem Stand der Technik als Nebenprodukt in einer Ausbeute von bis zu 40% entstehende (+), (-) oder (+/- )-Famesol, nur in Konzentrationen von weniger als 2 Gew.-% entsteht.
Produktgemische, die neben dem erwünschten alpha-Bisabolol auch einen nennenswerten Anteil an Farnesolen umfassen, unterliegen bei der anschließenden destillativen Abtrennung der Farnesole einer längeren thermischen Belastung, die zu Nebenreaktionen und insbesondere zur Zersetzung der gebildeten Produkte führen kann. Das erfindungsgemäße Verfahren führt wie erwähnt zu einem Produktgemisch, das alpha - Bisabolol in einem besonders hohen Anteil enthält und daneben nur geringe Mengen an Farnesol. Die destillative Abtrennung des Farnesol nimmt daher keine lange Zeit in Anspruch, was letztlich dazu führt, dass sich Bisabolol bei Einsatz des
erfindungsgemäßen Verfahrens in guten Raum/Zeit-Ausbeuten, in hoher Reinheit und mit einer sensorisch optimalen Qualität herstellen lässt.
Die nachfolgenden Beispiele erläutern das erfindungsgemäße Verfahren näher. Die Anwendung des erfindungsgemäßen Verfahrens ist jedoch nicht auf diese Beispiele beschränkt.
Herstellung von alpha-Bisabolol
Beispiel 1
In einer Standardapparatur bestehend aus 500ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44g (0,2 mol) (+/-)- Nerolidol und 88g (1 ,57 mol) Aceton vorgelegt. Dann wurde bei 15°C innerhalb von 15 min eine Lösung aus 3,6 g ( 32mmol) Methansulfonsäure und 1 ,8 g (11 ,2 mmol) Perchlprsäure 60%ig zugetropft. Anschließend wurde das Reaktionsgemisch 24 Stunden bei 15 °C gerührt und nach erfolgter GC- Kontrolle aufgearbeitet.
Die Aufarbeitung erfolgte derart, dass das Reaktionsgemisch mit 100g Wasser und 100 Diethylether versetzt und danach die org. Phase abgetrennt wurde. Die org. Phase wurde dann mit Soda-Lsg. und Wasser neutralgewaschen. Nach dem Abdestillieren des Lösungsmittels verblieben 43 g Rohprodukt. Die anschließende destillative Reinigung erfolgte an einer 1 m Drehband-Kolonne.

Beispiel 2 (Vergleichsbeispiel)
In einem '500 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-)-Nerόlidol und 88 g (1 ,57 mol) Aceton vorgelegt. Dann wurde bei 15°C innerhalb von 15 min 1 ,8 g (11 ,2 mmol) Perchlorsäure 60%ig zugetropft. Anschließend wurde das Reaktionsgemisch 24 Stunden bei 20 °C gerührt und nach erfolgter GC-Kontrolle aufgearbeitet.
Die Aufarbeitung erfolgte derart, dass das Reaktionsgemisch mit 100g Wasser und 100 g Diethylether versetzt und danach die organische Phase abgetrennt wurde. Die org. Phase wurde dann mit Soda-Lösung und Wasser neutral gewaschen. Nach dem Abdestillieren des Lösungsmittels verblieben 42,5 g Rohprodukt. Die anschließende destillative Reinigung erfolgte an einer 1 m Drehbandkolonne.

Beispiel 3
In einem 550 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-) - Nerolidol und 102 g (1,65 mol) Butan-2-on vorgelegt. Dann wurde bei einer Temperatur von 15 °C bis 20°C innerhalb von 15 min eine Lösung aus 1 ,8 g (18,7 mmol) Methansulfonsäure und 1 ,8 g (11 ,2 mmol) Perchlorsäure 60%ig zugetropft. Die einsetzende Reaktion war leicht exotherm. Anschließend wurde das Reaktionsgsemisch 18 Stunden bei 15 - 20°C gerührt und nach erfolgter GC-Kontrolle aufgearbeitet.
Die Aufarbeitung erfolgte derart, ass das Reaktionsgemisch mit 100 g Wasser und 100 g Diethylether versetzt wurde. Danach wurde die org. Phase
abgetrennt und mit Soda-Lsg. und Wasser neutral gewaschen. Es verblieben 41 g Rohprodukt. Dieses wurde an einer 1 m Drehbandkolonne destilliert.

Beispiel 4
In einem 550 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-) - Nerolidol und 86 g ( 1 moi) Pentan-3-on vorgelegt. Dann wurde bei einer Temperatur von 20°C innerhalb von 15 min eine Lösung aus 3,6 g ( 37,5 mmol) Methansulfonsäure und 0,9 g ( 5,4 mmol) Perchlorsäure 60%ig zugetroft. Anschließend wurde das Reaktionsgemisch 24 Stunden bei 20°C gerührt und nach erfolgter GC - Kontrolle aufgearbeitet.

Beispiel 5
In einem 550 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-) - Nerolidol und 200 g (1 ,67 mol) Acetophenon (Methyl-phenylketon) vorgelegt. Dann wurde bei einer Temperatur von 20°C innerhalb von 10 min eine Lösung aus 1 ,8 g ( 16 mmol ) Methans.ulfonsäure und 1 ,8 g (11 ,2 mmol) Perchlorsäure 60%ig zugetroft. Die einsetzende Reaktion war leicht exotherm. Anschließend wurde das Reaktionsgemisch 34 Stunden bei 20°C gerührt und nach erfolgter GC - Kontrolle aufgearbeitet

Beispiel 6
In einem 550 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-) - Nerolidol, 1 16 g (2 mol) Aceton und 0,9 g ( 0,05 mol) Wasser vorgelegt. Dann wurde bei einer Temperatur von 15°C innerhalb von 10 min eine Lösung aus 3,6g (16 mmol) Methansulfonsäure und 1 ,8g ( 11 ,2 mmol) Perchlorsäure 60 %ig zugetroft. Die einsetzende Reaktion war leicht exotherm. Anschließend wurde das Reaktionsgemisch 24 Stunden bei 20 °C gerührt und nach erfolgter GC - Kontrolle aufgearbeitet.

Beispiel 7
In einem 550 ml-Dreihalskolben mit Rückflußkühler, Tropftrichter und Thermometer wurden 44 g (0,2 mol) (+/-) - Nerolidol,116 g (2 mol) Aceton vorgelegt. Dann wurde bei einer Temperatur von 15°C innerhalb von 10 min eine Lösung aus 7,65 g ( 50 mmol) Trifluormethansulfonsäure und 1 ,8 g ( 11 ,2 mmol) Perchlorsäure 60 %ig zugetropft. Die Reaktion war exotherm. anschließend wurde das Reaktionsgemisch 8 Stunden bei 15°C bis 20°C gerührt und nach erfolgter GC - Kontrolle aufgearbeitet.

Claims
1. Verfahren zur Herstellung von alpha-Bisabolol, mit folgendem Schritt:
Umsetzen von Nerolidol mit einem Gemisch aus einem Keton, einer Sulfonsäure und Perchlorsäure.
2. Verfahren nach Anspruch 1 , wobei eine Sulfonsäure der Formel C
R2S03H
mit R2 = Alkyl, Aryl oder Alkaryl, jeweils verzweigt oder unverzweigt, substituiert oder unsubstituiert, eingesetzt wird.
3. Verfahren nach Anspruch 2, wobei R2 = Me, Et, n-Propyl, iso-Propyl, n-Butyl, iso-Butyl, n-Pentyl, n-Hexyl, Benzyl, para-Tolyl, CF3, CF3CH2.
4. Verfahren nach einem der Ansprüche 1-3, wobei ein Keton der Formel B
B mit R = Me, Et, n-Propyl, iso-Propyl, n-Butyl, iso-Butyl, tert.-Butyl R1 = Me, Et, n-Propyl, iso-Propyl, n-Butyl, iso-Butyl, tert.-Butyl
eingesetzt wird.
5. Verfahren nach einem der vorangehenden Ansprüche, wobei das molare Verhältnis zwischen Keton und Nerolidol im Bereich zwischen 0,1 :1 und 30:1 , vorzugsweise 1 :1 und 10:1 liegt.
6. Verfahren nach einem der vorangehenden Ansprüche, wobei das molare Verhältnis zwischen Sulfonsäure und Nerolidol in einem Verhältnis zwischen 0,001 :1 und 10:1 , vorzugsweise 0,01 :1 und 0,5:1 liegt.
7. Verwendung einer Sulfonsäure als Katalysator oder Reaktionspartner bei der Umwandlung von Nerolidol in alpha-Bisabolol.
